By treatment of the run-on RNAs with a mild iodination reaction and subjection of the products to reverse transcription into complementary DNA (cDNA), base mismatch occurs at the original a6A incorporation sites, thus making the RNPII locations detected in the high-throughput cDNA sequencing. The mGRO-seq yields both the map of RNPII sites and the chromatin RNA abundance and holds great promise for the study of single-cell transcriptional activity.Material efficiency (ME) can support rapid climate change mitigation and circular economy. Here, we comprehensively assess the circularity of ME strategies for copper use in the U.S. housing services (including residential buildings and major household appliances) by integrating use-phase material and energy demand. Although the ME strategies of more intensive floor space use and extended lifetime of appliances and buildings reduce the primary copper demand, employing these strategies increases the commonly neglected use-phase share of total copper requirements during the century from 23-28 to 22-42%. Use-phase copper requirements for home improvements have remained larger than the demand gap (copper demand minus scrap availability) for much of the century, limiting copper circularity in the U.S. housing services. Further, use-phase energy consumption can negate the benefits of ME strategies. For instance, the lifetime extension of lower-efficiency refrigerators increases the copper use and net environmental impact by increased electricity use despite reductions from less production. This suggests a need for more attention to the use phase when assessing circularity, especially for products that are material and energy intensive during use. To avoid burden shifting, policymakers should consider the entire life cycle of products supporting services when pursuing circular economy goals.Rheumatoid arthritis (RA) is an autoimmune disease with nearly 1.6 billion patients worldwide and an incidence of 0.5-1%. In recent years, basic and clinical studies have revealed that immune cell responses and corresponding secretion of inflammatory factors are important in the control of RA development. Our study found that a natural plant ingredient, menthone, could be used as a potential antirheumatism compound. In vivo observations demonstrated that menthone alleviates collagen II-induced arthritis (CIA) in mice. Furthermore, we found that menthone regulates the number of Th1 and Th17 cells in CIA mice. Importantly, menthone significantly inhibits the release of pro-inflammatory cytokines, including TNF-α, IL-1β, and IL-6, in CIA mice. Our study suggests a potential component for the development of drugs to treat rheumatoid arthritis.Herein, a N-rich metal-organic framework (MOF) with four kinds of cages, Zn4(ade)2(TCA)2(H2O) (NENU-1000, Hade = adenine, H3TCA = 4,4',4″-tricarboxytriphenylamine, NENU = Northeast Normal University), was prepared by the mixed-ligand strategy. Cationic dyes can be selectively absorbed by NENU-1000 at proper concentrations, but not neutral and anionic dyes, which perhaps can be assigned to the N-rich neutral framework of NENU-1000. The recently proposed sparse Bayesian learning (SBL)-based method can significantly improve the baseline correction performance. However, it has at least two disadvantages (i) it works poorly for large-scale datasets and (ii) it completely ignores the burst-sparsity structure of the sparse representation of the pure spectrum. In this paper, we present a new fast burst-sparsity learning method for baseline correction to overcome these shortcomings. The first novelty of the proposed method is to jointly adopt a down-sampling strategy and construct a multiple measurements block-sparse recovery problem with the down-sampling sequences. The down-sampling strategy can significantly reduce the dimension of the spectrum; while jointly exploiting the block sparsity among the down-sampling sequences avoids losing the information contained in the original spectrum. The second novelty of the proposed method is introducing the pattern-coupled prior into the SBL framework to characterize the inherent burst-sparsity in the sparse representation of spectrum. As illustrated in the paper, burst-sparsity commonly occurs in peak zones with more denser nonzero coefficients. Properly utilizing such burst-sparsity can further enhance the baseline correction performance. Results on both simulated and real datasets (such as FT-IR, Raman spectrum, and chromatography) verify the substantial improvement, in terms of estimation accuracy and computational complexity.This paper reports how the endosomal pathways of nanoparticle (NP) constructs with different surface curvatures are affected by their order of delivery. Sequential incubation of cytosine-phosphate-guanine (CpG)-conjugated spiky and spherical gold NPs with macrophages resulted in different nanoconstruct ratios at the interior edges of endosomes. Application of spiky NPs after spherical NPs accelerated the formation of late-stage endosomes and resulted in larger endosomes, and the spherical NPs were enclosed by the spiky NPs. In contrast, the reverse incubation order produced an asymmetric distribution of the two nanoconstruct shapes in smaller endosomes. Macrophages with a higher proportion of the enclosed spherical NPs as well as a larger ratio of spiky to spherical NPs at the endosomal edge showed enhanced toll-like receptor 9 activation and secretion of proinflammatory cytokines and chemokines. Our results indicate that the subcellular trafficking of targeting nanoconstructs by vesicles is affected by both the delivery order and the endosomal distribution. Our study also establishes a new approach for nanoscale monitoring of intracellular therapeutics delivery with conventional electron microscopy.Detecting metal ions in vivo with a high spatiotemporal resolution is critical to understanding the roles of the metal ions in both healthy and disease states. Although spatiotemporal controls of metal-ion sensors using light have been demonstrated, the lack of penetration depth in tissue and in vivo has limited their application. To overcome this limitation, we herein report the use of high-intensity focused ultrasound (HIFU) to remotely deliver on-demand, spatiotemporally resolved thermal energy to activate the DNAzyme sensors at the targeted region both in vitro and in vivo. A Zn2+-selective DNAzyme probe is inactivated by a protector strand to block the formation of catalytic enzyme structure, which can then be activated by an HIFU-induced increase in the local temperature. With this design, Zn2+-specific fluorescent resonance energy transfer (FRET) imaging has been demonstrated by the new DNAzyme-HIFU probes in both HeLa cells and mice. The current method can be applied to monitor many other metal ions for in vivo imaging and medical diagnosis using metal-specific DNAzymes that have either been obtained or can be selected using in vitro selection.The molecular composition of atmospheric particulate matter (PM) in the urban environment is complex, and it remains a challenge to identify its sources and formation pathways. Here, we report the seasonal variation of the molecular composition of organic aerosols (OA), based on 172 PM2.5 filter samples collected in Beijing, China, from February 2018 to March 2019. We applied a hierarchical cluster analysis (HCA) on a large nontarget-screening data set and found a strong seasonal difference in the OA chemical composition. Molecular fingerprints of the major compound clusters exhibit a unique molecular pattern in the Van Krevelen-space. We found that summer OA in Beijing features a higher degree of oxidation and a higher proportion of organosulfates (OSs) in comparison to OA during wintertime, which exhibits a high contribution from (nitro-)aromatic compounds. OSs appeared with a high intensity in summer-haze conditions, indicating the importance of anthropogenic enhancement of secondary OA in summer Beijing. Furthermore, we quantified the contribution of the four main compound clusters to total OA using surrogate standards. With this approach, we are able to explain a small fraction of the OA (∼11-14%) monitored by the Time-of-Flight Aerosol Chemical Speciation Monitor (ToF-ACSM). However, we observe a strong correlation between the sum of the quantified clusters and OA measured by the ToF-ACSM, indicating that the identified clusters represent the major variability of OA seasonal cycles. This study highlights the potential of using nontarget screening in combination with HCA for gaining a better understanding of the molecular composition and the origin of OA in the urban environment.The hybrid coupling of biocatalysts and chemical catalysts plays a vital role in the fields of catalysis, sensing, and medical treatment due to the integrated advantages in the high activity of natural enzymes and the excellent stability of nanozymes. Herein, a new nanozyme/natural enzyme hybrid biosensor was established for ultrasensitive glutamate detection. The MIL-88B(Fe)-NH2 material with remarkable peroxidase mimic activity and stability was used as a nanozyme and carrier for immobilizing glutamate oxidase (GLOX) through Schiff base reaction to construct a chem-enzyme cascade detector (MIL-88B(Fe)-NH2@GLOX). The resultant MIL-88B(Fe)-NH2@GLOX exhibited a wide linear range (1-100 μM), with a low detection limit of 2.5 μM for glutamate detection. Furthermore, the MIL-88B(Fe)-NH2@GLOX displayed excellent reusability and storage stability. After repeated seven cycles, MIL-88B(Fe)-NH2-GLOX (GLOX was adsorbed on MIL-88B(Fe)-NH2) lost most of its activity, whereas MIL-88B(Fe)-NH2@GLOX still retained 69% of its initial activity.
