Functional and structural data were recorded from 26 participants performing motor, auditory, and visual tasks in different conditions with the 20- and 64-channel Siemens head/neck coil and the prescan normalize filter turned ON or OFF. Data were analyzed with the MRIQC tool to evaluate data quality differences. The functional data were statistically evaluated by comparison of the β estimates and the time-course signal-to-noise ratio (tSNR) in four regions of interest, i.e., the auditory, visual, and motor cortices and the thalamus. The MRIQC tool indicated a better data quality for both functional and structural data with the prescan normalize filter, with an advantage for the 20-channel head coil in functional data and an advantage for the 64-channel head coil in structural measurements. Nevertheless, recommendations for the functional data regarding choice of head coils and prescan normalize filter depend on the brain regions of interest. Higher β estimates and tSNR values occurred in the auditory cortex and thalamus with the prescan normalize filter, whereas the contrary was true for the visual and motor cortices. Due to higher β estimates in the visual cortex in the 64-channel head coil, this head coil is recommended for studies investigating the visual cortex. For most of the research questions, the 20-channel head coil is better suited for functional experiments, with the prescan normalize filter, especially when investigating deep brain areas. For anatomical studies, the 64-channel head coil seemed to be the better choice.Introduction Autism Spectrum Disorder (ASD) is a heterogeneous neurodevelopmental disorder characterized by a complex pathogenesis, by impairment social communication and interaction, and may also manifest repetitive patterns of behavior. Many studies have recognized an alteration of the immune response as a major etiological component in ASDs. Despite this, it is still unclear the variation of the function of the immune response. Aim Our aim is to investigate the levels of immunological markers in peripheral blood of children with ASD such as regulatory B and T cells, memory B and natural killer (NK) cells. Materials and Methods We assessed various subsets of immune cells in peripheral blood (regulatory B and T cells, B-cell memory and natural killer cells) by multi-parametric flow cytometric analysis in 26 ASD children compared to 16 healthy controls (HCs) who matched age and gender. Results No significant difference was observed between B-cell memory and NK cells in ASDs and HCs. Instead, regulatory B cells and T cells were decreased (p less then 0.05) in ASD subjects when compared to HCs. Discussion Regulatory B and T cells have a strategic role in maintaining the immune homeostasis. Their functions have been associated with the development of multiple pathologies especially in autoimmune diseases. According to our study, the immunological imbalance of regulatory B and T cells may play a pivotal role in the evolution of the disease, as immune deficiencies could be related to the severity of the ongoing disorder.In social animals, identifying sounds is critical for communication. In humans, the acoustic parameters involved in speech recognition, such as the formant frequencies derived from the resonance of the supralaryngeal vocal tract, have been well documented. However, how formants contribute to recognizing learned sounds in non-human primates remains unclear. To determine this, we trained two rhesus monkeys to discriminate target and non-target sounds presented in sequences of 1-3 sounds. After training, we performed three experiments (1) We tested the monkeys' accuracy and reaction times during the discrimination of various acoustic categories; (2) their ability to discriminate morphing sounds; and (3) their ability to identify sounds consisting of formant 1 (F1), formant 2 (F2), or F1 and F2 (F1F2) pass filters. Our results indicate that macaques can learn diverse sounds and discriminate from morphs and formants F1 and F2, suggesting that information from few acoustic parameters suffice for recognizing complex sounds. We anticipate that future neurophysiological experiments in this paradigm may help elucidate how formants contribute to the recognition of sounds.Background Movement fluctuations are the main complication of Parkinson's disease (PD) patients receiving long-term levodopa (L-dopa) treatment. We compared and ranked the efficacy and safety of dopamine agonists (DAs) with regard to motor fluctuations by using a Bayesian network meta-analysis (NMA) to quantify information from randomized controlled trials (RCTs). Methods and Findings We carried out a systematic review and meta-analysis, and only RCTs comparing DAs for advanced PD were included. Electronic databases (PubMed, Embase, and Cochrane Library) were systematically searched for relevant studies published until January 2021. Two reviewers independently extracted individual study data and evaluated studies for risk of bias using the Cochrane Risk of Bias tool. Network meta-analyses using a Bayesian framework were used to calculate the related parameters. The pre-specified primary and secondary outcomes were efficacy ("ON" time without troublesome dyskinesia, "OFF" time, "ON" time, "UPDRS-III," and "UPDimproved the UPDRS III scores, and ultimately ameliorated the UPDRS II scores, thereby maximizing its benefit. This NMA of pramipexole, ropinirole, and rotigotine represents an effective treatment option and has an acceptable safety profile in patients with advanced PD.Multiple sclerosis (MS) is a complex disease of the central nervous system (CNS), characterized by inflammation, demyelination, neuro-axonal loss, and gliosis. Inflammatory demyelinating lesions are a hallmark of the disease. Spontaneous remyelination, however, is often incomplete and strategies that promote remyelination are needed. As a result, accurate and sensitive in vivo measures of remyelination are necessary. The visual pathway provides a unique opportunity for in vivo assessment of myelin damage and repair in the MS-affected brain since it is highly susceptible to damage in MS and is a very frequent site of MS lesions. The visually evoked potential (VEP), an event-related potential generated by the striate cortex in response to visual stimulation, is uniquely placed to serve as a biomarker of the myelination along the visual pathway. The multifocal VEP (mfVEP) represents a most recent addition to the array of VEP stimulations. This article provides a current view on the role of mfVEP as a biomarker of demyelination, spontaneous remyelination, and myelin repair in MS.Objective Executive function refers to the conscious control of thinking and behavior in psychological process. Executive dysfunction widely exists in a variety of neuropsychiatric diseases, and is closely related to the decline of daily living ability and function. This study intends to explore the effect of low-frequency repetitive transcranial magnetic stimulation (rTMS) on executive function and its neural mechanism by using event-related potential (ERP), so as to provide basis for further study on the relationship between cerebral cortex and executive function. Methods Task switching paradigm was used to study the cognitive flexibility in executive function. Thirty-one healthy subjects were randomly assigned to receive rTMS stimulations (1 Hz rTMS or sham rTMS) to the left dorsolateral prefrontal cortex (DLPFC) twice. The switching task and the electroencephalography EEG recordings were performed before (pre-rTMS/pre-sham rTMS) and immediately after the end of the rTMS application (post-rTMS/post-sham rTMS). Results The analysis of RTs showed that the main effects of switching and time were statistically significant. Further analysis revealed that the RT of rTMS stimulation was longer than sham rTMS at post-stimulation. ERP analysis showed that there was a significant switching effect in frontal and central scalp location, and the P2 amplitude in switch trials was greater than that in non-switch trials. At post-stimulation, the N2 amplitude of rTMS is more negative than that of sham rTMS at non-switch trials, whereas no such difference was found at switch trials. The P3 amplitude and LPC amplitude are significantly reduced by rTMS at post-stimulation. Conclusion Low-frequency rTMS of the left DLPFC can cause decline of cognitive flexibility in executive function, resulting in the change of N2 amplitude and the decrease of P3 and LPC components during task switching, which is of positive significance for the evaluation and treatment of executive function.Mild traumatic brain injury (TBI) involves widespread axonal injury and activation of microglia, which initiates secondary processes that worsen the TBI outcome. The upregulation of cannabinoid type-2 receptors (CB2) when microglia become activated allows CB2-binding drugs to selectively target microglia. CB2 inverse agonists modulate activated microglia by shifting them away from the harmful pro-inflammatory M1 state toward the helpful reparative M2 state and thus can stem secondary injury cascades. We previously found that treatment with the CB2 inverse agonist SMM-189 after mild TBI in mice produced by focal cranial blast rescues visual deficits and the optic nerve axon loss that would otherwise result. We have further shown that raloxifene, which is Food and Drug Administration (FDA)-approved as an estrogen receptor modulator to treat osteoporosis, but also possesses CB2 inverse agonism, yields similar benefit in this TBI model through its modulation of microglia. As many different traumatic events produce TBI in humans, it is widely acknowledged that diverse animal models must be used in evaluating possible therapies.
