The objective of the current study is to accomplish a relative exploration of the biological roles of differentially dysregulated genes (DRGs) in type 2 diabetes mellitus (T2DM). The study aimed to determine the impact of these DRGs on the biological pathways and networks that are related to the associated disorders and complications in T2DM and to predict its role as prospective biomarkers.
Datasets obtained from metabolomic and proteomic profiling were used for investigation of the differential expression of the genes. A subset of DRGs was integrated into IPA software to explore its biological pathways, related diseases, and their regulation in T2DM. #link# Upon entry into the IPA, only 94 of the DRGs were recognizable, mapped, and matched within the database.
The study identified networks that explore the dysregulation of several functions; cell components such as degranulation of cells; molecular transport process and metabolism of cellular proteins; and inflammatory responses. Top disorders associated wits made it probable to advantage in understanding the role of these biomarkers in the identification of significant biological processes, pathways, and regulators that are associated with T2DM and its comorbidities.
Diabetes can increase oxidative stress in various tissues of the body, and the progress of this process is associated with intensification of the complications of diabetes. The current study purposed to evaluate the protective effect of
(PG) seed on oxidative stress induced by diabetes in the liver and serum of streptozotocin (STZ)-induced diabetic rats.
This interventional study was performed on 36 male Wistar rats. The rats were randomly divided into 6 groups (healthy controls, healthy treated with 3% and 6% (PG), diabetic control, diabetic treated with 3% and 6% (PG) doses). After 6 weeks of intervention, weight, glucose, and oxidative stress parameters in serum and liver including total antioxidant capacity, malondialdehyde, total thiol and superoxide dismutase activity were measured. Data analysis was done by statistical software version 16 and Tukey's one-way ANOVA tests.
Diabetic rats showed significantly higher malondialdehyde and fasting glucose levels (12±1.2 mmol/L) and significant reductiht loss, and improve oxidative stress indices in the serum and liver of STZ-induced diabetic rats. It is a potential therapeutic food supplement for the treatment and prevention of hyperglycemia and high oxidative stress of diabetes.
Emerging evidence suggests that garlic (
L.) and its bioactive components can mitigate hepatic steatosis by the modulation of hepatic lipid metabolism. We aimed to assess the efficacy of the garlic administration on hepatic steatosis in patients with NAFLD.
This clinical trial was conducted on adult patients with ultrasound-diagnosed NAFLD. Eligible participants were randomly assigned, with the use of the stratified blocked procedure, to receive 800 mg garlic or placebo for 15 weeks. The primary outcome was the improvement in the hepatic steatosis diagnosed by ultrasound technique after 15 weeks of intervention.
A total of 110 patients underwent randomization, and 98 patients completed the trial. Twenty-four (51.1%) patients in the garlic group achieved improvement in the hepatic steatosis compared to eight (15.7%) patients in the placebo group with the relative risk of 5.6 (95% CI 2.17 to 14.5; P=0.001), which remained significant after adjusting for baseline value of hepatic steatosis. There were significant reductions in weight and serum ALT, AST, FBS, Hb A1C, total cholesterol, LDL-cholesterol, and TG concentration with the garlic intake compared to placebo (P<0.05). The results were also significant after adjusting for weight change, energy intake, and physical activity. No serious adverse effects were observed with the garlic intake.

The optimal blood pressure (BP) level for diabetic patients remains controversial, while studies provided limited evidence on BP management for individuals with normoglycemia or prediabetes. We aimed to investigate the associations between systolic blood pressure (SBP) and all-cause and cardiovascular (CVD) mortality among US adults with different glycemic profiles.
We used data from the 1999-2014 National Health and Nutrition Examination Survey (NHANES, n=40,046) with comprehensive baseline examination and follow-up assessment. Restricted cubic spline analysis was performed to examine dose-response relationship between SBP and mortality risk. Cox regression models were used to estimate hazard ratios of all-cause mortality and CVD mortality for SBP categories.
U-shaped associations between SBP and all-cause mortality were observed regardless of glucose status. The relationship between SBP and CVD mortality was found to be U-shaped only in normoglycemic participants. The lowest mortality risk of optimal e optimal SBP range was gradually higher with worsening glucose status.

) is a second-generation once-daily basal insulin. Previous randomized controlled trials showed comparable HbA1c reductions with lower rates of hypoglycemia of Gla-300 versus Gla-100.
We report the 12 months results of the Swiss cohort of Toujeo-1, a prospective, observational multicenter study exploring the real-world effectiveness of Gla-300 in adult patients with type 2 diabetes (T2D) uncontrolled (HbA1c 7.5-10%) on oral therapy and compared these to the overall Toujeo-1 cohort (conducted in Switzerland and Germany). Primary endpoint was the percentage of patients achieving individual HbA1c targets. Secondary endpoints included changes in HbA1c, fasting plasma glucose (FPG), body weight, insulin dose, incidence of hypoglycemia and overall safety.
The analysis included 47 patients (14 women) with a mean age of 64.1 years and a diabetes duration of 8.4 years. Swiss physicians determined a higher HbA1c treatment target (7.4 vs. 7.0%) and patients received highn, Gla-300 improves glycemic control with a low risk of hypoglycemia and no increase of body weight. The results for Switzerland are consistent with those reported for the overall Toujeo-1 cohort and reveal that treatment targets and approaches slightly differ between both countries.
Carotenoids are a large group of natural pigments that occur in many foods, fruits, and vegetables. link3 Several studies have shown a number of biological properties of carotenoids, particularly beneficial impacts on cancer, metabolic, neurodegenerative, and cardiovascular diseases. However, recent evidence has shown that these compounds could prevent, delay, and ameliorate diabetic retinopathy (DR). The aim of current study was to review the therapeutic effects of carotenoids in the treatment of DR and discuss the molecular mechanisms that are behind these pharmacological activities.
Six online databases (Medline/PubMed, Scopus, Web of Knowledge, Embase, ScienceDirect, and ProQuest) were searched until September 2019. The systematic review was carried out using the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) checklist.
A total of 25 studies were included after the final retrieval. A relationship was observed between carotenoids and management of DR. Findings also demonstrated that the underlying mechanism of beneficial effects of these compounds was antioxidant, anti-inflammatory, anti-angiogenic, and neuroprotective properties.
Carotenoids potentially delay the initiation and prevent the progression of DR; however, ample preclinical studies are required to confirm their effect, and adequate clinical trials are needed to really understand how well these compounds influence DR among humans.

Type 2 diabetes mellitus (T2DM) is characterized by insulin resistance and abnormalities in insulin production. Apelin is associated with insulin resistance. According to the anti-diabetic properties of curcumin, the purpose of this study was to compare the effects of curcumin and nano-curcumin intake on insulin resistance and serum levels of fasting blood sugar (FBS), Apelin, and lipid profile (cholesterol, triglyceride, LDL, HDL and VLDL) in T2DM rats.
Forty-eight male Wistar rats were divided into six groups Control, diabetic, diabetic treated with two doses of curcumin (100 and 200 mg/kg) and diabetic treated with two doses of nano-curcumin (100 and 200 mg/kg). Induction of T2DM was performed by intraperitoneal injection of Nicotinamide (110 mg/kg) and Streptozotocin (45 mg/kg) in the fasting state. Luminespib clinical trial received different doses of nano-curcumin and curcumin by gavage (daily) for 28 days. At the end of the intervention period, insulin resistance and serum levels of FBS, apelin and lipid profiles were measured.
Insulin resistance and serum levels of FBS, Apelin, cholesterol, triglycerides, LDL, and VLDL were significantly decreased in diabetic rats treated with curcumin and nano-curcumin (p<0.05) so that nano-curcumin in reducing lipid profile is more effective than curcumin (P<0.05). Serum level of HDL in nano-curcumin groups was significantly higher than diabetic and curcumin groups (p<0.05). Also, with increasing insulin resistance, serum level of apelin increased (P<0.05).
The therapeutic effects of curcumin and nano-curcumin were effective in decreasing insulin resistance, serum levels of FBS, apelin and lipid profile. The dose of 100 mg/kg nano-curcumin was more effective in reducing lipid profile.
