M-cadherin is a skeletal muscle-specific transmembrane protein mediating the cell-cell adhesion of myoblasts during myogenesis. It is expressed in the proliferating satellite cells and highly induced by myogenic regulatory factors (MRFs) during terminal myogenic differentiation. Several conserved cis-elements, including 5 E-boxes, 2 GC boxes, and 1 conserved downstream element (CDE) were identified in the M-cadherin proximal promoter. We found that E-box-3 and -4 close to the transcription initiation site (TIS) mediated most of its transactivation by MyoD, the strongest myogenic MRF. Including of any one of the other E-boxes restored the full activation by MyoD, suggesting an essential collaboration between E-boxes. Stronger activation of M-cadherin promoter than that of muscle creatine kinase (MCK) by MyoD was observed regardless of culture conditions and the presence of E47. Furthermore, MyoD/E47 heterodimer and MyoD ∼ E47 fusion protein achieved similar levels of activation in differentiation medium (DM), suggesting high affinity of MyoD/E47 to E-boxes 3/4 under DM. We also found that GC boxes and CDE positively affected MyoD mediated activation. The CDE element was predicted to be the target of the chromatin-modifying factor Meis1/Pbx1 heterodimer. Knockdown of Pbx1 significantly reduced the expression level of M-cadherin, but increased that of N-cadherin. Using ChIP assay, we further found significant reduction in MyoD recruitment to M-cadherin promoter when CDE was deleted. Taken together, these observations suggest that the chromatin-modifying function of Pbx1/Meis1 is critical to M-cadherin promoter activation before MyoD is recruited to E-boxes to trigger transcription.BK virus nephropathy in kidney transplantation is widely recognized as an important cause of graft dysfunction and loss. In the case of transplants of organs other than kidney, BK virus nephropathy in native kidneys has been recognized as a cause of chronic kidney disease, which is related with immunosuppression; however, the diagnosis is usually late because the renal dysfunction is attributed to other causes, such as toxicity by anticalcineurinic drugs, interstitial nephritis due to medications, hemodynamic changes, diabetes, hypertension, etc. We report a case of BK virus nephropathy in a patient who underwent heart transplantation due to peripartum cardiomyopathy. The kidney biopsy reported active chronic tubulointerstitial nephritis associated with late stage polyomavirus nephritis and the blood viral load for BK virus was positive (logarithm 4.5). The immunosuppressive treatment was reduced, and after two years of follow-up, the patient had stable renal function with a serum creatinine of 2.5 mg/dL (GFR of 23.4 mL/min/1.73m2). We recommend that the BK virus be considered as a cause of renal dysfunction in heart transplant recipients, with the aim of detecting its replication in time to reduce immunosuppressive therapy before irreversible compromise of renal function may manifest.Developing the highly efficient and low-cost electrocatalysts for the oxygen evolution reactions (OERs), as vital half reactions of water splitting, is crucial for renewable energy technology. Benefiting from the ultrathin amorphous structure and multi-metal coordination, Fe-CoMo UH nanosheets exhibit outstanding performance for OERs with a low overpotential of 245 mV at 10 mA cm-2, a small Tafel slope of 37 mV dec-1 and an excellent stability for 90 h. Previous investigations have shown that an anti-proliferative drug eluting coating on the IOL surface provides an effective way to inhibit PCO. However, due to the undesirable elution of the anti-proliferative drug, the safety of such modification is one of the important issues to be solved. In this investigation, photodynamic coating was introduced into IOL surface modification. Here, we report the presence of unique topological electronic properties in a 2D planar octagon TiB4 compound. Particularly, without considering the spin-orbit coupling (SOC), we found that the material showed a coexistence of novel quadratic node (QN), and two different types of nodal rings (NRs), namely type-I and type-II. The protection mechanism of fermions has been fully clarified in this study. Furthermore, these fermions showed clear edge states. It is worth noting that QN had a topological charge of 2 since it is different from linear nodes and exhibit clear Fermi arc edge states. Under lattice strain, we found that the system could further exhibit rich topological phase transition. link3 When SOC was included, we determined that these crossing points open very tiny energy gaps, which were smaller than previously reported 3D and 2D examples. Here, we study the effect of the phase state of this nanoscale meniscus-consisting of coexisting monolayer, bilayer and trilayer phase domains-on the frictional behavior during tip sliding by means of molecular dynamics simulations. While the meniscus experiences a domain-by-domain liquid-to-solid phase transition induced by lateral compression, we observe an evident transition in measured friction curves from continuous sliding to stick-slip and meanwhile a gradual increase in friction forces. Moreover, the stick-slip friction can be modulated by varying lattice orientation of the monolayer ice domain in the meniscus, choosing the sliding direction or applying in-plane strains to the substrate. Our results shed light on the rational design of high-performance micro- and nano-electromechanical systems relying on hydration lubrication.
