This research work primarily focussed on the production of biochar from sugarcane bagasse through HTC followed by NaOH activation at inert atmosphere for removing SMX from water. The biochar was characterized for structural morphology and presence of functional groups. XRD and FTIR analysis confirmed that presence of aromatized graphitic structure accumulated with oxygenated functional groups are responsible for the elimination of SMX. Coelenterazine SEM analysis portrayed the sphere-shaped structure of biochar with hydrophobic groups interior and hydrophilic groups exterior. BET isotherm revealed the active surface area equal to 1099 m2/g with high coverage of mesopores structure. Pzpc of adsorbent is evaluated to 6.5 stating that effective removal of SMX depends on ionization effects induced due to reaction medium. Kinetics study revealed the sorption of SMX followed chemical interaction pertaining to Elovich model. Isotherm studies revealed that Freundlich model fitted well stating heterogeneous mode of interaction. Immobilization of SMX on surface of ABC is due to charge assisted hydrogen bonding and π-π interaction with graphitized carbon, showing maximum sorption capacity of 400 mg/g through spontaneous reaction. The results suggested that HTC derived biochar had great adsorption affinity with respect to pH towards SMX and could be employed as an effective sorbent in cleaning water contaminants.In this work, the effectiveness of a biodegradable and natural surfactant synthesized through a novel method has been studied through the ion flotation process to treat waters containing Per/Poly Fluoroalkyl Substances (PFAS) and heavy-metal ions. This cysteine-based surfactant, which is environmentally acceptable, showed considerable solubility and foaming ability over a wide range of pH. It also could remove 97-99(%) of 5 mg/L of cadmium, chromium, copper, nickel, zinc, and manganese ions in a single batch physicochemical process. Moreover, for the first time, a foam fractionation method in association with using this cysteine-based surfactant was applied for perfluorooctanoic acid (PFOA) removal from water. This surfactant was used as a co-surfactant and could readily remove 72% of PFOA (40 mg/L) from water. The characterization of the surfactant was undertaken using 1H NMR, FT-IR, elemental analysis, melting point, and determination of its critical micelle concentration (CMC). This environmentally friendly surfactant has high potential applications in green chemistry especially in the treatment of waters contaminated with PFAS/heavy-metal ions.The contamination of the environment with anticancer drugs, which show recalcitrance to conventional wastewater treatment, has become a significant ecological threat. Fungi represent a promising non-conventional biological alternative for water conditioning. The aim of this work was to evaluate the efficacy of five white-rot fungi (Fomes fomentarius (CB13), Hypholoma fasciculare (CB15), Phyllotopsis nidulans (CB14), Pleurotus ostreatus (BWPH) and Trametes versicolor (CB8)) in the removal of bleomycin and vincristine. The removal capacity was measured at 0, 4, 9, and 14 days of incubation using SPE-UPLC-MS. The enzymatic profiles of laccase, manganese, and lignin peroxidases and wide range of eco- and cytotoxicity, assays of the post-process samples were also conducted. We observed >94% vincristine elimination by F. fomentarius, H. fasciculare and T. versicolor after only 4 days. Bleomycin removal occurred after a minimum of 9 days and only when the drug was incubated with T. versicolor (36%) and H. fasciculare (25%). The removal of both cytostatics was associated with laccase production, and the loss of eco- and cytotoxicity, especially in regard to viability of Lemna minor and Daphnia magna, as well as fibroblasts morphology.The mycotoxin ochratoxin A (OTA) is a widespread contaminant in human and animal food products. Previous studies in rats revealed that melatonin (Mel) exhibits a preventive effect against OTA-induced oxidative stress in liver. However, it remains unknown whether gut microbiota respond to Mel and, if so, whether it can prevent OTA-induced inflammation and mitophagy in the liver. In the present study, mice received an oral gavage of Mel and OTA for 3 weeks before harvesting colonic digesta and liver tissue for analyses. In another study, the role of intestinal microbiota on the effects of Mel on OTA-induced liver inflammation and mitophagy was assessed through clearance of intestinal microbiota with antibiotics followed by gut microbiota transplantation (GMT). Oral Mel supplementation ameliorated mitophagy in the liver and reversed gut microbiota dysbiosis. Intriguingly, in antibiotic-treated mice, Mel and OTA failed to induce mitophagy in the liver. Using the GMT approach in which mice were colonised with intestinal microbiota from control-, OTA-, or Mel + OTA-treated mice led us to elucidated the involvement of intestinal microbiota in liver inflammation and mitophagy induced by OTA. The findings suggested that intestinal microbiota play some role in the Mel-induced amelioration of liver inflammation and mitophagy induced by OTA.Gold-based plasmonic nanocrystals have been extensively developed for noninvasive photothermal therapy. In this study, gold nanorods (AuNRs) with (200) plane and gold nanobipyramids (AuNBPs) with (111) plane were utilized as photothermal agents for noninvasive photothermal therapy. With longitudinal surface plasma bands at ~808 nm, both of AuNRs and AuNBPs revealed photothermal capability and reversibility of laser response under 808-nm near-infrared (NIR) laser irradiation. Moreover, AuNBPs with (111) plane exhibited higher photothermal performance than that of AuNRs with (200) plane under NIR laser irradiation. Density function theory (DFT) simulations revealed that water adsorption energy followed the order Au(111) less then Au(100), indicating that the water was easily desorbed on the Au(111) surface for photothermal heating. For the photothermal therapy against Escherichia coli (E. coli), AuNBPs also exhibited higher efficiency compared to that of AuNRs under NIR laser irradiation. Combination of experimental photothermal therapy and DFT simulations demonstrated that AuNBPs with (111) plane were better photothermal agents than that of AuNRs with (100) plane.In-tube solid-phase microextraction (IT-SPME) was developed about 24 years ago as an effective sample preparation technique using an open tubular capillary column as an extraction device. link2 IT-SPME is useful for micro-concentration, automated sample cleanup, and rapid online analysis, and can be used to determine the analytes in complex matrices simple sample processing methods such as direct sample injection or filtration. IT-SPME is usually performed in combination with high-performance liquid chromatography using an online column switching technology, in which the entire process from sample preparation to separation to data analysis is automated using the autosampler. Furthermore, IT-SPME minimizes the use of harmful organic solvents and is simple and labor-saving, making it a sustainable and environmentally friendly green analytical technique. Various operating systems and new sorbent materials have been developed to improve its extraction efficiency by, for example, enhancing its sorption capacity and selectivity. In addition, IT-SPME methods have been widely applied in environmental analysis, food analysis and bioanalysis. link3 This review describes the present state of IT-SPME technology and summarizes its current trends and future perspectives, including method development and strategies to improve extraction efficiency.Canine babesiosis, a tick-borne haemoprotozoan disease of dogs, is of significance globally due to its rapid spread. A precise confirmatory diagnosis is required to curtail the rapid spread of infection. Our study described the evaluation of recombinant BgSA3 protein based indirect ELISA for sero-diagnosis and sero-surveillance of Babesia gibsoni infection in dogs. A partial BgSA3 gene segment (1921 bp) of B. gibsoni, encoding for recombinant truncated BgSA3 (75 kDa) protein devoid of predicted signal peptide (23 aa) at N-terminus and transmembrane region (20 aa) at C-terminus, was expressed in E. coli using a pET28a(+) vector. The rBgSA3 protein purified under native conditions using Ni-NTA superflow cartridge was confirmed by SDS-PAGE and Western blotting using sera from dogs infected/uninfected with B. gibsoni, and erythrocyte lysate/ plasma from infected/uninfected dogs. The rBgSA3 protein was specific only to B. gibsoni antibodies but did not react with uninfected sera. Further, rBgSA3 protein was evaluated for sero-diagnosis/sero-surveillance using Indirect-ELISA format. There was no cross reactivity to B. vogeli, E. canis, H. canis and D. repens infected dogs serum samples. The diagnostic sensitivity and specificity of rBgSA3 based I-ELISA was found to be 86.4 and 93.1 % respectively, in comparison with cytb based PCR assay. Additionally, rBgSA3-ELISA evaluated using survey serum samples (n = 287), detected 11.85 % samples as positive. In conclusion, B. gibsoni infection, an emerging disease is prevalent in the present study area and the standardized rBgSA3 protein based indirect-ELISA was found to be a specific and sensitive test for large scale sero-diagnosis and sero-surveillance of B. gibsoni infection in dogs.Studies conducted to support registration or approval of veterinary anthelmintics generally follow study design recommendations provided by the VICH (International Cooperation on Harmonisation of Technical Requirements for Registration of Veterinary Medicinal Products), "Efficacy of Anthelmintics General Requirements" (VICH GL7). For dose confirmation studies, VICH GL7 provides recommendations for determining that the control animals had an adequate infection "to permit the appropriate standards of efficacy to be met with acceptable statistical and biological certitude/confidence." In the simulation studies described in this report, we investigated the performance of one method, the statistical criterion given in Section 4.5 of VICH GL7, for evaluating the adequacy of infection in anthelmintic studies, in combination with the conventional criterion of a minimum of six adequately infected animals. We conducted numerical simulations, based on parasite data from previously conducted dose confirmation studies in dogs and cattle, to investigate how the statistical criterion impacts adequacy of infection determinations when used with the conventional criterion at various sample sizes. Simulation studies in common nematode species in both dogs and cattle indicated that under certain circumstances the statistical criterion can guard against overinterpreting the evaluation of adequacy of infection as sample size is increased. However, the statistical criterion may be overly restrictive for samples with adequate infection but containing multiple zero parasite counts and adding it to the conventional criterion does not provide any additional benefit when the sample contains no zero counts. It is important for investigators designing efficacy studies to understand the potential impact this criterion may have when establishing adequacy of infection criteria in study protocols.
