The Psoriasis Area and Severity Index (PASI) is widely used to score disease severity, but evaluation is subjective, and an objective biomarker would be useful. The stratum corneum (SC), which can be non-invasively harvested, may reflect psoriasis-associated changes in epidermal keratinocytes, such as the upregulation of the calprotectin proteins S100A8 and S100A9. The aim of this study was to examine the availability of S100A8/A9 protein levels in SC as a biomarker of psoriasis disease activity. Fifty-three patients with psoriasis, 30 with psoriasis vulgaris (PsV), and 23 with psoriatic arthritis (PsA) participated. SC cells from lesional and non-lesional skin were collected by tape-stripping. S100A8/A9 levels in serum and in SC were quantified by enzyme-linked immunosorbent assay and compared with PASI score before and after treatment initiation or switching. Atopic dermatitis (AD) patients and disease-free individuals were used as controls. Expression of S100A8/A9 in SC of lesional skin of psoriasis patients was significantly higher than in non-lesional skin or AD skin. There was no significant difference of SC S100A8/A9 levels between PsV and PsA patients. The S100A8/A9 levels in SC of psoriasis patients were significantly positively correlated with the PASI score. When patients' skin lesions cleared (PASI clear) in response to treatment, expression of S100A8/A9 in SC was no longer detectable. S100A8/A9 protein levels in SC may be available as an objective, non-invasive biomarker of psoriasis activity to complement PASI scoring.
The longitudinal time-course of dd-cfDNA after kidney transplant (KTx) is not well-described. The cut off values of dd-cfDNA in KTx derive from biopsy-coupled single measurements. Meaningful interpretation necessitates understanding of (1) time variance of dd-cfDNA levels post-KTx, (2) factors determining biologic variability, and (3) relationship to donor and recipient characteristics. We hypothesized that an understanding of the aforementioned factors would better inform clinical decision-making using dd-cfDNA.

One hundred and twenty five KTx patients with dd-cfDNA obtained longitudinally were included. Univariate analyses were directed at inter-patient variability and intra-patient inter-occasion variability of dd-cfDNA. Multivariate linear regression was used in analyses accounting for repeat measures.

At 1-month post KTx median dd-cfDNA (1) were higher in repeat KTx (.57%, P<.001), and dual KTx (1.10%, P=ns) versus a first KTx (.31%); (2) showed a significant difference in donor after cardiac death (DCD [.45%]) versus living related (LRD [.27%]) donors (P=.036). Longitudinal (1-3months) dd-cfDNA measurements showed a significant downtrend for all donor types. Panel-reactive antibodies (PRA) were positively correlated with dd-cfDNA.

Previous iterations of National Blood Collection and Utilization Survey (NBCUS) have demonstrated declines in blood collection and transfusion in the United States since 2008, including declines of 3.0% and 6.1% in red blood cell (RBC) collections and transfusions between 2015 and 2017, respectively. This study describes results of the 2019 NBCUS.

The survey was distributed to all US blood collection centers, all hospitals performing ≥1000 surgeries annually, and a 40% random sample of hospitals performing 100-999 surgeries annually. Weighting and imputation were used to generate national estimates for units of blood and components collected, distributed, transfused, and outdated.

In 2019, 11,590,000 RBC units were collected (95% confidence interval [CI], 11,151,000-12,029,000 units), a 5.1% decrease compared with 2017, while 10,852,000 RBC units were transfused (95% CI, 10,444-11,259 units), a 2.5% increase from 2017. Between 2017 and 2019, platelet distributions (2,508,000 units; 95% CI, 2,375,000-2,6trends in blood supply safety and availability.
Neurofeedback is considered a promising intervention for the treatment of attention-deficit hyperactivity disorder (ADHD). NEWROFEED is a prospective, multicentre, randomized (32), reference drug-controlled trial in children with ADHD aged between 7 and 13years. The main objective of NEWROFEED was to demonstrate the noninferiority of personalized at-home neurofeedback (NF) training versus methylphenidate in the treatment of children with ADHD.

The NF group (n=111) underwent eight visits and two treatment phases of 16 to 20 at-home sessions with down-training of the theta/beta ratio (TBR) for children with high TBR and enhancing the sensorimotor rhythm (SMR) for the others. The control group (n=67) received optimally titrated long-acting methylphenidate. The primary endpoint was the change between baseline and endpoint in the Clinician ADHD-RS-IV total score in the per-protocol population (90 NF/59 controls).

Our study failed to demonstrate noninferiority of NF versus methylphenidate (mean between-group difference 8.09 90% CI [8.09; 10.56]). However, both treatment groups showed significant pre-post improvements in core ADHD symptoms and in a broader range of problems. Reduction in the Clinician ADHD-RS-IV total score between baseline and final visit (D90) was 26.7% (SMD=0.89) in the NF and 46.9% (SMD=2.03) in the control group. Emissions were modeled using an inverse diffusion model in conjunction with measured concentrations and turbulence. The potential for lagoon mixing was estimated by the Brunt frequency using a theoretical Fourier series temperature profile model constructed from measured air and lagoon temperatures. Annual H2 S emissions from the dairy parlor and holding area liquid waste, based on 318 d of measurement, were 212 g m-2 or 807 g head of cattle-1 . Hydrogen sulfide emissions were highest in the spring and the fall. Eleven days with emissions >7 g d-1 head-1 accounted for 25% of the annual emissions. Shear mixing appeared to dominate the mixing in the lagoon when the lagoon was estimated to be nearly isothermal. Wind shear correlated with significantly greater daily mean emissions. The H2 S emissions from this lagoon appeared to result from a series of processes; biogenic production of H2 S in the sludge, H2 S-enriched bubbles rise through the lagoon by buoyancy and wind shear induced mixing, and bubbles bursting at the surface either due to desiccation of the bubbles or surface disturbances induced by wind and precipitation.Salinity stress results in significant losses in plant productivity and loss of cultivable lands. Although Pongamia pinnata is reported to be a salt-tolerant semiarid biofuel tree, the adaptive mechanisms to saline environments are elusive. Despite a reduction in carbon exchange rate (CER), the unchanged relative water content provides no visible salinity induced symptoms in leaves of hydroponic cultivated Pongamia seedlings for 8 days. Our Na+ -specific fluorescence results demonstrated that there was an effective apoplastic sodium sequestration in the roots. Salinity stress significantly increased zeatin (~5.5-fold), and jasmonic acid (~3.8-fold) levels in leaves while zeatin (~2.5-fold) content increased in leaves as well as in roots of salt-treated plants. Metabolite analysis suggested that osmolytes such as myo-inositol and mannitol were enhanced by ~12-fold in leaves and roots of salt-treated plants. Additionally, leaves of Pongamia showed a significant enhancement in carbohydrate content, while fatty acids were accumulated in roots under salt stress condition. At the molecular level, salt stress enhanced the expression of genes related to transporters, including the Salt Overly Sensitive 2 gene (SOS2), SOS3, vacuolar-cation/proton exchanger, and vacuolar-proton/ATPase exclusively in leaves, whereas the Sodium Proton Exchanger1 (NHX1), Cation Calcium Exchanger (CCX), and Cyclic Nucleotide Gated Channel 5 (CNGC5) were up-regulated in roots. Antioxidant gene expression analysis clearly demonstrated that peroxidase levels were significantly enhanced by ~10-fold in leaves, while Catalase and Fe-superoxide Dismutase (Fe-SOD) genes were increased in roots under salt stress.
