Furthermore, by using scanning ultrafast electron microscopy, we examine the spatiotemporal dynamics of charge carriers generated at this junction and find that lateral carrier separation is enabled by differences in the band bending in the vertical direction, which we verify with simulations. Band-bending junctions may therefore enable new optoelectronic devices that rely solely on band bending arising from thickness variations to separate charge carriers.Ursolic acid (UA), a natural triterpenoid widely distributed within fruits and edible plants, has been proven to relieve alcoholic liver disease (ALD). However, the mechanisms involved largely remain unclear. This study investigated whether the beneficial effects of UA on ALD could be related to gut-liver axis (GLA) modulation. Special attention was paid to the contribution of gut microbiome manipulation. UA ameliorated intestinal oxidative stress and barrier dysfunction induced by alcohol. As a consequence of gut leakiness amelioration, the related endotoxemia-mediated liver toll-like receptor 4 pathway induction and the subsequent reactive oxygen species overproduction were reverted. UA also counteracted alcohol-induced gut dysbiosis. A fecal microbiota transplantation study indicated that liver injury as well as ileum oxidative stress and gut barrier dysfunction of recipient mice were partly ameliorated as a result of microbiome remodeling. These results suggest that dietary UA alleviates ALD through GLA homeostasis modulation. We herein report the role of DNA quadruplex folding in liquid droplet formation via LLPS induced by interactions between DNA and linker histone H1 (H1), a key regulator of chromatin organization. Fluidity measurements inside the droplets, binding assays using G-quadruplex-selective probes, and structural analyses based on circular dichroism demonstrated that quadruplex DNA structures, such as the G-quadruplex and i-motif, promote droplet formation with H1 and decrease molecular motility within droplets. The dissolution of the droplets in the presence of additives and the LLPS of the DNA structural units indicated that, in addition to electrostatic interactions between the DNA and the intrinsically disordered region of H1, π-π stacking between quadruplex DNAs could potentially drive droplet formation, unlike in the electrostatically driven LLPS of duplex DNA and H1. According to phase diagrams of anionic molecules with various conformations, the high LLPS ability associated with quadruplex folding arises from the formation of interfaces consisting of organized planes of guanine bases and the side surfaces with a high charge density. Given that DNA quadruplex structures are well-documented in heterochromatin regions, it is imperative to understand the role of DNA quadruplex folding in the context of intranuclear LLPS.Doubly oxido-bridged transition metal moieties, M2(μ-O)2, play important roles as oxidation reaction centers in nature. This work features a diruthenium(III,IV) complex with a doubly oxido-bridged core Ru2III,IV(μ-O)23+ with a carbonato bridged between the two ruthenium centers, M[RuIII,IV(ebpma)2(μ-O)2(μ-O2CO)]2(PF6)3 (M[1CO3]2(PF6)3; Carbonato complex, ebpma; ethylbis(2-pyridymethyl)amine), and explores the interactions of this complex with cations (H+ and M+). M[1CO3]2(PF6)3 was formed via reactions of a singly oxido-bridged complex, [RuIII,IVCl2(ebpma)2(μ-O)]PF6·(CH3)2CO, with M2CO3 (M = K, Na) or with CO2(g), adjusted to around pH 12 with NaOH(aq.), in a water-acetone mixed solvent. The Carbonato complex was isolated as a powder in the form of M[1CO3]2(PF6)3 (M = K, Na), because of the interactions between the carbonato moiety and K+ or Na+ in the solid structure. In acidic aqueous solutions, unexpectedly, the carbonato ligand remained bound to the doubly bridged core, Ru2III,IV(μ-O)23+ or Ru2III,IV(μ-O)(μ-OH)4+, without decarboxylation even under pH 1.0. Two-step one-protonation/deprotonation occurred reversibly between pH 1.0 and 13.2 to the bridging oxido and carbonato ligands. The structures of the corresponding one- and two-protonated complexes ([1CO3H]2+ and [1CO32H]3+) were successfully characterized.Models intended to predict intestinal absorption are an essential part of the drug development process. Although many models exist for capturing intestinal absorption, many questions still exist around the applicability of these models to drug types like "beyond rule of 5" (bRo5) and low absorption compounds. This presents a challenge as current models have not been rigorously tested to understand intestinal absorption. Here, we assembled a large, structurally diverse dataset of ∼1000 compounds with known in vitro, preclinical, and human permeability and/or absorption data. In silico (quantitative structure-activity relationship), in vitro (Caco-2), and in vivo (rat) models were statistically evaluated for predictive performance against this human intestinal absorption dataset. We expect this evaluation to serve as a resource for DMPK scientists and medicinal/computational chemists to increase their understanding of permeability and absorption model utility and applications for academia and industry.
