Sepsis is characterized by organ dysfunction resulting from a patient's dysregulated response to infection. Sepsis-associated acute kidney injury (S-AKI) is the most frequent complication contributing to the morbidity and mortality of sepsis. The prevention and treatment of S-AKI remains a significant challenge worldwide. In the recent years, human amnion epithelial cells (hAECs) have drawn much attention in regenerative medicine, yet the therapeutic efficiency of hAECs in S-AKI has not been evaluated.
Septic mice were induced by cecal ligation and puncture (CLP) operation. hAECs and their derived exosomes (EXOs) were injected into the mice
tail vein right after CLP surgery. The 7-day survival rate was observed. Serum creatinine level was measured and H&E staining of tissue sections were performed 16 h after CLP. Transmission electron microscopy was used to examine the renal endothelial integrity in CLP mice. However, existing methods for computing the characteristic average and corresponding response corridors of experimental data are often customized to specific types or shapes of signal and therefore limited in general applicability. In addition, simple methods such as point-wise averaging can distort or misrepresent important features if signals are not well aligned and highly correlated. In this study, an improved method of computing the characteristic average and response corridors of a set of experimental signals is presented based on arc-length re-parameterization and signal registration. The proposed arc-length corridor method was applied to three literature datasets demonstrating a range of characteristics common to biomechanical data, such as monotonic increasing force-displacement responses with variability, oscillatory acceleration-time signals, and hysteretic load-unload data. The proposed method addresses two challenges in assessing experimental data arc-length re-parameterization enables the assessment of complex-shaped signals, including hysteretic load-unload data, while signal registration aligned signal features such as peaks and valleys to prevent distortion when determining the characteristic average response. The arc-length corridor method was shown to compute the characteristic average and response corridors for a wide range of biomechanical data, while providing a consistent statistical framework to characterize variability in the data. No study has demonstrated the specific differences between habitually barefoot and shod cohorts based on foot morphology and dynamic plantar pressure during walking and running. The present study aimed to assess and classify foot metrics and dynamic plantar pressure patterns of barefoot and shod people via machine learning algorithms. One hundred and forty-six age-matched barefoot (n = 78) and shod (n = 68) participants were recruited for this study. Gaussian Naïve Bayes were selected to identify foot morphology differences between unshod and shod cohorts. The support vector machine (SVM) classifiers based on the principal component analysis (PCA) feature extraction and recursive feature elimination (RFE) feature selection methods were utilized to separate and classify the barefoot and shod populations via walking and running plantar pressure parameters. Peak pressure in the M1-M5 regions during running was significantly higher for the shod participants, increasing 34.8, 37.3, 29.2, 31.7, and 40.1%, respectively. The test accuracy of the Gaussian Naïve Bayes model achieved an accuracy of 93%. The mean 10-fold cross-validation scores were 0.98 and 0.96 for the RFE- and PCA-based SVM models, and both feature extract-based and feature select-based SVM models achieved an accuracy of 95%. The foot shape, especially the forefoot region, was shown to be a valuable classifier of shod and unshod groups. This study aims to address this gap by evaluating different modelling and post-processing approaches in simulations of a patient-specific aorta. Methods Magnetic resonance imaging (MRI) and 4D flow MRI from a patient with aortic valve stenosis were used to reconstruct the aortic geometry and derive patient-specific inlet and outlet boundary conditions. Three different computational approaches were considered based on assumed laminar or assumed disturbed flow states including low-resolution laminar (LR-Laminar), high-resolution laminar (HR-Laminar) and large-eddy simulation (LES). Each simulation was ran for 30 cardiac cycles and post-processing was conducted on either the final cardiac cycleR-Laminar simulation over an LES simulation based on their comparable computational cost. It has attracted much attention in recent years owing to its ability to create complex vascular network-like channels through thick tissue constructs while maintaining endothelial cell activity. Biodegradable materials play a crucial role in tissue engineering. Placental dysfunction contributes to major obstetric complications, such as fetal growth restriction and preeclampsia. Currently, there is no effective treatment for placental dysfunction in the perinatal period, and prophylaxis is often delivered too late, at which point the disease manifestation cannot be prevented. However, with recent integration of nanoscience and medicine to perform elaborate experiments on the human placenta, it is expected that novel and efficient nanotherapies will be developed to resolve the challenge of managing placental dysfunction. The advent of nanomedicine has enabled the safe and targeted delivery of drugs using nanoparticles. These smart nanoparticles can load the necessary therapeutic substances that specifically target the placenta, such as drugs, targeting molecules, and ligands. Packaging multifunctional molecules into specific delivery systems with high targeting ability, diagnosis, and treatment has emerged as a novel theragnostic (both therapeutic and diagnostic) approach. In this review, the authors discuss recent advances in nanotechnology for placental dysfunction treatment. In particular, the authors highlight potential candidate nanoparticle-loaded molecules that target the placenta to improve utero-placental blood flow, and reduce reactive oxygen species and oxidative stress. The authors intend to provide basic insight and understanding of placental dysfunction, potential delivery targets, and recent research on placenta-targeted nanoparticle delivery systems for the potential treatment of placental dysfunction. The authors hope that this review will sensitize the reader for continued exploration of novel nanomedicines.
