In this paper, we report the metabonomic analysis of V. volvacea in response to low temperature and compare the differences in metabolite expression between the low-temperature-resistant strain VH3 and the low-temperature-sensitive strain V23. Finally, the putative low-temperature resistance mechanism of VH3 is revealed at the metabolic level. This study provides a theoretical basis for revealing the regulatory mechanism of low-temperature resistance in V. volvacea and for future molecular breeding efforts.Population-wide tolerance and persisters enable susceptible bacterial cells to endure hostile environments, including antimicrobial exposure. The SOS response can play a significant role in the generation of persister cells, population-wide tolerance, and shielding. The SOS pathway is an inducible DNA damage repair system that is also pivotal for bacterial adaptation, pathogenesis, and diversification. In addition to the two key SOS regulators, LexA and RecA, some other stressors and stress responses can control SOS factors. Bacteria are exposed to DNA-damaging agents and other environmental and intracellular factors, including cigarette smoke, that trigger the SOS response at a number of sites within the host. The Escherichia coli TisB/IstR module is as yet the only known SOS-regulated toxin-antitoxin module involved in persister formation. Nevertheless, the SOS response plays a key role in the formation of biofilms that are highly recalcitrant to antimicrobials and can be abundant in persisters. Furthermore, the dynamic biofilm environment generates DNA-damaging factors that trigger the SOS response within the biofilm, fueling bacterial adaptation and diversification. This review highlights the SOS response in relation to antimicrobial recalcitrance to antimicrobials in four clinically significant species, Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, and Mycobacterium tuberculosis.Cryoconite holes are miniature freshwater aquatic ecosystems that harbor a relatively diverse microbial community. This microbial community can withstand the extreme conditions of the supraglacial environment, including fluctuating temperatures, extreme and varying geochemical conditions and limited nutrients. We analyzed the physiological capabilities of microbial isolates from cryoconite holes from Antarctica, Greenland, and Svalbard in selected environmental conditions extreme pH, salinity, freeze-thaw and limited carbon sources, to identify their physiological limits. In this study, we analyzed the produced water from 14 unconventional wells in the Bakken Shale using geochemical measurements, quantitative PCR (qPCR), and 16S rRNA gene sequencing with the overall goal of understanding the complex dynamics present in hydraulically fractured wells. Bakken Shale produced waters from this study exhibit high measurements of total dissolved solids (TDS). These conditions inhibit microbial growth, such that all samples had low microbial loads except for one sample (well 11), which had lower TDS concentrations and higher 16S rRNA gene copnated by sulfate reducers. Our findings expand the current understanding of microbial life in the Bakken region and provide new insights into how the unique produced water conditions shape microbial communities. Finally, our analysis suggests that produced water chemistry is tightly linked with microbiota in the Bakken Shale and shows that additional research efforts that incorporate coupled microbial and geochemical datasets are necessary to understand this ecosystem.Pastures are an important part of crop and food systems in cold climates.
