Comprehensive understanding of gene-diet interactions is necessary to establish proper dietary guidelines to prevent and manage general and abdominal obesity. We investigated the role of genetic variants and their interactions with general and abdominal obesity-associated nutrients using a largescale genome-wide association study of Korean adults.
A total of 50,808 participants from a Korean genome and epidemiology study were included. Dietary intake was assessed using a food frequency questionnaire. Obesity was defined as a body mass index ≥25kg/m
. Abdominal obesity (AO) was defined as waist circumference ≥90cm and 80cm in males and females, respectively. Dietary nutrient intake was classified based on Korean Dietary Reference Intakes (DRIs). Odds ratios and 95% confidence intervals were calculated after adjusting for age, sex, exercise, smoking, alcohol drinking, total energy consumption, PC1, and PC2.
Among the individuals consuming fat (%) above DRI, carriers of Ca binding protein 39 (CAB39)- rs6722579 minor allele (A) have a higher risk of AO than those not carrying the SNP (odds ration [OR]=3.73, p-value=2.05e-07; interaction p-value=1.80e-07). Among the individuals consuming vitamin C above DRI, carriers of carboxypeptidase Q (CPQ)- rs59465035 minor allele (T) have a lower risk of AO than those without that SNP (OR=0.89, p-value=1.44e-08; interaction p-value=9.50e-06). The genetic association with obesity was stronger among individuals with a genetic variant rs4130113 near GHR gene region in those consume folate above DRI and with a genetic variant rs5760920 near CRYBB2 gene region in those consume vitamin B2 above DRI.

To investigate the relationship of dietary copper intake with new-onset hypertension among Chinese adults.
A total of 12,245 participants who were free of hypertension at baseline from the China Health and Nutrition Survey (CHNS) were included. Dietary intake was measured by 3 consecutive 24-hdietary recalls combined with a household food inventory. New-onset hypertension was defined as systolic blood pressure ≥140mmHg and/or diastolic blood pressure ≥90mmHg or diagnosed by a physician or under antihypertensive treatment during the follow-up.
During a median follow-up of 6.1 years, 4304 participants developed new-onset hypertension. Overall, the associations between dietary copper intake and new-onset hypertension followed a U-shape (P for nonlinearity <0.001). The risk of new-onset hypertension significantly decreased with the increment of dietary copper intake (per SD increment HR, 0.71; 95% CI, 0.57-0.88) in participants with copper intake <1.57mg/day, and increased with the increment of dietary copper intake (per SD increment HR, 1.09; 95% CI 1.07-1.12) in participants with copper intake ≥1.57mg/day.
There was a U-shaped association between dietary copper intake and new-onset hypertension in general Chinese adults, with an inflection point at about 1.57mg/day. Our results emphasized the importance of maintaining optimal copper intake levels for the primary prevention of hypertension.

Our results emphasized the importance of maintaining optimal copper intake levels for the primary prevention of hypertension.The objective of this study was to investigate the effects of exposure to endotoxin on the reproductive performance of humans and animals in pregnancy and delivery period. Mucin is considered to play a critical role in protecting the tissue epithelium. At pregnancy period, the MUC2 expression of uterus in the High LPS group was significantly higher than that in the Control group. The glycosaminoglycans of gland cells were secreted into the uterine cavity to protect the uterus. Then, the MUC2 layer became thinner, and LPS entered the lamina propria of the uterus. The mRNA expression of tight junction proteins showed a marked drop, and morphological damage of the uterus occurred. Subsequently, the glycosaminoglycans of gland cells in the High LPS and Low LPS groups increased with the increasing LPS dose, and the damage to the endometrial epithelium was repaired in female mice at Day 5 postdelivery. A low dose of LPS activated the PI3K/AKT signaling pathways to increase the glycosaminoglycans particles, while a high dose of LPS inhibited the PI3K/AKT signaling pathway to decrease the glycosaminoglycans particles. Taken together, our results suggest that gland cells secreted glycosaminoglycans particles into the uterine cavity by exocytosis to increase the thickness of the mucus layer to protect the uterus and that this process was regulated by PI3K/AKT signaling pathways.Ammonia exposure can be considered more stressful for aquatic animals when it coincides with high temperature. This study was conducted to detect the effects of ammonia exposure and heat stress and their interactions on the histological features of gills and liver tissues and the expression responses of immune and antioxidative related genes in Nile tilapia. Thus, 180 fish were divided into four groups (triplicates), where the first and third groups were kept in clean water without total ammonium nitrogen (TAN) exposure. At the same time, the second and fourth groups were exposed to 5 mg TAN/L. After seven days, the water temperature was raised in the third (without ammonia toxicity) and fourth (exposed with 5 mg TAN/L) groups up to 32 °C and kept under these conditions for 24 h. While the first (without ammonia toxicity) and second (exposed with 5 mg TAN/L) groups were kept under optimum water temperature (27.28 °C) then gills and liver tissues were dissected. Marked upregulation of keap1 was seen in the gilhe liver showed hepatic vascular congestion and visible necrotic changes with nuclear pyknosis. Calcareous substances (CSs) are also commonly used as robust and cost-effective amendments for increasing the pH of acidic soils and decreasing the mobility of metals in soils. CTP-656 mouse In this study, we developed a new remediation scheme for cadmium (Cd)-contaminated soil remediation by coupling HS and CS. The results showed that regardless of the addition of fulvic acid (FA), all the CS-containing treatments significantly increased the soil pH by 0.32-0.60, and the concentration of bioavailable Cd decreased in the moderately (field experiment soil, maximum 62%) and highly (pot experiment soil, maximum 57%) Cd-contaminated soils. The Cd content in rice (Oryza sativa L.) tissues significantly decreased after all the treatments. The bioaccumulation factors (BAFs) decreased by over 50% in the roots, stems, leaves and husks in all treatments, while the translocation factors (TFs) only significantly decreased in the highly contaminated soil. Among all treatments, the two HS+CS treatments (FA+CaCO3 and FA+CaO) had the greatest effect on decreasing the concentration of bioavailable Cd in soil and Cd in brown rice grains. The suggested mechanism for the effectiveness of coupled HS and CS was that CS first mitigated the pH and precipitated Cd, followed by a complexation effect between HS and Cd. Although the Cd in rice grains in both cases was higher than the standard limit, HS+CS remediation can be advocated as a robust, simple and cost-effective scheme for Cd remediation if the additive dose is slightly increased, as this approach can simultaneously improve the pH of acidic soil and adsorb Cd in soil.The transfer of antibiotic resistance genes (ARGs) in soil under reclaimed water irrigation poses a potential environmental risk. Regulation of NPK fertilizer could influence the behavior of bacterial communities, mobile genetic elements (MGEs), and soil properties, which determine the fate of ARGs. To identify the key element in NPK fertilizer and realize efficient regulation, we explored the effect of individual N, P, K fertilization on ARG variation in tomato rhizosphere and bulk soils. Compared with an unfertilized treatment, N fertilization resulted in greater decreases in the abundance of ARGs (decreases of 24.06%-73.09%) than did either P fertilization (increases of up to 35.84%, decreases of up to 58.80%) or K fertilization (decreases of 13.47%-72.47%). The influence of different forms of N (CO(NH2)2, NaNO3, and NH4HCO3), P (Ca(H2PO4)2 and CaMgO4P+), and K (KCl and K2(SO4)) fertilizers was also investigated in this study, and showed the influence of NaNO3, CaMgO4P+, and K2(SO4) on reducing ARGs abundance was greater in different types of N, P, K fertilizers. Bacterial communities showed the strongest response to N fertilization. The reduced bacterial diversity and abundance of ARG-host and non-host organisms explained the decline of total ARG abundance in soil. In soils fertilized with either P or K, the effect of soil properties, especially total nitrogen and pH, on ARG variation was greater than that of bacterial community and MGEs. These results suggest that N regulation of in NPK fertilizer may be an effective way to reduce the risks of ARGs in soil associated with reclaimed water irrigation.
After heavy metals enter the body, they affect a variety of organs, particularly the main metabolic organ, the liver. Moreover, people are more likely to be exposed to multiple metals than to a single metal. We explored the associations between exposure to a heavy metal mixture and liver function biomarkers.
This study involved 1171 residents living in areas with or without heavy metal exposure in northeast China. Urine concentrations of chromium (Cr), cadmium (Cd), lead (Pb), and manganese (Mn) were measured. Total protein (TP), albumin (ALB), aspartate aminotransferase (AST), and alanine aminotransferase (ALT) were used as biomarkers of liver function. A generalized linear model (GLM), quantile g-computation, and Bayesian kernel machine regression (BKMR) were used to explore the associations between the four metals and liver function.
GLM analysis revealed that Cr level was negatively associated with TP (β=-0.57; 95% CI -0.89, -0.26) and ALB (β=-0.27; 95% CI -0.47, -0.07) levels, and Cd level was positively associated with AST (β=1.04; 95% CI 0.43, 1.65) and ALT (β=0.94; 95% CI 0.08, 1.79) levels. ALB (β=0.26; 95% CI 0.10, 0.41) and ALT (β=0.52; 95% CI 0.02, 1.02) levels were positively associated with urine Mn concentration. The quantile g-computation indicated that exposure to a mixture of the four metals was significantly associated with TP (β=-0.56; 95% CI -0.94, -0.18) and ALT (β=0.84; 95% CI 0.04, 1.63) levels. Among the metals, Cr had the strongest effect on TP and Cd had that on AST. The BKMR model indicated that mixed metal exposure was negatively associated with TP and ALB levels and positively associated with ALT and AST levels.
