Colorectal cancer (CRC) is prevalent worldwide and novel diagnostic and prognostic biomarkers are needed to improve precision medicine. Circular RNAs (circRNAs) are currently being considered as emerging tumor biomarkers. Herein, we aimed to explore the possible clinical application of circRNAs in the early diagnosis and prognostic prediction of CRC. First, candidate circRNA was selected by integrating analysis of Gene Expression Omnibus (GEO) database using GEO2R program. ROC curve analysis demonstrated the predictive values and likelihood ratios of circ_001659 were satisfactory for the diagnosis of CRC, including patients in early-stage disease or patients with carcinoembryonic antigen (CEA)-negative status. Moreover, serum circ_001659 may be a novel biomarker in the assessment of successful treatment and remission of cancer tracking. We further investigated the oncogenic role of circ_001659. In vivo and in vitro experiments indicated that circ_001659 could promote CRC cell invasion and migration. Mechanistically, circ_001659 was localized in the nucleus, recruited the RBBP5 to Vimentin promoter and increased H3K4 trimethylation level on the Vimentin promoter region, which epigenetically activated Vimentin transcription. Our findings demonstrate that circ_001659 could be a useful serum biomarker for CRC diagnosis and prognosis. Targeting circ_001659 and its pathway may be meaningful for treating patients with CRC.Oenothera drummondii is a native species from the coastal dunes of the Gulf of Mexico that has nowadays extended to coastal areas in temperate zones all over the world, its invasion becoming a significant problem locally. The species grows on back beach and incipient dunes, where it can suffer flooding by seawater, and sea spray. TMP269 mw We were interested in knowing how salinity affects this species and if invasive populations present morphological or functional traits that would provide greater tolerance to salinity than native ones. To this end, we conducted a greenhouse experiment where plants from one native and from one invading population were irrigated with five salinity treatments. We measured functional traits on photosynthetic, photochemical efficiency, water content, flowering, Na+ content, pigment content, and biomass. Although O. drummondii showed high resistance to salinity, the highest levels recorded high mortality, especially in the invasive population. Plants exhibited differences not only in response to time under salinity conditions, but also according to their biogeographic origin, the native population being more resistant to long exposure and high salt concentration than the invasive one. Native and invasive populations showed different response to salt stress in photosynthesis and transpiration rates, stomatal conductance, water use efficiency, carboxylation efficiency, electron transport rate, electron transport efficiency, energy used in photochemistry, among others. The increasing salinity levels resulted in a progressive reduction of photosynthesis rate due to both stomatal and biochemical limitations, and also in a reduction of biomass and number and size of flowers, compromising the reproductive capacity.This is an introductory chapter for the Special Issue Ex vivo dual perfusion of an isolated cotyledon of the human placenta. Uteroplacental acute atherosis is frequently observed in preeclampsia, and shares features with early atherosclerotic lesions, including artery wall foam cells. The lipid-associated proteins FABP4 (fatty acid binding protein 4), perilipin-2, and LOX-1 (lectin-like oxidized LDL-receptor 1) are involved in atherosclerotic foam cell formation. Increased levels of these proteins have been associated with preeclampsia systemically and in placental tissue. Their role in acute atherosis is yet unidentified. Our aim was to describe the presence of these proteins in acute atherosis, and compare our findings to what is known in early atherosclerotic lesions.

Serial sections of decidua basalis tissue from 12 normotensive (4 with acute atherosis) and 23 preeclamptic pregnancies (16 with acute atherosis) were stained with HE and immunostained for CK7, CD68, FABP4, perilipin-2, and LOX-1. Artery wall and perivascular protein expression was assessed in 190 spiral artery sections; 55 with acute atherosis.

Acute atherosis foam cell generation.Loneliness is common in youth, with suggestions that these negative emotions confer vulnerability for anxiety and depression. Here, we investigated for the first time whether, consistent with psychological models of loneliness, biased interpretations of social situations could prospectively predict loneliness in youth. 104 young people completed measures of loneliness and interpretations of ambiguous social and non-social (bodily or health-related) situations at three time-points with intervals of three months between each. As government-imposed social distancing measures (to control the COVID-19 outbreak) occurred between Times 2 and 3 (but not between Times 1 and 2), this enabled us to assess whether restricted social activity could provoke greater predictive power of biased interpretational styles on loneliness. Using cross-lagged panel models, we showed that after estimating paths representing within-time across-variable ("concurrent") paths and across-time within-variable ("stability") paths, there were no significant cross-lag 'causal' paths between earlier interpretational style and later loneliness. Between Time 2 and 3, we demonstrated a significant cross-lag 'consequential' path between earlier loneliness and later threatening interpretations of social situations, but this became non-significant after controlling for concurrent anxiety and depression. Biased interpretational style may reflect a concurrent maintenance factor of youth loneliness.Suspended mariculture has significantly influences on the benthic sediment. However, our understanding on how bacterial communities respond to mariculture induced changes in redox profiles is limited. In present study, sediments from two maricultures and reference areas were collected and incubated for 28 day. The results indicated that the dominant pathway of organic matter mineralization in the sediment varied from groups, in the reference, it was the iron reduction, but in the two mariculture groups it was the SO42- reduction. Remarkable changes of bacteria community were recorded in the aerobic zone, where the abundances of 14 OTUs belonging to Gammaproteobacteria and Alphaproteobacteria were significantly higher than that in oxidation and anaerobic zones. However, 4 keystone OTUs were strictly anaerobic and belonging to Desulfobacteraceae (n = 3) and Marinilabiaceae (n = 1). The main environmental drivers determining sediment bacterial distribution were the particle organic carbon, dissolve oxygen, NO3-, and moisture content.The level of heavy metals (Cr, Co, Ni, Cu, Zn, and Pb) and Al2O3 were quantified in 140 surface sediment samples from the continental shelf of Hainan Island (CSHI). The mean heavy metal concentration in the decreasing order was Cr > Zn > Pb > Ni > Cu > Co. Although heavy metals were locally enriched in the CSHI, the overall pollution level was relatively low. The biohazard assessment results of the mean effects range-median quotient (M-ERM-Q) and hazard quotient (HQ) for a single metal element (Cu, Pb, Zn) in the surface sediments showed that the exposure to individual heavy metals posed a low health risk. However, the biohazard assessment of multiple metals was higher than the single metals. Particular attention should be paid to the potential biological hazard from Cr and Ni in the CSHI.We report the discovery of a dead Magellanic penguin (Spheniscus magellanicus) found on Juquehy Beach (23°46'S 45°44'W), municipality of São Sebastião, Brazil, on September 9th 2020. Following necropsy, we noted the presence of an adult size PFF-2 protective mask within the stomach of the penguin which we inferred as the cause of death. As far as we are aware, this is the first recorded instance of marine animal mortality by protective face mask ingestion. Whilst concerns have been raised relating microplastic contamination in marine environments from Covid-19 related waste, there has been relatively less attention paid to the potential risk of macro-scale contaminants, such as protective face coverings. We suggest that Covid-19 related macro contaminants be considered in coastal marine risk assessments and urge further research on this topic.Thieno[2,3-b]pyridines are a class of compounds known for their potent anti-proliferative activities against a range of human cancer cell lines. In this research, a number of strategies to generate analogues that have improved aqueous solubility whilst retaining the potent anti-proliferative actions, compared to previously-explored compounds in this class, were made. Herein we report the synthesis of 80 novel compounds, comprising two series, all based on the thieno[2,3-b]pyridine core structure. Overall, it was found that introducing alternative heterocycles did not notably improve the solubility or retain anti-proliferative activity seen in previously-reported analogues. However, pleasingly it was discovered, that the best strategy for improving the solubility was the alteration of the appended alkyl ring to introduce polar groups such as alcohols, ketones and substituted amine groups. In addition to this finding, we have discovered a thieno[2,3-b]pyridine, 15e, with greater aqueous solubility that has ever been seen for this class of compounds that is also a potent inhibitor of cancer cell growth, with IC50's in the nanomolar range. This new lead structure will form the basis of future explorations into this class of compounds.Faecal carriage of extended-spectrum cephalosporin-resistant Escherichia coli (ESC-R E. coli) in dogs has been reported worldwide and can reduce the effectiveness of treatments against bacterial infections. However, the drivers that influence faecal carriage of ESC-R E. coli in dogs are poorly understood. The aims of this study were to estimate the prevalence of ESC-R E. coli among dogs prior to their admission to a veterinary teaching hospital and to identify risk factors associated with the faecal carriage of ESC-R E. coli. Rectal swabs (n = 130) were collected from dogs and screened for ESC-R E. coli using MacConkey agar supplemented with cefotaxime (2 μg/mL). E. coli species was confirmed by MALDI-TOF and screening of extended-spectrum beta-lactamase (ESBL) genes was conducted by multiplex PCR. Questionnaires were completed by each dog's owner to test several human and dog characteristics associated with ESC-R E. coli. The prevalence of faecal carriage of ESC-R E. coli was 9.2 % and 67 % of ESC-R E. coli isolates harboured ESBL genes including CTX-M alone or in combination with TEM.
