To investigate the molecular mechanism by which a novel naphthalene allyl trifluoromethyl benzocyclopentanone XX0335 inhibits the proliferation and induces apoptosis of lung cancer A549 cells.

Lung cancer A549 cells were treated with 0.1% DMSO (control) or different concentrations (6.25, 12.5, and 25 μg/mL) of XX0335, and the changes in cell viability, cell cycle, proliferation and apoptosis were assessed with CCK-8 assay, EdU experiment, and flow cytometry. The effects of different concentrations of XX0335 on phosphorylation levels of proliferation-related proteins Akt, mTOR, Akt/mTOR and the expressions of cleaved PARP and cyclin D1 were determined using Western blotting. We also assessed the effect of XX0335 on tumor growth in a mouse model bearing A945 cell xenograft.

Treatment with XX0335 reduced the viability of A549 cells in a dose-dependent manner (
< 0.01) and significantly inhibited cell proliferation (
< 0.001). Flow cytometry showed that XX0335 treatment promoted apoptosis of the cells (
< 0.01) and caused an obvious increase of the number of G1-phase cells. Compared with DMSO, XX0335 significantly inhibited the phosphorylation of Akt and mTOR, increased the expression of cleaved PARP, and lowered the protein expression of cyclin D1. In the tumor-bearing mouse models, injection of XX0335 significantly decreased the tumor volume (
< 0.01).

XX0335 inhibits the proliferation, cycle and induces apoptosis of lung cancer A549 cells possibly by inhibiting the Akt/mTOR signal pathway.

To analyze the expression of immunoglobulin mucin molecule 3 (TIM-3) in epithelial ovarian cancer (EOC) and the effects of TIM-3 knockdown and overexpression on proliferation and migration of ovarian cancer cells.

We analyzed TIM-3 expression in EOC and normal ovarian tissues using GEPIA database. We also detected TIM-3 expression levels in 82 surgical specimens of EOC and 18 specimens of normal ovarian tissues using immunohistochemistry, and analyzed the correlation of TIM-3 expression with clinicopathological parameters and survival outcomes of the patients. The expression of TIM-3 and Wnt1 mRNA in the tissues were detected using qRT-PCR. We constructed SKOV3 cell models of TIM-3 knockdown and overexpression and examined the changes in proliferation, apoptosis, migration and invasion of the cells using MTT assay, Annexin V-FITC/PI staining, scratch test and Transwell assay. The activity of Wnt/β-catenin pathway in the transfected was detected using dual luciferase reporter assay, and the mRNA levels of viors of the tumor cells possibly by activating the Wnt/β-catenin signal pathway.
TIM-3 is highly expressed in EOC tissue to promote malignant behaviors of the tumor cells possibly by activating the Wnt/β-catenin signal pathway.
To explore the effect of transforming growth factor-β (TGF-β)-activated kinase 1 (TAK1) on toluene diisocyanate (TDI)-induced allergic airway inflammation in mice.

Thirty-two mice were randomly divided into AOO group, AOO+5Z-7-Oxozeaenol group, TDI group, and TDI+5Z-7-Oxozeaenol group. Another 32 mice were randomly divided into AOO group, TDI group, TDI +5Z-7-Oxozeaenol group, and TDI +5Z-7-Oxozeaenol + Necrostatin-1 group. TAK1 inhibitor (5Z-7-Oxozeaenol, 5 mg/kg) and/or RIPK1 inhibitor (Necrostatin-1, 5 mg/kg) were used before each challenge. Airway responsiveness, airway inflammation and airway remodeling were assessed after the treatments. We also examined the effect of TDI-human serum albumin (TDI-HSA) conjugate combined with TAK1 inhibitor on the viability of mouse mononuclear macrophages (RAW264.7) using CCK8 assay. The expressions of TAK1, mitogen-activated protein kinase (MAPK) and receptor interacting serine/threonine protease 1 (RIPK1) signal pathway in the treated cells were detected with West.

Inhibition of TAK1 aggravates TDI-induced airway inflammation and hyperresponsiveness and may increase the death of macrophages by enhancing the activity of RIPK1 and causing persistent activation of caspase 8.

To study the therapeutic mechanism of
(LQF) for diabetic kidney disease (DKD) based on GEO database and network pharmacology.

LQF and DKD targets were obtained using the databases including GEO, TCMSP, CNKI, ChemDraw, and SwissTarget Prediction, and LQF-DKD intersection targets were obtained with VENNY. String was used for protein-protein interaction (PPI) analysis, and R package for KEGG and GO enrichment analysis. Cytoscape 3.7.2 software Network graphs were constructed. The results of network pharmacology analysis were verified in SD rat models of DKD by daily treatment of the rats with LQF at low (1 g/kg), medium (2 g/kg), and high (2 g/kg) doses, and kidney pathology was observed with HE staining and the changes in renal function were assessed. Western blotting was used to detect the expression levels of NF-κB and p-NF-κB proteins.

We identified 760 main targets of LQF, and obtained 1026 differential genes using GEO database and 61 LQF-DKD intersection targets using Venny database. The core targhe renal function. Objective Circular RNAs (circRNAs) are non-coding RNAs (ncRNA) circularized without a 3' polyadenylation [poly-(A)] tail or a 5' cap, resulting in a covalently closed loop structure. circRNAs were first discovered in RNA viruses in the 1970s, but only a small number of circRNAs were discovered at that time due to limitations in traditional polyadenylated transcriptome analyses. With the development of specific biochemical and computational methods, recent studies have shown the presence of abundant circRNAs in eukaryotic transcriptomes. circRNAs play vital roles in many physiological and pathological processes, such as acting as miRNA sponges, binding to RNA-binding proteins (RBPs), acting as transcriptional regulatory factors, and even serving as translation templates. Current evidence has shown that circRNAs can be potentially used as excellent biomarkers for diagnosis, therapeutic effect evaluation, and prognostic assessment of a variety of diseases, and they may also provide effective therapeutic targets due to their stability and tissue and development-stage specificity. This review focuses on the properties of circRNAs and their immune relationship to disease, and explores the role of circRNAs in immune-related diseases and the directions of future research.
The objective of this study was to establish local diagnostic reference levels (LDRLs) for the full-field digital mammography (FFDM) and tomosynthesis (DBT) in Moroccan health facilities.

Data from 146 women were collected from three facilities. The proposed DRLs were defined as the 75th percentile of the mean average glandular dose (AGD) distribution.

The mean AGD recorded in this study for the three centers was 1.47mGy for all centers, and 1.42mGy and 1.64mGy for the CC and MLO projections, respectively. The mean compressed breast thickness (CBT) values recorded in this current study were 55mm, the LDRLs reported for all centers was 1.7mGy, the CC projection was 1.6mGy, and the MLO projection was 1.8mGy. In addition, the LDRLs reported in the current study were compared with those from previous studies for other countries, including the United Kingdom, Japan, Ghana, and Sri Lanka.

This work provides an assessment of local DRLs for mammography in Morocco and is suggested as a starting point that will allow professionals to evaluate and optimize their practice. Furthermore, the definition of national DRLs is a necessary process in optimizing Moroccan medical exposures.

The aim of this study is to evaluate the effective dose and cancer risk of examinations in EOS imaging system in different age and gender groups.

In total, 555 patients who had undergone common EOS imaging examinations were entered into the study. Exposure parameters and patients' characteristics for lower limb, full spine and full body imaging techniques, at different gender and age groups, were evaluated. NexturastatA Finally, effective dose and risk of exposure induced cancer death (REID) was calculated with the Monte Carlo based PCXMC software.

The difference between average effective doses of male and female was not significant (p≥0.05), however, the corresponding REID showed statistically significant difference (p≤0.001). The average effective dose of patients (without considering technique, age and gender) was obtained as 0.13 mSv. The corresponding average REID was 8.84 per million. The maximum average effective dose value was obtained for patients over 10 years of age with the full body technique (0.17±0.05 examinations.
Although the ACGME has called for outcomes-based evaluation of residency programs, few metrics or benchmarks exist connecting educational processes with resident educational outcomes. To address this deficiency, a national Education Quality Improvement Program (EQIP) for General Surgery training is proposed.

We describe the initial efforts to create this platform. In addition, a national survey was administered to 330 Program Directors to assess their interest in and concerns about a continuous educational quality improvement project.

We demonstrate that through a collaborative process and the support of the Association of Program Directors in Surgery (APDS), we were able to develop the groundwork for a national surgical educational improvement project, now called EQIP. The survey response rate was 45.8% (152 of 332 programs) representing a mix of university (55.3%), university-affiliated (18.4%), independent (24.3%), and military (2.0%) programs. Most respondents (66.2%) had not previously heard of EQIors have expressed enthusiasm for EQIP and are willing to participate in the program examining outcomes of General Surgery training programs, with an ultimate goal of improving overall residency training.Secondary metabolism in plants gives rise to a vast array of small-molecule natural products. The discovery of operon-like gene clusters in plants has provided a new perspective on the evolution of specialized metabolism and the opportunity to rapidly advance the metabolic engineering of natural product production. Here, we review historical aspects of the study of plant metabolic gene clusters as well as general strategies for identifying plant metabolic gene clusters in the multi-omics era. We also emphasize the exploration of their natural variation and evolution, as well as new strategies for the prospecting of plant metabolic gene clusters and a deeper understanding of how their structure influences their function.
The aim of this study is to review the evidence available suggesting that environmental conditions represent a risk factor associated with non-infective acute respiratory illness in athletes.

Systematic review.

PubMed, EBSCOhost and Web of Science (1st January 1990-31 July 2020) were searched systematically using keywords related to male and female athletes (i.e. from physically active individuals to elite athletes), aged 15-65 years and a combination of the terms (non-infective acute respiratory illness AND [pollution OR allergies OR climate] AND athletes AND prevalence/incidence/risk factors).

A total of seven papers (n = 1567 athletes) addressed our question. Among these, one focused on indoor air pollution, four on chlorinated swimming pool exposure and two on cold air conditions. None was selected for allergies, outdoor air pollution or other climatic conditions. Except rhinitis induced by swimming in chlorinated pools (n = 1), no respiratory disease due to the environment was identified specifically in athletes.
