Platelet rich plasma (PRP) is widely used in orthopaedics, but is still heavily debated. Therefore, a survey among the German "Working Group for Clinical Tissue Regeneration" of the German Society of Orthopaedics and Traumatology was conducted to achieve a consensus about the current therapeutical potential of PRP.

A first survey (n = 65 experts, all orthopaedic/trauma surgeons) was conducted (n = 13 questions). Following, a second round (n = 40 experts) was conducted with 31 questions to achieve consensus in 5 categories three most common indications, PRP application, future research areas.

Therapeutic PRP application was regarded as useful (89%), possibly even more important in the future (90%). Most common indications were tendon pathologies (77%), osteoarthritis (OA) (68%), muscle injuries (57%) and cartilage damage (51%). Consensus was reached in 16/31 statements. The application of PRP for early knee OA (Kellgren-Lawrence grade II) was regarded as potentially useful, as well as for acute and chronic tendinopathies. For chronic lesions (cartilage, tendons), multiple injections (2-4) were seen preferable to singular injections. However, no sufficient data exists on the time interval between the injections. Standardization of PRP preparation, application, frequency, as well as determining the range of indication is strongly recommended.

There is a need of further standardization of the PRP preparation methods, indication and application protocols for knee OA and other indications, which must be further evaluated in basic science studies and randomized controlled clinical trials.

The challenging target in the workup of thyroid nodule(s) is to exclude or diagnose thyroid cancer efficiently prior to surgical intervention. The present work studied a panel of eight serum biomarkers to differentiate benign from malignant thyroid nodules, aiming at reducing unnecessary thyroidectomy performed for inconclusive preoperative fine needle aspiration cytology. DNA Repair inhibitor Serum interleukin-5 (IL-5), interleukin-8 (IL-8), hepatocyte growth factor (HGF), epidermal growth factor (EGF), angiopietin (Ang1), nonokine induced by interferon gamma (MIG), galectin (Gal-3), and vitamin D-binding protein (VDRP) were quantified by multiplex bead assay using Luminex xMAP technology. The study was conducted on 60 subjects of three groups (20 each; healthy controls, benign thyroid nodule, and malignant thyroid nodule).

Significant increase of the following biomarkers in the malignant group compared to the benign group was found; IL-8 29.7 vs 8.75 pg/ml, p < 0.001, EGF 128.7 vs 6.72 pg/ml, p < 0.001, HGF 173.2 vs 112.2 pg/ml, p = 0.012, MIG 776.7 vs 438 pg/ml, p = 0.023, and Ang-1 95016 vs 33327.5 pg/ml, p = 0.014. No significant differences were detected for IL-5, Gal-3, and VDBP. Serum IL-8 and EGF showed the highest diagnostic performance individually with area under the curve (AUC) 0.849 and 0.848, respectively. The combined biomarker panels of IL-8 and EGF and IL-8, EGF, and MIG have reached a sensitivity and specificity of 95% and 65%, respectively, with a negative predictive value of 92.9%.

Hydrochlorothiazide is found to form cocrystals with picolinamide (H1), tetramethylpyrazine (H2) and piperazine (H3). It was characterized using IR spectra, and quantum mechanical calculations for geometry and other properties. Frontier orbital energies are used to predict the energy properties and model the possible charge transfer between the constituents of the cocrystal. The frontier molecular orbital analysis indicates chemical reactivity and bioactivity of the cocrystals. The MEP surface reveals the various reactive surfaces in the cocrystal system, which is very important in deciding various biological activities. The UV-Vis spectra show the possible electronic transitions of the molecules. Simulated electronic spectra using TDDFT method with CAM-B3LYP functional were used to investigate the suitability of the cocrystals to be used in DSSC. Moreover, the molecular docking analysis proves that the cocrystals can act as potential inhibitors and paves the way for developing effective drugs.Two novel ecological and low-cost adsorbents were prepared from seed residues of the tree species Anadenanthera macrocarpa and Cedrela fissilis for the removal of methylene blue dye in water. The materials were comminuted and characterized by different techniques. The particles of samples have a rough surface with cavities. The optimum dosage and pH for both materials were 1 g L-1 and pH 8. The pseudo-second-order model was the most suitable for describing the adsorption kinetics for both systems. The Anadenanthera macrocarpa presented a maximum experimental capacity of 228 mg g-1, while the Cedrela fissilis, a similar capacity of 230 mg g-1 at 328 K. The Tóth model was proper for describing the equilibrium curves for both systems. The thermodynamic indicators show that the adsorption process is spontaneous and endothermic for both materials. The application of materials for the simulated effluent treatment showed 74 and 78% of color removal using Anadenanthera macrocarpa and Cedrela fissilis samples, respectively. Overall, seed residues of Anadenanthera macrocarpa and Cedrela fissilis could be potentially applied for adsorptive removal of colored contaminants in wastewater.Better understanding of soil water storage and photosynthetic regulation of maize production will be useful to develop a water-saving strategy in rain-fed conditions. Therefore, maize crop was grown under the different cultivation practices for analyzed light and CO2-response curves under various mulching strategies during 2017-2018 years. Six different treatments were used such as the following PP, ridges and furrows zone covered with plastic film mulching; PS, ridges covered with plastic film and furrows zone with stalk mulching; PN, ridges covered with plastic film and furrows zone without mulching; TP, conventional flat planting with plastic film mulching; TS, conventional flat planting with stalk mulching; and TN, conventional flat planting without mulching. The PP treatment had considerable effects on rainwater collection, improved SWS, and maize productivity than that of TP treatment. Significantly increase of SWS was observed under the PP treatment as a result photosynthetic capacity (An) improved under light and CO2-response curves, apparent quantum efficiency (α), respiration rate, total chlorophyll ab content, and 13C-photosynthates distribution in different organs. Under the PP and TP treatments, the maize might keep a great photosynthetic capacity at the post-flowering stage through improving An, LAI, soluble protein, Rubisco contents, and grain yield. The CO2 and light-response curves were significantly enhanced at the PP treatment due to higher 13C carbon isotope (Δ‰) and Ci/Ca as a result lower bundle sheath to leakiness of CO2 (ɸ) compared with the rest of all treatments. The results suggested that PP cultivation practice was the best water-saving strategy because it reduced bundle sheath leakiness to CO2 (ɸ); as a result there's a significant improvement in soil water storage, LAI, 13C-photosynthates distribution, photosynthetic capacity parameters, and maize production.One of the consequences of phytosanitary treatments applied to crops is the generation of a great volume of agro-wastewater having pesticide residues. These pollutants can be considered a serious threat to the environment and human health due to their capacity to affect distant areas remaining for a long time after their application. We have assessed the degradation of five pesticides in agro-waste water produced in two farms by the cleaning pesticide containers and phytosanitary treatment equipment used in the farms. For this purpose, a pilot facility was installed in both farms and advanced oxidation treatments were conducted using natural sunlight by means of Na2S2O8 and heterogeneous photocatalysis (TiO2/Na2S2O8). The remaining percentages obtained at the end of the experiments ranged from 5 to 90.1% for chlorantraniliprole, 5 to 82.3% for difenoconazole, 0.02 to 19.1% for metalaxyl, 1.4 to 74.4% for myclobutanil, and 0.3 to 61% for triadimenol. We observed a correlation between the higher remaining percentages and the total initial concentration of pollutant because of other commercial formulations applied in the farms. The results showed that this equipment could be used to eliminate or reduce the presence of pesticide residues in agro-waste water using an innovative facility installed in the farms and a renewable and economical source of energy (sunlight).The respiratory tract is the route of entry for accidentally inhaled AgNPs, which can reach the lungs and redistribute to other main organs through systemic circulation. In the present work, we aimed to evaluate silver biodistribution and biological effects after 1 or 2 intratracheal instillations (IT) of two differently sized PVP-coated AgNPs (5 and 50 nm-3 mg/kg) and ionic silver (AgNO3-1 mg/kg bw) in mice. Furthermore, nuclear magnetic resonance (NMR) metabolomics was applied to unveil pulmonary metabolic variations. Animals exposed to 5 nm AgNP (AgNP5) showed higher levels of ionic silver in organs, especially in the lung, spleen, kidney and liver, while animals exposed to 50 nm AgNP (AgNP50) showed higher levels of silver in the blood. Animals exposed to AgNP50 excreted higher amounts of silver than those exposed to AgNP5, which is consistent with higher tissue accumulation of silver in animals exposed to the latter. Lung metabolic profiling revealed several Ag-induced alterations in metabolites involved in different pathways, such as glycolysis and tricarboxylic acid (TCA) cycle, amino acid and phospholipid metabolism, and antioxidant defense. Notably, most of the metabolic changes observed after 1 IT were absent in animals subjected to 2 IT of AgNO3, or reversed for AgNPs, suggesting adaptation mechanisms to cope with the initial insult and recover homeostasis. Graphical abstract.γ-Glutamyl compounds have unveiled their importance as active substances or precursors of pharmaceuticals. In this research, an approach for enzymatic synthesis of γ-glutamyl compounds was developed using γ-glutamylmethylamide synthetase (GMAS) from Methylovorus mays and polyphosphate kinase (PPK) from Corynebacterium glutamicum. GMAS and PPK were co-recombined in pETDuet-1 plasmid and co-expressed in E. coli BL21 (DE3), and the enzymatic properties of GMAS and PPK were investigated, respectively. Under the catalysis of the co-expression system, L-theanine was synthesized with 89.8% conversion when the substrate molar ratio of sodium glutamate and ethylamine (11.4) and only 2 mM ATP were used. A total of 14 γ-glutamyl compounds were synthesized by this one-pot method and purified by cation exchange resin and isoelectric point crystallization with a yield range from 22.3 to 72.7%. This study provided an efficient approach for the synthesis of γ-glutamyl compounds by GMAS and PPK co-expression system.
