In the APE group, levels of TNF-α and IL-2 were significantly down-regulated and IL-4 and IL-10 significantly up-regulated in contrasted with that of model group. In addition, levels of TGF-β1 and bFGF in the APE group were significantly induced compared with that of model group. CONCLUSION The results suggest that the extracts from Chuanwu (Aconitum Carmichaelii) and Banxia (Rhizoma Pinelliae) promote wound healing in the rats, which is associated with enhancing anti-inflammatory ability and inducing fibroblast formation.OBJECTIVE To investigate the effect of Daiqin phlegm-expelling pill, prepared with Traditional Chinese Medicine (TCM), on the development of inflammation in Sprague-Dawley (SD) rats with chronic obstructive pulmonary disease (COPD) induced by lipopolysaccharide (LPS) and smoke, and to identity the possible underlying mechanism. METHODS Sixty male rats were divided into 6 groups (healthy control group, untreated group, Daiqin phlegm-expelling pill low, middle and high dose, and ambroxol hydrochloride tablet group). COPD was established in SD rats by sootiness and tracheal instillation with LPS. The rats were treated with Daiqin phlegm-expelling pill at the indicated doses for 28 d, the inflammatory cells in bronchoalveolar lavage fluid (BALF), the concentration of tumor necrosis factor-α (TNF-α), interleukin (IL)-8 and IL-6 in blood and the inflammation in lung were evaluated. RESULTS The number of inflammatory cells in the BALF and TNF-α, IL-8 and IL-6 level in plasma were significantly reduced in rats with COPD when treated with Daiqin phlegm-expelling pill or ambroxol hydrochloride tablet, compared with those without any treatment. The Daiqin phlegm-expelling pill treated rats with COPD had a attenuated inflammation in lung tissue, compared with the untreated group. CONCLUSION Daiqin phlegm-expelling pill can significantly restrain the airway inflammation in rats with LPS-smoke induced COPD.OBJECTIVE To examine the effects of Yokukansan (YKS) extract on two endogenous modulators of anxiety, brain-derived neurotrophic factor (BDNF) and serotonin (5-HT)2A receptors pharmacologically, in the ischemic rat model of dementia. METHODS The cerebral ischemia (CI) was induced by bilateral occlusion of the vertebral and common carotid arteries (4-vessel occlusion ischemia). The CI was combined with the amyloid-β42 peptide (Aβ42) injected intracerebroventricularly, and referred to as CI+Aβ. Anxiety-like behaviors were assessed by elevated plus maze (enclosed arm), light/dark transition test (dark chamber), and open-field test. Wet-dog shakes were induced by the 5-HT2A receptor agonist 2, 5-dimethoxy-4-iodoamphetamine (DOI). The concentration of BDNF in serum was determined by enzyme-linked immuno sorbent assay. RESULTS CI + Aβ increased anxiety, as demonstrated by the increase of time spent in the enclosed arms and dark chambers, and locomotion in the outer zone of the open field (thigmotaxis). CI + Aβ decreased the serum concentration of BDNF. YKS reduced the anxiety-like behaviors, suppressed the DOI-induced wet-dog shakes and increased serum BDNF concentrations. CONCLUSION Our findings suggest that YKS extract improves CI + Aβ-induced anxiety by antagonizing 5-HT2A receptors and increasing BDNF.OBJECTIVE To investigate the efficacy of Tiaobu Feishen formulae (TBFS), including Bufei Jianpi formula (BJF), Bufei Yishen formula (BYF), and Yiqi Zishen formula (YZF), on inflammatory response, protease-anti-protease imbalance and collagen deposition in rats. METHODS In present work, we used an in vitro model of cigarette smoking extract (CSE)- and tumor necrosis factor-α (TNF-α)-induced A549 cells to examine the efficacy of BJF, BYF and YZF on the production of inflammatory cytokines, including TNF-α and interleukin (IL)-8, IL-6, matrix metalloproteinases (MMP)-9, and IL-10 in CSE or TNF-α-induced A549 cells. And their related transcription factors and signaling pathway were also analyzed. RESULTS The results showed that BJF, BYF and YZF could significantly decrease the expression levels of the pro-inflammatory cytokines induced by CSE or TNF-α. Furthermore, BJF, BYF and YZF could suppress CSE- or TNF-α-induced activation of nuclear factor-kappa B (NF-κB) transcription factors and its corresponding pathways. Taken together, these data implied that BJF, BYF and YZF effectively inhibited CSE- or TNF-α-induced inflammatory response in alveolar epithelial cell, which was due to their inhibition effect on NF-κB pathways. CONCLUSION Our findings suggest that the Tiaobu Feishen therapies may protect human alveolar epithelial cells against cigarette smoking and TNF-α-induced inflammation. NF-κB pathway may involve in the actions.OBJECTIVE To evaluate the effects of the Xiaoyaosan (XYS) decoction on the food intake and body weight of rats with chronic immobilization stress (CIS), as well as the concentration of serum leptin and the expression of feeding-related neuropeptides [leptin receptor (Ob-R), proopiomelanocortin (POMC), and α-melanocyte-stimulating hormone (α-MSH)] in the hypothalamic arcuate nucleus (ARC). METHODS We subjected rats to CIS for 3 h a day. The rats were randomly divided into the following groups control group, 7-day stress group, 21-day stress group and XYS-treated group. The rats in the two stress groups were exposed to CIS for 7 or 21 d. The rats in the XYS-treated group were also exposed to CIS for 21 d and were intragastrically administered the XYS decoction before stress. The body weight and food intake of the rats were measured every day. The content of leptin in serum and α-MSH in the ARC were detected by ELISA, and the expression of neuropeptides in the ARC was assayed by immunofluorescence, Western blot and qRT-PCR. RESULTS The food intake and body weight of rats exposed to CIS were lower than those of control rats. The serum leptin, and expression levels of Ob-R, POMC and α-MSH in the ARC were significantly higher than those in the control rats. Treatment with the XYS decoction improved the appetite and the body weight, and down-regulated serum leptin and Ob-R, POMC and α-MSH in hypothalamus ARC. CONCLUSION The leptin-Ob-R-POMC pathway might be the part of the mechanism underlying XYS's improvement of somatic symptoms such as reduction in food intake and body weight related to CIS.OBJECTIVE To observe the effect of Yanggan Jiedu Sanjie (YGJDSJ) formula on human hepatocellular carcinoma Bel-7402 cells. METHODS Bel-7402 cells were treated with YGJDSJ. Cell proliferation was detected by cell counting kit-8 assay. Cell apoptosis was identified by Hoechst 33258 staining and flow cytometric analysis. Cell cycle distribution was quantified by flow cytometric analysis. Caspase activities were measured by commercial kit. Cell senescence was detected by senescence-associated β-galactosidase (SA-β-gal) staining. Protein expression and phosphorylation were identified by Western blot. Protein expression was knocked-down by siRNA. RESULTS YGJDSJ inhibited proliferation of Bel- 7402 cells in a dose- and time-dependent manner. YGJDSJ induced apoptosis and activated caspase- 3, 8, and 9 in Bel-7402 cells. YGJDSJ-induced apoptosis was completely abrogated by a pan caspase inhibitor, Z-VAD-FMK. YGJDSJ also induced cell senescence, up-regulated cyclin-dependent kinase inhibitor 1a (CDKN1a) and CDKN2a expression and down-regulated retinoblastoma protein (RB) phosphorylation in Bel-7402 cells. Specific knockdown of CDKN1a and CDKN2a significantly reduced YGJDSJ-induce cell senescence in Bel-7402 cells. CONCLUSION YGJDSJ inhibited cell proliferation, induced caspase-dependent apoptosis and CDKN1a/CDKN2a-RB signalling mediated cell senescence in Bel-7402 cells. Our findings suggest that YGJDSJ might be potential for hepatocellular carcinoma treatment.OBJECTIVE To find out the combination of the extracts from Paeonia lactiflora Pallas (PL), Reh- mannia Glutinosa var. Purpurea Makino (RG), Perilla Frutescens var. Acuta Kudo (PF) to increase endometrial receptivity. METHODS Herbal medicines were extracted with boiling water and polysaccharides were removed. We examined the effect of PL, RG, and PF (PRP), a most effective herbal formula deduced from constitutive ingredient herbs of Antai Yin which is composed of PRP, on the leukemia inhibitory factor (LIF) expression and endometrial receptivity. RESULTS The combination of the extracts from PRP induced the LIF expression in Ishikawa cells and increased the adhesion between Ishikawa and JAr cells. In addition, PRP-induced attachment of JAr cells onto Ishikawa cells and expression of adhesion molecules, ITGAV, ITGB5, CD44s, and L-selectin, are significantly reduced by knock-down of LIF expression. CONCLUSION Induced by the combination of the PRP extracts, the adhesion between trophoblast and endometrial cells are mediated by expression of LIF and adhesion molecules. Thus, we suggest the combination of the PRP extracts may be a novel therapy for enhancing embryo implantation rate.OBJECTIVE To evaluate current evidence on the efficacy and safety of Cordyceps sinensis (cordyceps) or its fermented products used as an adjunctive treatment in patients undergoing maintenance hemodialysis. METHODS The Cochrane Central Register of Controlled Trials (CENTRAL), EMBASE, MEDLINE, China National Knowledge Infrastructure (CNKI) and Wanfang Database were searched for relevant randomized controlled trials up to March 2016. learn more Two review authors independently selected trials for inclusion, extracted data, assessed the methodological quality and rated the quality of evidence with the Grading of Recommendations, Assessment, Development and Evaluation approach. RESULTS Twelve studies involving 655 participants were included. Evidence of low to moderate-quality showed that cordyceps plus conventional treatment compared to conventional treatment alone significantly improved C-reactive protein [standardized mean difference (SMD) -0.61; 95% confidence intervals (CI) -1.00 to -0.22], high-sensitivity C-reactive protein [weighted mean difference (WMD) -3.44 mg/L; 95% CI -3.89 to -2.99], serum albumin (WMD 3.07 g/L; 95% CI 1.59 to 4.55), malondialdehyde (WMD -1.95 nmol/L; 95% CI -2.24 to -1.66), and hemoglobin (WMD 9.56 g/L; 95% CI 3.65 to 15.47) levels. However, there was no significant improvement for serum creatinine and low-density lipoprotein cholesterol. Overall, most trials either did not monitor adverse events or poorly documented them. CONCLUSION Given the small number of trials included, the unclear methodological quality of the included trials, and the high heterogeneity in pooled analyses, the evidence obtained in this review is insufficient to recommend the use of cordyceps as adjunctive treatment in hemodialysis patients.OBJECTIVE To research the Traditional Chinese Medicine (TCM) practice of Professor Xu Runsan for treatment of chronic pelvic pain (CPP) caused by sequelae of pelvic inflammatory disease (SPID) by data mining. METHODS The medical records of inpatients at China-Japan Friendship Hospital confirmed to have CPP caused by SPID were collected (274 visits in total). The data extracted from the medical records were analyzed by frequency statistics, correlation analyses, cluster analyses, and complex network analyses. RESULTS The most frequently used medicines were warm medicines, bitter medicines, and medicines distributed to the liver meridian. The most common medicinal combinations were Chishao (Radix Paeoniae Rubra) plus Huangqi (Radix Astragali Mongolici) plus Sanqi (Radix Notoginseng) and Ezhu (Rhizoma Curcumae Phaeocaulis); Guizhi (Ramulus Cinnamomi) plus Fuling (Poria) and Chishao (Radix Paeoniae Rubra); and Chaihu (Radix Bupleuri Chinensis) plus Zhishi (Fructus Aurantii Immaturus) and Gancao (Radix Glycyrrhizae).
