In this paper we present a novel conditional Triplet loss for solving few-shot problems using deep metric learning. While the conventional Triplet loss suffers the limitation of random sampling of triplets which leads to slow convergence in training process, our proposed network tries to distinguish between samples so that it improves the training speed. Our main contributions are two-fold. (i) We propose a conditional Triplet loss to train a deep Triplet network for deep metric embedding. The proposed Triplet loss employs a penalty-reward technique to enhance the convergence of standard Triplet loss. (ii) We improve the performance of the existing image co-segmentation model by replacing the conventional loss function by our proposed conditional Triplet loss. To demonstrate the performance of the proposed network, experiments carry out on MNIST and CIFAR. Simulation results are evaluated by AUC and Recall (sensitivity) and indicate that the proposed conditional Triplet network achieves higher accuracy in comparison to state-of-the-arts.Deep neural networks (DNNs) with a complex structure and multiple nonlinear processing units have achieved great successes for feature learning in image and visualization analysis. Due to interpretability of the "black box" problem in DNNs, however, there are still many obstacles to applications of DNNs in various real-world cases. This paper proposes a new DNN model, knowledge-based deep stacked denoising auto-encoders (KBSDAE), which inserts the knowledge (i.e., confidence and classification rules) into the deep network structure. This model not only can offer a good understanding of the representations learned by the deep network but also can produce an improvement in the learning performance of stacked denoising auto-encoder (SDAE). The knowledge discovery algorithm is proposed to extract confidence rules to interpret the layerwise network (i.e., denoising auto-encoder (DAE)). The symbolic language is developed to describe the deep network and shows that it is suitable for the representation of quantitative reasoning in a deep network. The confidence rule insertion to the deep network is able to produce an improvement in feature learning of DAEs. The classification rules extracted from the data offer a novel method for knowledge insertion to the classification layer of SDAE. The testing results of KBSDAE on various benchmark data indicate that the proposed method not only effectively extracts knowledge from the deep network, but also shows better feature learning performance than that of those typical DNNs (e.g., SDAE).Soil salinity is among the crucial factors that impact on crop productivity, including oat (Avena sativa L.). Herein, we used two distinct oat cultivars with varied salt tolerance levels to unravel adaptive responses to salt stress by metabolomic and transcriptomic characterization. Metabolomic profiling revealed 201 metabolites, including saccharides, amino acids, organic acids, and secondary metabolites. The levels of most saccharides and amino acids were elevated in Baiyan 2 (BY2) as well as in Baiyan 5 (BY5) exposed to salt stress. In the tolerant cultivar BY2 exposed to 150 mM NaCl, concentrations of most of the metabolites increased significantly, with sucrose increased by 38.34-fold, Sophorose increased by 314.15-fold and Isomaltose 2 increased by 25.76-fold. In the sensitive cultivar BY5, the concentrations of most metabolites increased after the plant was exposed to 150 mM NaCl but decreased after the plant was exposed to 300 mM NaCl. Although plants have no caspase-3 homologs, PBA1 acts as a plant caspase-3-like enzyme in plant programmed cell death (PCD). PCD occurs during the formation of secretory cavities in Citrus fruits; hence, secretory cavities could be utilized as a new cell biology model for investigating the regulatory mechanisms of plant PCD. To further study the association between PBA1 and PCD during secretory cavity development in Citrus fruits, CgPBA1 was identified in the fruit of Citrus grandis 'Tomentosa'. The temporal and spatial expression of CgPBA1 during secretory cavity development were analyzed using quantitative real-time PCR and in situ hybridization, and the morphological changes in the apoptotic cell nuclei were observed using TUNEL assay and ultra-thin section technology. The results revealed that the full-length cDNA of CgPBA1 contains a 711 bp ORF that encodes a putative protein containing 236 amino acid with a proteasome-β-6 functional domain that belongs to the Ntn hydrolase super family. CgPBA1 was predominantly expressed in the secretory cavities; its expression changes coincided with the morphological changes and DNA fragmentation in apoptotic cell nuclei. The green fluorescent fusion protein of CgPBA1 is also located in the nucleus of tobacco epidermal cells. Based on previous research and the findings of the present study, we speculate that CgPBA1 is a highly functional conserved protein in plants, and it might be involved in nuclear degradation during PCD for secretory cavity formation in C. grandis 'Tomentosa' fruits.Riboswitches are metabolite sensing aptamer domains present in non-coding regions in RNA and act as gene-regulating elements. Thiamine pyrophosphate (TPP) riboswitch is evolved as a new target for developing antibiotics against many pathogenic bacteria. The earlier reports suggest that the modification of the pyrophosphate group in the ligand molecule can enhance gene expression. In this work, we have examined the binding affinity and efficacy of TPP and two recently reported ligands, CH2-TPP, and CF2-TPP, using Well-tempered metadynamics (WT-MtD) simulations. The experimental in vitro assays show that both TPP and CH2-TPP repress the gene expression to the same extent. The calculated binding energies correlate well with the experimental study and show the same trend of binding affinity of ligands for the TPP riboswitch. The root mean square fluctuation profiles suggest that the CH2-TPP and TPP trigger higher fluctuations in P1 and L3 region, and such fluctuations in the P1 region is involved in the gene regulation process. The metal ion mediated contact of TPP ligand with pyrophosphate binding helix is found to be critical in the gene regulation process. The simulation results corroborate the experimental observations that the role of conformational changes occurring in different riboswitch regions upon ligand binding is essential to repress the gene expression process. The analysis methods and results of this study may be of further use in understanding ventricular vascular coupling of transported flow variables in healthy sex differences, healthy aging, and various cardiovascular conditions.Muscle models are commonly based on intrinsic properties pooled across a number of individuals, often from a different species, and rarely validated against directly measured muscle forces. Here we use a rich data set of rat medial gastrocnemius muscle forces recorded during in-situ and in-vivo isometric, isotonic, and cyclic contractions to test the accuracy of forces predicted using Hill-type muscle models. We identified force-length and force-velocity parameters for each individual, and used either these subject-specific intrinsic properties, or population-averaged properties within the models. The modeled forces for cyclic in-vivo and in-situ contractions matched with measured muscle-tendon forces with r2 between 0.70 and 0.86, and root-mean square errors (RMSE) of 0.10 to 0.13 (values normalized to the maximum isometric force). The modeled forces were least accurate at the highest movement and cycle frequencies and did not show an improvement in r2 when subject-specific intrinsic properties were used; however, there was a reduction in the RMSE with fewer predictions having higher errors. We additionally recorded and tested muscle models specific to proximal and distal regions of the muscle and compared them to measures and models from the whole muscle belly there was no improvement in model performance when using data from specific anatomical regions. These results show that Hill-type muscle models can yield very good performance for cyclic contractions typical of locomotion, with small reductions in errors when subject-specific intrinsic properties are used.The effect of dry heat (DH, 130 °C, 1, 3 and 9 h), cold plasma (CP, 40 V, 1, 5 and 10 min) and their combination (D-P) treatment on the structure, physicochemical and digestive properties of red adzuki bean starch were studied. The results showed that DH or CP had slight change in morphology while diffraction pattern of starch was remained. With the extension of treated time of DH and CP, the amylose content, crystallinity, molecular weight, short-range order, the long chain of amylopectin, enthalpy value, swelling power, digestibility were reduced, while gelatinization temperatures, the short chain of amylopectin and solubility were increased. The D-P had deeper modification than the single treatment. The combination of dry heat and cold plasma is a simple and green method to improve the starch structure and enhance starch properties and this modified starch could be implemented to tailor starch to the desired food applications.
