Treatment of FN-RMS cells in vitro with the recently described HES1 small-molecule inhibitor, JI130, limited FN-RMS cell growth. Inhibition of HES1 in vivo via conditional expression of a HES1-directed shRNA or JI130 dosing impaired FN-RMS tumor xenograft growth. Lastly, targeted transcriptomic profiling of FN-RMS xenografts in the context of HES1 suppression identified associations between HES1 and RAS-MAPK signaling. In summary, these in vitro and in vivo preclinical studies support the further investigation of HES1 as a therapeutic target in FN-RMS.[This corrects the article DOI 10.1021/acsaem.2c00977.].
Antiretroviral therapy (ART) is remarkably effective to prevent perinatal transmission (PT) of HIV-1. We evaluated the PT rate in a population of women with widespread access to ART before conception.

The analysis included 14630 women living with HIV-1 delivering from 2000 to 2017 in centers participating in the nationwide prospective multicenter French Perinatal Cohort (ANRS-EPF). PT was analyzed according to time period, timing of ART initiation, maternal plasma viral load (pVL), and gestational age at birth. No infants were breastfed and all received neonatal prophylaxis.

PT decreased between the three periods, from 1.1% in 2000-2005 (58/5123), to 0.7% in 2006-2010 (30/4600), and 0.2% in 2011-2017 (10/4907; p < 0.001). Restricting the analysis to the 6316/14630 (43%) women on ART at conception, PT decreased from 0.42% (6/1434) in 2000-2005 to 0.03% (1/3117) in 2011-2017 (p = 0.007). Among women treated at conception, if maternal pVL was undetectable near delivery, no PT was observed whatever the ART combination, [95%CI 0-0.07] (0/5482). Among women starting ART during pregnancy and with undetectable pVL near delivery, PT was 0.57% [95%CI 0.37-0.83] (26/4596). Among women treated at conception but having a detectable pVL near delivery, PT was 1.08% [95%CI 0.49-2.04] (9/834). Bozitinib c-Met inhibitor We also qualitatively described the 10 cases of transmission occurring during the 2011-2017 period.

In a setting with free access to ART, monthly pVL assessment, infant ART prophylaxis, and in the absence of breastfeeding, suppressive ART initiated before pregnancy and continued throughout the pregnancy can reduce perinatal transmission of HIV to almost zero.

WHO guidelines strongly recommend a single high-dose of liposomal amphotericin B as part of preferred treatment, but this drug remains unaffordable in most low- and middle-income countries. A proactive approach is needed from manufacturers and other stakeholders to improve access. The skeletal system is generated and maintained by its progenitors, skeletal stem cells (SSCs), across the duration of life. Gradual changes associated with aging result in significant differences in functionality of SSCs. Declines in bone and cartilage production, increase of bone marrow adipose tissue, compositional changes of cellular microenvironments, and subsequent deterioration of external and internal structures culminate in the aged and weakened skeleton. The features and mechanisms of skeletal aging, and of its stem and progenitor cells in particular, are topics of recent investigation. The discovery of functionally homogeneous SSC populations with a defined cell surface phenotype has allowed for closer inspection of aging in terms of its effects on transcriptional regulation, cell function, and identity. Here, we review the aspects of SSC aging on both micro- and macroscopic levels. Up-to-date knowledge of SSC biology and aging is presented, and directions for future research and potential therapies are discussed. The realm of SSC-mediated bone aging remains an important component of global health and a necessary facet in our understanding of human aging.Detection of bare gas microbubbles by magnetic resonance (MR) at low concentrations typically used in clinical contrast-ultrasound studies was recently demonstrated using hyperCEST. Despite the enhanced sensitivity achieved with hyperCEST, in vivo translation is challenging as on-resonance saturation of the gas-phase core of microbubbles consequently results in saturation of the gas-phase hyperpolarized 129 Xe within the lungs. Alternatively, microbubbles can be condensed into the liquid phase to form perfluorocarbon nanodroplets, where 129 Xe resonates at a chemical shift that is separated from the gas-phase signal in the lungs. For ultrasound applications, nanodroplets can be acoustically reverted back into their microbubble form to act as a phase-change contrast agent. Here, we show that low-boiling point perfluorocarbons, both in their liquid and gas form, generate phase-dependent hyperCEST contrast. Magnetic resonance detection of ultrasound-mediated phase transition demonstrates that these perfluorocarbons could be used as a dual-phase dual-modality MR/US contrast agent.Quercetin is the most abundant polyphenolic flavonoid (flavonol subclass) in vegetal foods and medicinal plants. This dietary chemopreventive agent has drawn significant interest for its multiple beneficial health effects ("polypharmacology") largely associated with the well-documented antioxidant properties. However, controversies exist in the literature due to its dual anti-/pro-oxidant character, poor stability/bioavailability but multifaceted bioactivities, leaving much confusion as to its exact roles in vivo. Increasing evidence indicates that a prior oxidation of quercetin to generate an array of chemical diverse products with redox-active/electrophilic moieties is emerging as a new linkage to its versatile actions. The present review aims to provide a comprehensive overview of the oxidative conversion of quercetin by systematically analyzing the current quercetin-related knowledge, with a particular focus on the complete spectrum of metabolite products, the enzymes involved in the catabolism and the underlying molecular mechanisms. Herein we review and compare the oxidation pathways, protein structures and catalytic patterns of the related metalloenzymes (phenol oxidases, heme enzymes and specially quercetinases), aiming for a deeper mechanistic understanding of the unusual biotransformation behaviors of quercetin and its seemingly controversial biological functions. The study of science popularization in dictatorships, such as Franco's regime, offers a useful window through which to review definitions of controversial categories such as "popular science" and the "public sphere." It also adds a new analytical perspective to the historiography of dictatorships and their totalitarian nature. Moreover, studying science popularization in these regimes provides new tools for a critical analysis of key contemporary concepts such as nationalism, internationalism, democracy, and technocracy. In the late 1940s in Spain, a group of young scholars, most of them newly appointed university lecturers, gained control of Arbor, the promotional journal of the Consejo Superior de Investigaciones Científicas (CSIC The Spanish National Research Council), the institution that General Franco had founded after the Spanish Civil War (1936-9) to organize Spanish science. This group constituted the intellectual core of the more reactionary, Catholic traditionalist faction of Franco's regime, and they coveted greater political power, in competition with other factions of the regime. Lacking the opportunity to launch an overt political campaign within a dictatorship, the group started a fight for the cultural conquest of Spain. In this cultural struggle for hegemony, journals, magazines, cultural associations, publishing houses, newspapers, and cultural centers became their weapons. By analyzing this faction's views on and activities within the popularization of science, particularly regarding theories of evolution, this article argues that popular discourse on science played a critical role in the cultural struggle both as a "safe" channel in which to forward their claims and as a tool to gather popular attention through topics of general interest. A covert political campaign was conducted through the popularization of science and this, in turn, fueled the construction of a public sphere for science in a dictatorial context. Scientific popularization became a much-appreciated tool to achieve cultural hegemony and, as such, it also became a central element in constructing and legitimating the ideological foundations of Franco's regime.A sulfhydryl monomicelles interfacial assembly strategy is presented for the synthesis of fully exposed single-atom-layer Pt clusters on 2D mesoporous TiO2 (SAL-Pt@mTiO2 ) nanosheets. This synthesis features the introduction of the sulfhydryl group in monomicelles to finely realize the controllable co-assembly process of Pt precursors within ordered mesostructures. The resultant SAL-Pt@mTiO2 shows uniform SAL Pt clusters (≈1.2 nm) anchored in ultrathin 2D nanosheets (≈7 nm) with a high surface area (139 m2  g-1 ), a large pore size (≈25 nm) and a high dispersion (≈99 %). Moreover, this strategy is universal for the synthesis of other SAL metal clusters (Pd and Au) on 2D mTiO2 with high exposure and accessibility. When used as a catalyst for hydrogenation of 4-nitrostyrene, the SAL-Pt@mTiO2 shows a high catalytic activity (TOF up to 2424 h-1 ), 100 % selectivity for 4-aminostyrene, good stability, and anti-resistance to thiourea poisoning under relatively mild conditions (25 °C, 10 bar).
To provide useful insights into the measles elimination progress in China, measles surveillance data was reviewed, and the transmission patterns of measles viruses circulating in China during 1993-2021 were analyzed.

Measles incidence data from the National Notifiable Disease Reporting System of the China Center for Disease Control and Prevention was analyzed. A total of 17,570 strains were obtained from 30 of 31 provinces in mainland China during 1993-2021. The recommended genotyping window was amplified. Genotyping analysis was conducted in comparison with the reference strains. Furthermore, phylogenetic analyses were performed to identify genetic relationships among different lineages within the genotypes.

With high coverage of routine immunization and intensive supplementary immunization activities, measles incidence has shown a downward trend since 1993, despite two resurgences, reaching a historic low level in 2020-2021 (average 0.5 per million). During 1993-2021, nine genotypes including domestic genotype H1, imported genotypes B3, D4, D8, D9, D11, G3, and H2, and vaccine-associated genotype A were identified. Among them, genotype H1 strain circulated endemically in China for over 25 years; the last strain was detected in Yunnan province in September 2019. Multiple imported genotypes were identified since 2009 showing different transmission patterns. Since April 2020, no imported strains have been detected, while vaccine-associated genotype A continues to be detected.

The evidence of low incidence during 2020-2021 and virological surveillance data in this study confirmed that China is currently approaching measles elimination.
