Using a mixed electronic-ionic device model based on experimentally determined parameters, the current-voltage characteristics of single-carrier devices are accurately reproduced. Our results reveal that in our solution processed MAPbI3 thin films transport of electrons dominates over holes. Furthermore, we show that the direction of the hysteresis in the current-voltage characteristics provides a fingerprint for the sign of the dominant moving ionic species.Phenotypic plasticity, the ability of a genotype of producing different phenotypes when exposed to different environments, may impact ecological interactions. We study here how within-individual plasticity in Moricandia arvensis flowers modifies its pollination niche. During spring, this plant produces large, cross-shaped, UV-reflecting lilac flowers attracting mostly long-tongued large bees. However, unlike most co-occurring species, M. arvensis keeps flowering during the hot, dry summer due to its plasticity in key vegetative traits. Changes in temperature and photoperiod in summer trigger changes in gene expression and the production of small, rounded, UV-absorbing white flowers that attract a different assemblage of generalist pollinators. This shift in pollination niche potentially allows successful reproduction in harsh conditions, facilitating M. arvensis to face anthropogenic perturbations and climate change.Cooperative breeding may be selected for in animals when, on average, it confers greater benefits than solitary breeding. In a number of eusocial insects (i.e., ants, bees, wasps, and termites), queens join together to co-create new nests, a phenomenon known as colony co-founding. It has been hypothesised that co-founding evolved because queens obtain several fitness benefits. However, in ants, previous work has suggested that co-founding is a random process that results from high queen density and low nest-site availability. We experimentally examined nest-founding behaviour in the black garden ant, Lasius niger. We gave newly mated queens the choice between two empty nesting chambers, and compared their distribution across the two chambers with that expected under random allocation. We found that queens formed associations of various sizes; in most instances, queens group together in a single chamber. Across all experiments, the frequency of larger groups of queens was significantly higher than expected given random assortment. These results indicate colony co-founding in ants may actually be an active process resulting from mutual attraction among queens. That said, under natural conditions, ecological constraints may limit encounters among newly mated queens.By time-of-flight (TOF) neutron diffraction experiments, the influence of segregation-induced microstructure bands of austenite (γ) and martensite (α' ) phases on the partitioning of stress and strain between these phases was investigated. Initially, tensile specimens of a Co-added stainless steel were heat treated by quenching and partitioning (Q&P) processing. Tensile specimens were subsequently loaded at 350 °C parallel to the length of the bands within the apparent elastic limit of the phase mixture. Lattice parameters in both axial and transverse directions were simultaneously measured for both phases. The observation of a lattice expansion for the γ phase in the transverse direction indicated a constraint on the free transverse straining of γ arising from the banded microstructure. The lateral contraction of α' imposed an interphase tensile microstress in the transverse direction of the γ phase. The multiaxial stress state developed in the γ phase resulted in a large deviation from the level of plastic strain expected for uniaxial loading of single phase γ. Since segregation-induced banded microstructures commonly occur in many engineering alloys, the analysis of stress and strain partitioning with the present Q&P steel can be used to interpret the observations made for further engineering alloys with two-phase microstructures.Whether the presence of adsorbates increases or decreases thermal conductivity in metal-organic frameworks (MOFs) has been an open question. Here we report observations of thermal transport in the metal-organic framework HKUST-1 in the presence of various liquid adsorbates water, methanol, and ethanol. Experimental thermoreflectance measurements were performed on single crystals and thin films, and theoretical predictions were made using molecular dynamics simulations. We find that the thermal conductivity of HKUST-1 decreases by 40 - 80% depending on the adsorbate, a result that cannot be explained by effective medium approximations. Our findings demonstrate that adsorbates introduce additional phonon scattering in HKUST-1, which particularly shortens the lifetimes of low-frequency phonon modes. As a result, the system thermal conductivity is lowered to a greater extent than the increase expected by the creation of additional heat transfer channels. Finally, we show that thermal diffusivity is even more greatly reduced than thermal conductivity by adsorption.Psoriatic arthritis (PsA) is a chronic musculoskeletal inflammatory disease found in up to 30% of psoriasis patients. Prolargin-an extracellular matrix (ECM) protein present in cartilage and tendon-has been previously shown elevated in serum of patients with psoriasis. ECM protein fragments can reflect tissue turnover and pathological changes; thus, this study aimed to develop, validate and characterize a novel biomarker PROM targeting a matrix metalloproteinase (MMP)-cleaved prolargin neo-epitope, and to evaluate it as a biomarker for PsA. #link# A competitive ELISA was developed with a monoclonal mouse antibody; dilution- and spiking-recovery, inter- and intra-variation, and accuracy were evaluated. Serum levels were evaluated in 55 healthy individuals and 111 patients diagnosed with PsA by the CASPAR criteria. Results indicated that the PROM assay was specific for the neo-epitope. Inter- and intra- assay variations were 11% and 4%, respectively. PROM was elevated (p = 0.0003) in patients with PsA (median 0.24, IQR 0.19-0.31) compared to healthy controls (0.18; 0.14-0.23) at baseline. AUROC for separation of healthy controls from PsA patients was 0.674 (95% CI 0.597-0.744, P  less then  0.001). In conclusion, MMP-cleaved prolargin can be quantified in serum by the PROM assay and has the potential to separate patients with PsA from healthy controls.Antibiotics that interfere with translation, when combined, interact in diverse and difficult-to-predict ways. link2 Here, we explain these interactions by "translation bottlenecks" points in the translation cycle where antibiotics block ribosomal progression. To elucidate the underlying mechanisms of drug interactions between translation inhibitors, we generate translation bottlenecks genetically using inducible control of translation factors that regulate well-defined translation cycle steps. These perturbations accurately mimic antibiotic action and drug interactions, supporting that the interplay of different translation bottlenecks causes these interactions. We further show that growth laws, combined with drug uptake and binding kinetics, enable the direct prediction of a large fraction of observed interactions, yet fail to predict suppression. However, varying two translation bottlenecks simultaneously supports that dense traffic of ribosomes and competition for translation factors account for the previously unexplained suppression. Here, we show that the AHR pathway is selectively active in IDO/TDO-overexpressing tumors and is associated with resistance to immune checkpoint inhibitors. We demonstrate that IDO-Kyn-AHR-mediated immunosuppression depends on an interplay between Tregs and tumor-associated macrophages, which can be reversed by AHR inhibition. Selective AHR blockade delays progression in IDO/TDO-overexpressing tumors, and its efficacy is improved in combination with PD-1 blockade. Our findings suggest that blocking the AHR pathway in IDO/TDO expressing tumors would overcome the limitation of single IDO or TDO targeting agents and constitutes a personalized approach to immunotherapy, particularly in combination with immune checkpoint inhibitors.One major bottleneck in natural product drug development is derivatization, which is pivotal for fine tuning lead compounds. Although intense studies have established various methodologies for protein engineering of type I modular polyketide synthase(s) (PKSs), the accurate targeting of desired regions in the PKS gene is still challenging due to the high sequence similarity between its modules. Here, we report an innovative technique that adapts in vitro Cas9 reaction and Gibson assembly to edit a target region of the type I modular PKS gene. Proof-of-concept experiments using rapamycin PKS as a template show that heterologous expression of edited biosynthetic gene clusters produced almost all the desired derivatives. Our results are consistent with the promiscuity of modular PKS and thus, our technique will provide a platform to generate rationally designed natural product derivatives for future drug development.The central role played by cerebrospinal-fluid (CSF) examinations including antineuronal autoantibody (Ab) testing is increasingly recognized in psychiatry. The rationale of this study was to present a multimodally investigated group of patients. In total, 992 patients were analyzed for CSF alterations 456 patients with schizophreniform and 536 with affective syndromes. Ab measurement included testing for established antineuronal IgG-Abs against intracellular antigens in serum (Yo/Hu/Ri/cv2[CRMP5]/Ma1/Ma2/SOX1/TR[DNER]/Zic4/amphiphysin/GAD65) and for cell surface antigens in the CSF (NMDAR/AMPA-1/2-R/GABA-B-R/LGI1/CASPR2/DPPX). In 30 patients with "red flags" for autoimmune psychosis, "tissue tests" were performed. Additional diagnostics included MRI and EEG analyses. CSF white-blood-cell counts were increased in 4% and IgG indices in 2%; CSF-specific oligoclonal bands were detected in 4%; overall, 8% displayed signs of neuroinflammation.
