Two- to three-week-old pea seedlings (Pisum sativum L.) and their protoplasts were investigated. Photosynthesis and respiration in intact seedlings were measured using a GFS-3000 gas analyzer, Dual-PAM-100 Pulse-Amplitude-Modulation (PAM)-fluorometer, and a Dual-PAM gas-exchange Cuvette 3010-Dual. Electrical activity was measured using extracellular electrodes. The parameters of photosynthetic light reactions in protoplasts were measured using the Dual-PAM-100; photosynthesis- and respiration-related changes in O2 exchange rate were measured using an Oxygraph Plus System. We found that preliminary changes in the activity of H+-ATPase in the plasma membrane (its inactivation by sodium orthovanadate or activation by fusicoccin) influenced the amplitudes and magnitudes of VP-induced photosynthetic and respiratory responses in intact seedlings. Decreases in H+-ATPase activity (sodium orthovanadate treatment) induced fast decreases in photosynthetic activity and increases in respiration in protoplasts. Following US FDA approval of the first immune checkpoint inhibitor (ICI), ipilimumab (human IgG1 k anti-CTLA-4 monoclonal antibody), in 2011, and of the first oncolytic virus, Imlygic (talimogene laherparepvec), in 2015, there has been renewed interest in IO. In the past decade, ICIs have changed the treatment paradigm for many cancers by enabling better therapeutic control, resuming immune surveillance, suppressing tumor immunosuppression, and restoring antitumor immune function. However, ICI therapies are effective only in a small subset of patients and show limited therapeutic potential due to their inability to demonstrate efficacy in 'cold' or unresponsive tumor microenvironments (TMEs). Relatedly, oncolytic viruses (OVs) have been shown to induce antitumor immune responses, augment the efficacy of existing cancer treatments, and reform unresponsive TME to turn 'cold' tumors 'hot,' increasing their susceptibility to checkpoint blockade immunotherapies. For this reason, OVs serve as ideal complements to ICIs, and multiple preclinical studies and clinical trials are demonstrating their combined therapeutic efficacy. This review will discuss the merits and limitations of OVs and ICIs as monotherapy then progress onto the preclinical rationale and the results of clinical trials of key combination therapies.Non-destructive testing (NDT) methods are an important means to detect and assess rock damage. To better understand the accuracy of NDT methods for measuring damage in sandstone, this study compared three NDT methods, including ultrasonic testing, electrical impedance spectroscopy (EIS) testing, computed tomography (CT) scan testing, and a destructive test method, elastic modulus testing. link2 Sandstone specimens were subjected to different levels of damage through cyclic loading and different damage variables derived from five different measured parameters-longitudinal wave (P-wave) velocity, first wave amplitude attenuation, resistivity, effective bearing area and the elastic modulus-were compared. The results show that the NDT methods all reflect the damage levels for sandstone accurately. The damage variable derived from the P-wave velocity is more consistent with the other damage variables, and the amplitude attenuation is more sensitive to damage. The damage variable derived from the effective bearing area is smaller than that derived from the other NDT measurement parameters. Resistivity provides a more stable measure of damage, and damage derived from the acoustic parameters is less stable. By developing P-wave velocity-to-resistivity models based on theoretical and empirical relationships, it was found that differences between these two damage parameters can be explained by differences between the mechanisms through which they respond to porosity, since the resistivity reflect pore structure, while the P-wave velocity reflects the extent of the continuous medium within the sandstone.Bilayer ion-exchange membranes are mainly used for separating single and multiply charged ions. link3 It is well known that in membranes in which the layers have different charges of the ionogenic groups of the matrix, the limiting current decreases, and the water splitting reaction accelerates in comparison with monolayer (isotropic) ion-exchange membranes. It was revealed that in bilayer membranes, the limiting current's value is determined by the properties of a thin surface film (modifying layer). A linear regularity of the dependence of the non-equilibrium effective rate constant of the water-splitting reaction on the resistance of the bipolar region, which is valid for both bilayer and bipolar membranes, has been revealed. It is shown that the introduction of the catalyst significantly reduces the water-splitting voltage, but reduces the selectivity of the membrane. In this article, we review how the identification of TVM cells necessitates a re-evaluation of accepted paradigms for conventional memory T (TMEM) cells, the potential for heterogeneity within the TVM population, and the defining characteristics of TVM cells. Further, we highlight recent literature documenting the development of TVM cells as a distinct CD8+ T cell lineage as well their biological significance in the context of disease.A phase-field description of brittle fracture is employed in the reported four-point bending analyses of monolithic and laminated glass plates. Our aims are (i) to compare different phase-field fracture formulations applied to thin glass plates, (ii) to assess the consequences of the dimensional reduction of the problem and mesh density and refinement, and (iii) to validate for quasi-static loading the time-/temperature-dependent material properties we derived recently for two commonly used polymer foils made of polyvinyl butyral or ethylene-vinyl acetate. As the nonlinear response prior to fracture, typical of the widely used Bourdin-Francfort-Marigo model, can lead to a significant overestimation of the response of thin plates under bending, the numerical study investigates two additional phase-field fracture models providing the linear elastic phase of the stress-strain diagram. The typical values of the critical fracture energy and tensile strength of glass lead to a phase-field length-scale parameter that is challenging to resolve in the numerical simulations. Therefore, we show how to determine the fracture energy concerning the applied dimensional reduction and the value of the length-scale parameter relative to the thickness of the plate. The comparison shows that the phase-field models provide very good agreement with the measured stresses and resistance of laminated glass, despite the fact that only one/two cracks are localised using the quasi-static analysis, whereas multiple cracks evolve during the experiment. It was also observed that the stiffness and resistance of the partially fractured laminated glass can be well approximated using a 2D plane-stress model with initially predefined cracks, which provides a better estimation than the one-glass-layer limit.Citrinin (CIT) is a polyketide mycotoxin occurring in a variety of food and feedstuff, among which cereal grains are the most important contaminated source. Pigs and poultry are important livestock animals frequently exposed to mycotoxins, including CIT. Concerns are rising related to the toxic, and especially the potential nephrotoxic, properties of CIT. The purpose of this study was to clarify the histopathological effects on kidneys, liver, jejunum and duodenum of pigs, broiler chickens and laying hens receiving CIT contaminated feed. During 3 weeks, pigs (n = 16) were exposed to feed containing 1 mg CIT/kg feed or to control feed (n = 4), while 2 groups of broiler chickens and laying hens (n = 8 per group) received 0.1 mg CIT/kg feed (lower dose group) and 3 or 3.5 mg CIT/kg feed (higher dose group), respectively, or control feed (n = 4). CIT concentrations were quantified in plasma, kidneys, liver, muscle and eggs using a validated ultra-high performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) method.
