Taking on metastatic triple-negative breast cancer using sacituzumab govitecan.
OBJECTIVE Dihydroartemisinin (DHA), a predominant phytoconstituent in Artemisia annua L. (a plant widely used as a traditional medicine in China), inhibits lung tumorigenesis and metastasis. However, its anticancer effect against hepatocellular carcinoma has not yet been investigated. In this study, the anti-tumor potential of DHA was evaluated in vitro against the hepatocellular carcinoma HCCLM6 cell line. RESULTS DHA (1-100 μM) treatment suppressed the cell proliferation in dose-dependently. In addition, expression of all genes, involved in cellular proliferation (e.g. E2F1, BCL2, PCNA, MKI67 and CCNE2) and cellular motility (e.g. DOCK1, ITGA2, VCL, MMP2, FN1), was significantly downregulated by DHA (50 and 100 μM). Global gene expression profile identified 1731 differentially expressed genes (DEGs); among them, 211 were up-regulated and 1520 were down-regulated. Besides, the extracellular matrix (ECM)-receptor interaction, focal adhesion, regulation of actin cytoskeleton and TNF pathways were enriched by DEGs. Based on the KEGG signal pathway enrichment, the FN1 and integrin-β1 could be a potential target for DHA for inhibiting proliferation. The expression of FN1 and integrin-β1 was further analyzed by the qPCR, immunohistochemistry and Western blot assay in vitro and in vivo. The results indicated that DHA decreased the FN1 and integrin-β1 protein levels and interfered with PI3K-AKT signal transduction pathway. CONCLUSIONS Our findings revealed that DHA could inhibit proliferation and migration of human hepatocellular carcinoma cells targeting FN1 and ITGB1 via the PI3K-AKT pathway. Therefore, DHA might be a novel drug with a potential effect against liver tumorigenesis and metastasis.BACKGROUND The undifferentiated embryonic sarcoma of the liver (UESL) is a rare, aggressive tumor mainly affecting children. Since UESL has no specific clinical symptoms or imaging characteristics, many cases of UESL are diagnosed late. The paraneoplastic leukemoid reaction (PLR) is a very rare concomitant of oncological patients associated with poor prognosis. This report describes the clinical course of a patient combining these two rare entities and describes the diagnostic challenges and dynamics of paraneoplastic syndrome. CASE PRESENTATION We report a case of UESL in a 46-year-old male who became clinically conspicuous due to pronounced B symptoms. CT and MRI showed a suspicious unifocal liver lesion. As the histological analysis of a tissue sample did not reveal a clear result, an 18F-FDG-PET-CT examination was performed. In addition to a high glucose metabolism of the liver lesion, an increased glucose metabolism in the entire bone marrow was observed. This finding was considered as pronounced paraneoplasia and laparotomy with liver segment resection followed. Immediately after resection of the tumor the paraneoplastic symptoms completely declined and the patient showed no signs of recurrence in the 1-year follow-up. CONCLUSIONS Although UESL is rare and predominantly affects children, this diagnosis should always be considered for unclear unifocal cystic liver lesions, regardless of the patient's age, as appropriate treatment has a good prognosis.Land management and biodiversity protection are highly dependent on ecosystem classification. To identify the ecosystems, often ecologically homogenous areas are distinguished based on physical and biological features at various scales. These areas can also be considered as biodiversity surrogates for protection policies and planning. At a scale of 11,000,000, we obtained a total of 119 homogenous ecological units, and based on the dominant vegetation types, we classified them into 21 terrestrial ecosystems at the national level. It is important to detect stress early and unobtrusively. In this context, stress detection can be considered as a classification problem. In this study, it was investigated the effects of stress by using accelerometer and gyroscope sensor data of the writing behavior on a smartphone touchscreen panel. For this purpose, smartphone data including two states (stress and calm) were collected from 46 participants. The obtained sensor signals were divided into 5, 10 and 15 s interval windows to create three different data sets and 112 different features were defined from the raw data. To obtain more effective feature subsets, these features were ranked by using Gain Ratio feature selection algorithm. Afterwards, writing behaviors were classified by C4.5 Decision Trees, Bayesian Networks and k-Nearest Neighbor methods. As a result of the experiments, 74.26%, 67.86%, and 87.56% accuracy classification results were obtained respectively.This study aimed to investigate whether use of a computed tomography (CT)-based navigation system reduce the risk of dislocation after total hip arthroplasty (THA) in patients with osteonecrosis of the femoral head (ONFH). A total of 271 hips from 192 consecutive patients that underwent primary THA for ONFH were included. There were 110 hips in non-navigation group, and 161 hips in navigation group. After applying exclusion criteria, 209 hips from 149 patients were selected for analysis. Clinical outcomes and complication rates were evaluated, and implant alignments were also calculated. To identify whether the navigation system was useful to prevent dislocation, the inverse probability of treatment-weighted Cox regression analysis using a propensity score in relationship to sex, age at surgery, body mass index, and femoral head size was performed. No significant difference was observed in clinical scores between both groups.
