Host factors interacting with p8 and mediating p8 transfer are unknown. Here, we report that vasodilator-stimulated phosphoprotein (VASP), which promotes actin filament elongation, is a novel interaction partner of p8 and important for p8 and HTLV-1 Gag cell-to-cell transfer. VASP contains an Ena/VASP homology 1 (EVH1) domain that targets the protein to focal adhesions. Bioinformatics identified a short stretch in p8 (amino acids (aa) 24-45) which may mediate interactions with the EVH1 domain of VASP. Co-immunoprecipitations confirmed interactions of VASPp8 in 293T, Jurkat and HTLV-1-infected MT-2 cells. Co-precipitation of VASPp8 could be significantly blocked by peptides mimicking aa 26-37 of p8. Mutational studies revealed that the EVH1-domain of VASP is necessary, but not sufficient for the interaction with p8. Further, deletion of the VASP G- and F-actin binding domains significantly diminished co-precipitation of p8. Imaging identified areas of partial co-localization of VASP with p8 at the plasma membrane and in protrusive structures, which was confirmed by proximity ligation assays. Co-culture experiments revealed that p8 is transferred between Jurkat T-cells via VASP-containing conduits. Imaging and flow cytometry revealed that repression of both endogenous and overexpressed VASP by RNA interference or by CRISPR/Cas9 reduced p8 transfer to the cell surface and to target Jurkat T-cells. Stable repression of VASP by RNA interference in chronically infected MT-2 cells impaired both p8 and HTLV-1 Gag transfer to target Jurkat T-cells, while virus release was unaffected. Thus, we identified VASP as a novel interaction partner of p8, which is important for transfer of HTLV-1 p8 and Gag to target T-cells.Patellofemoral pain (PFP) is commonly caused by abnormal pressure on the knee due to excessive load while standing, squatting, or going up or down stairs. The JCS coordinates with statistically significant difference can serve as key biomarkers of patellar motion when evaluating a patient suspected of having PFP. The proposed method could reveal diagnostic biomarkers for accurately identifying PFP patients and be an effective addition to clinical diagnosis before surgery and to help plan rehabilitation strategies.Music is considered a vital element in young people's lives. It functions as an important means for expressing the emotions and feelings they experience in a daily basis. As such, this study explores the music preferences of high school Brazilian students (N = 940), 530 female (56.9%) and 410 male (43.1%) participants between 14 and 20 years old (M = 16.14 years old, SD = 1.22). The main instrument for data collection was the Questionnaire on Musical Style Preferences, which was adapted to the Brazilian context and encompassed 33 different music styles. A principal component analysis resulted in five dimensions representing different musical styles (1) Intense, (2) Unique, (3) Sophisticated, (4) Contemporary, and (5) Mellow. The results of this study reinforced theory of the five-factor model of musical preference. Results also suggest that Mellow music was the most preferred while Sophisticated music was the least preferred among participants. Regarding gender, male participants showed a greater preference towards Contemporary, Intense, and Sophisticated music, whereas women generally preferred Mellow and Unique. Regarding age, participants under 20 years old showed a greater preference towards Mellow musical styles as compared to older participants. On the one hand, regression analyses showed that preferences towards Intense music decrease with age. On the other hand, gender was a better predictor for music preferences than age. Although the results of this study correspond to those of previous studies, more research studies are necessary to further explain musical preferences within the Brazilian context.Banxia Houpu decoction (BXHPD) has been used to treat depression in clinical practice for centuries. However, the pharmacological mechanisms of BXHPD still remain unclear. Network Pharmacology (NP) approach was used to explore the potential molecular mechanisms of BXHPD in treating depression. Potential active compounds of BXHPD were obtained from the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform Database. STRING database was used to build a interaction network between the active compounds and target genes associated with depression. The topological features of nodes were visualized and calculated. Significant pathways and biological functions were identified using Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analyses. A total of 44 active compounds were obtained from BXHPD, and 121 potential target genes were considered to be therapeutically relevant. Pathway analysis indicated that MAPK signaling pathway, ErbB signaling pathway, HIF-1 signaling pathway and PI3K-Akt pathway were significant pathways in depression. They were mainly involved in promoting nerve growth and nutrition and alleviating neuroinflammatory conditions. The result provided some potential ways for modern medicine in the treatment of depression.We report on the results of a Covid-19 contact tracing app measurement study carried out on a standard design of European commuter tram. Our measurements indicate that in the tram there is little correlation between Bluetooth received signal strength and distance between handsets. We applied the detection rules used by the Italian, Swiss and German apps to our measurement data and also characterised the impact on performance of changes in the parameters used in these detection rules. We find that the Swiss and German detection rules trigger no exposure notifications on our data, while the Italian detection rule generates a true positive rate of 50% and a false positive rate of 50%. In the current study, we found that RNA interference of a C-type lectin (HaCTL3) in the cotton bollworm Helicoverpa armigera suppresses ecdysone and juvenile hormone signaling, thus reducing larval body size and delaying pupation. Depletion of HaCTL3 also results in an increased abundance of Enterocuccus mundtii in the hemolymph, which may escape from the gut. Furthermore, HaCTL3 and its controlled antimicrobial peptides (attacin, lebocin, and gloverin) are involved in the clearance of E. mundtii from the hemolymph via phagocytosis or direct bactericidal activity. Injection of E. mundtii into larval hemocoel mimics HaCTL3-depleted phenotypes and suppresses ecdysone and juvenile hormone signaling. Taken together, we conclude that HaCTL3 maintains normal larval growth and development of H. armigera via suppressing the abundance of E. mundtii in the hemolymph. Our results provide the first evidence of an immune system acting on an endocrine system to modulate development via shaping the composition of microbiota in insect hemolymph. Thus, this study will deepen our understanding of the interaction between immunity and development.
High-quality chest compressions is challenging for bystanders and first responders to out-of-hospital cardiac arrest (OHCA). Long compression pauses and compression rates higher than recommended are common and detrimental to survival. Our aim was to design a simple and low computational cost algorithm for feedback on compression rate using the transthoracic impedance (TI) acquired by automated external defibrillators (AEDs).

ECG and TI signals from AED recordings of 242 OHCA patients treated by basic life support (BLS) ambulances were retrospectively analyzed. Beginning and end of chest compression series and each individual compression were annotated. The algorithm computed a biased estimate of the autocorrelation of the TI signal in consecutive non-overlapping 2-s analysis windows to detect the presence of chest compressions and estimate compression rate.

A total of 237 episodes were included in the study, with a median (IQR) duration of 10 (6-16) min. The algorithm performed with a global sensitivity in the detection of chest compressions of 98.
