Epigenetic analyses, RNA sequencing, and flow cytometry revealed that EOMES is not expressed in CLL cells, but in CD8+ T cells for which EOMES is a known master regulator. We thus hypothesized that the increased CLL risk associated with the EOMES SNP might be explained by its negative impact on CD8+ T-cell-mediated immune control of CLL. Flow cytometry analyses revealed a higher EOMES expression in CD8+ T cells of CLL patients compared to healthy individuals, and an accumulation of PD-1+ EOMES+ CD8+ T cells in lymph nodes rather than blood or bone marrow in CLL. This was in line with an observed expansion of EOMES+ CD8+ T cells in the spleen of leukemic Eµ-TCL1 mice. As EOMES expression was highest in CD8+ T cells that express inhibitory receptors, an involvement of EOMES in T-cell exhaustion and dysfunction seems likely. Interestingly, Eomes-deficiency in CD8+ T cells resulted in their impaired expansion associated with decreased CLL control in mice. Overall, these observations suggest that EOMES is essential for CD8+ T-cell expansion and/or maintenance, and therefore involved in adaptive immune control of CLL.Newly learned information undergoes a process of awake reactivation shortly after the learning offset and we recently demonstrated that this effect can be observed as early as area V1. However, reactivating all experiences can be wasteful and unnecessary, especially for familiar stimuli. Therefore, here we tested whether awake reactivation occurs differentially for new and familiar stimuli. Subjects completed a brief visual task on a stimulus that was either novel or highly familiar due to extensive prior training on it. Replicating our previous results, we found that awake reactivation occurred in V1 for the novel stimulus. On the other hand, brief exposure to the familiar stimulus led to 'awake suppression' such that neural activity patterns immediately after exposure to the familiar stimulus diverged from the patterns associated with that stimulus. Further, awake reactivation was observed selectively in V1, whereas awake suppression had similar strength across areas V1-V3. These results are consistent with the presence of a competition between local awake reactivation and top-down awake suppression, with suppression becoming dominant for familiar stimuli.Focal adhesion kinase (FAK) is both a non-receptor tyrosine kinase and an adaptor protein that primarily regulates adhesion signalling and cell migration, but FAK can also promote cell survival in response to stress. FAK is commonly overexpressed in cancer and is considered a high-value druggable target, with multiple FAK inhibitors currently in development. Evidence suggests that in the clinical setting, FAK targeting will be most effective in combination with other agents so as to reverse failure of chemotherapies or targeted therapies and enhance efficacy of immune-based treatments of solid tumours. Here, we discuss the recent preclinical evidence that implicates FAK in anticancer therapeutic resistance, leading to the view that FAK inhibitors will have their greatest utility as combination therapies in selected patient populations.The presence of multiple Australopithecus species at Sterkfontein Member 4, South Africa (2.07-2.61 Ma), is highly contentious, and quantitative assessments of craniodental and postcranial variability remain inconclusive. Using geometric morphometrics, we compared the sacrum of the small-bodied, presumed female subadult Australopithecus africanus skeleton Sts 14 to the large, alleged male adult StW 431 against a geographically diverse sample of modern humans, and two species of Pan, Gorilla, and Pongo. The probabilities of sampling morphologies as distinct as Sts 14 and StW 431 from a single species ranged from 1.3 to 2.5% for the human sample, and from 0.0 to 4.5% for the great apes, depending on the species and the analysis. Sexual dimorphism and developmental or geologic age could not adequately explain the differences between StW 431 and Sts 14, suggesting that they are unlikely to be conspecific. This supports earlier claims of taxonomic heterogeneity at Sterkfontein Member 4.We investigated the practical aspects of the application of craniospinal irradiation using helical tomotherapy (HT-CSI) by evaluating interfractional setup errors and intrafractional movement during each treatment in 83 patients undergoing HT-CSI between January 2014 and December 2018. Interfractional setup errors in each axis (mediolateral; ML, craniocaudal; CC, and anteroposterior; AP) were assessed as differences between pre-treatment megavoltage computed tomography (MVCT) images scanned (zygomatic arch to the C4 spine) and planning CT images. Intrafractional movements were evaluated as the difference between pre-treatment and post-treatment MVCT (T12-L4 spine) images at each fraction. Median interfractional setup error was acceptable in every axis (ML 1.6 mm, CC 1.9 mm, AP 3.1 mm). Seven patients (8.4%) experienced significant intrafractional displacement from 1 to 10 fractions (0.34% for ML, 0.74% for CC, 1.21% for AP). Weight loss grade 1+ during treatment (p = 0.016) was an independent risk factor for significant intrafractional displacement. The risk factor for significant intrafractional movement in pediatric patients was weight loss grade 1+ (p = 0.020), while there was no factor in adults. HT-CSI could be a feasible treatment modality with acceptable setup verification. Inter- and intrafractional errors were acceptable; paying attention to weight loss during treatment is necessary, especially in pediatric patients.Standard treatment for HIV infection involves a combination of antiretrovirals. Additionally, opportunistic infections in HIV infected patients require further antimicrobial medications that might cause drug-drug interactions (DDIs). The objective of this study was to to compare the recognition of DDIs between antiretrovirals and antimicrobials by three proprietary databases and evaluate their concordance. 114 items of antiretrovirals and antimicrobials from the National List of Essential Medicines of Thailand 2018 were used in the study. However, 21 items were not recognised by Micromedex, Drugs.com, and Liverpool HIV interactions. Only 93 items were available for the detection of potential DDIs by the three databases. Potential DDIs detected from the three databases included 292 pairs. Liverpool showed the highest number of DDIs with 285 pairs compared with 259 pairs by drugs.com and 133 pairs by Micromedex. Regarding the severity classifications, Liverpool reported 10% Contraindicated; Micromedex reported 14% contraindicated and 59% major; Drugs.com reported 21% major. The Fleiss' kappa agreements were fair to poor among the three databases, higher agreement was observed for DDIs classified as severe. We aimed to show whether silver fir (Abies alba Mill.) can be an appropriate host partner for commercial mycorrhization with truffles, and how earthworms in the inoculation substrate would affect the mycorrhization dynamics. Silver fir seedlings inoculated with Tuber. aestivum were analyzed for root system parameters and mycorrhization, how earthworms affect the bare root system, and if mycorrhization parameters change when earthworms are added to the inoculation substrate. Seedlings were analyzed 6 and 12 months after spore inoculation. Mycorrhization with or without earthworms revealed contrasting effects on fine root biomass and morphology of silver fir seedlings. Here, we studied how visual distinctiveness signals along two feature dimensions-shape and surface texture-combine to determine the overall distinctiveness of an object in the scene. Distinctiveness scores between a target object and distractors were measured separately for shape and texture using a search task. These scores were then used to predict search times when a target differed from distractors along both shape and texture. Model comparison showed that the overall object distinctiveness was best predicted when shape and texture combined using a Euclidian metric, confirming the brain is computing independent distinctiveness scores for shape and texture and combining them to direct attention.This study aimed to replicate and validate concreteness and context effects on semantic word processing. In Experiment 1, we replicated the behavioral findings of Hoffman et al. (Cortex 63,250-266, https//doi.org/10.1016/j.cortex.2014.09.001 , 2015) by applying their cueing paradigm with their original stimuli translated into German. We found concreteness and contextual cues to facilitate word processing in a semantic judgment task with 55 healthy adults. The two factors interacted in their effect on reaction times abstract word processing profited more strongly from a contextual cue, while the concrete words' processing advantage was reduced but still present. For accuracy, the descriptive pattern of results suggested an interaction, which was, however, not significant. In Experiment 2, we reformulated the contextual cues to avoid repetition of the to-be-processed word. In 83 healthy adults, the same pattern of results emerged, further validating the findings. Our corroborating evidence supports theories integrating representational richness and semantic control mechanisms as complementary mechanisms in semantic word processing.
