Cholesterol-dependent cytolysins (CDCs) are key virulence factors involved in many lethal bacterial infections, including pneumonia, necrotizing soft tissue infections, bacterial meningitis, and miscarriage. Host responses to these diseases involve myeloid cells, especially macrophages. Macrophages use several systems to detect and respond to cholesterol-dependent cytolysins, including membrane repair, mitogen-activated protein (MAP) kinase signaling, phagocytosis, cytokine production, and activation of the adaptive immune system. However, CDCs also promote immune evasion by silencing and/or destroying myeloid cells. While there are many common themes between the various CDCs, each CDC also possesses specific features to optimally benefit the pathogen producing it. This review highlights host responses to CDC pathogenesis with a focus on macrophages. Due to their robust plasticity, macrophages play key roles in the outcome of bacterial infections. Understanding the unique features and differences within the common theme of CDCs bolsters new tools for research and therapy.The human mediator subunit MED25 acts as a coactivator that binds the transcriptional activation domains (TADs) present in various cellular and viral gene-specific transcription factors. Previous studies, including on NMR measurements and site-directed mutagenesis, have only yielded low-resolution models that are difficult to refine further by experimental means. Here, we apply computational molecular dynamics simulations to study the interactions of two different TADs from the human transcription factor ETV5 (ERM) and herpes virus VP16-H1 with MED25. Like other well-studied coactivator-TAD complexes, the interactions of these intrinsically disordered domains with the coactivator surface are temporary and highly dynamic ('fuzzy'). Due to the fact that the MED25 TAD-binding region is organized as an elongated cleft, we specifically asked whether these TADs are capable of binding in either orientation and how this could be achieved structurally and energetically. The binding of both the ETV5 and VP16-TADs in either orientation appears to be possible but occurs in a conformationally distinct manner and utilizes different sets of hydrophobic residues present in the TADs to drive the interactions. We propose that MED25 and at least a subset of human TADs specifically evolved a redundant set of molecular interaction patterns to allow binding to particular coactivators without major prior spatial constraints.Both climate change and migration present key concerns for global health progress. In doing so, we hope to provide insights that help in the more homogenous applications of conceptual frameworks for this area and more generally.Omega-3 long-chain polyunsaturated fatty acids (LCPUFA) have been shown to inhibit lipogenesis and adipogenesis in adult rats. Their possible early life effects on offspring fat deposition, however, remain to be established. To investigate this, female Wistar rats (n = 6-9 per group) were fed either a 91 ratio of linoleic acid (LA) to alpha-linolenic acid (ALA) or a lower 11.5 ratio during pregnancy and lactation. Each ratio was fed at two total fat levels (18% vs. 36% fat w/w) and offspring were weaned onto standard laboratory chow. Offspring exposed to a 36% fat diet, irrespective of maternal dietary LAALA ratio, were lighter (male, 27 g lighter; female 19 g lighter; p less then 0.0001) than those exposed to an 18% fat diet between 3 and 8 weeks of age. Offspring exposed to a low LA (18% fat) diet had higher proportions of circulating omega-3 LCPUFA and increased gonadal fat mass at 4 weeks of age (p less then 0.05). Reduced Srebf1 mRNA expression of hepatic (p less then 0.01), gonadal fat (p less then 0.05) and retroperitoneal fat (p less then 0.05) tissue was observed at 4 weeks of age in male and female offspring exposed to a 36% fat diet, and hepatic Srebf1 mRNA was also reduced in male offspring at 8 weeks of age (p less then 0.05). Thus, while offspring fat deposition appeared to be sensitive to both maternal dietary LAALA ratio and total fat content, offspring growth and lipogenic capacity of tissues appeared to be more sensitive to maternal dietary fat content.The coronavirus disease 2019 (COVID-19) pandemic continues to pose profound challenges to society. Its spread has been mitigated through strategies including social distancing; however, this may result in the adoption of a sedentary lifestyle. This study aimed to investigate (1) physical activity (PA) levels, sedentary behavior (SB), and sleep in young adults during the COVID-19 epidemic, and (2) the change in these behaviors before and during the pandemic. A total of 631 young adults (38.8% males) aged between 18 and 35 participated in the cross-sectional study and completed a one-off online survey relating to general information, PA, SB, and sleep. For the longitudinal study, PA, SB, and sleep data, obtained from 70 participants before and during the COVID-19 pandemic, were analyzed. Participants engaged in low PA, high SB, and long sleep duration during the COVID-19 pandemic. Moreover, a significant decline in PA while an increase in time spent in both SB and sleep was observed during the COVID-19 outbreak.
