Hemophilic arthropathy is the most common complication of severe hemophilia A. This study aims to investigated joint status and related risk factors in patients with severe hemophilia A (PWSHA).

This single-center study included 31 patients. Six index joints (both elbows, knees, and ankles) were evaluated using the Hemophilia Early Arthropathy Detection with UltraSound in China (HEAD-US-C) and Hemophilia Joint Health Score (HJHS). Treatment adherence was measured using the Validated Hemophilia Regimen Treatment Adherence Scale-Prophylaxis (VERITAS-Pro). We analyzed the influence of age, treatment delay (the interval between diagnosis and the initiation of treatment), prophylaxis, and treatment adherence on joint outcomes.

All patients were male (median age, 22 years). The median age at diagnosis was 1 year; that at initial treatment was 5 years. All patients experienced joint bleeding. HEAD-US-C and HJHS scores were positively correlated (R = 0.70,
< 0.0001). Median [range] HEAD-US-C and HJHS scores were 15 [0-36] and 32 [2-49], respectively. Age was positively correlated with both HEAD-US-C (
= 0.002) and HJHS scores (
< 0.0001). The difference of HEAD-US-C scores between groups with ≤1 year and >1 year treatment delay was close to significant (
= 0.055). HJHS scores were significantly different between these two groups (
= 0.03). Joint assessment scores were not significantly different between on-demand and low-dose prophylaxis groups. VERITAS-Pro scores were correlated with both HEAD-US-C and HJHS scores (
= 0.046 and
= 0.005, respectively).

Prompt treatment and adherence improvement may reduce severity.This study aimed to explore how cold acclimation (CA) modulates cold stress in tobacco leaves and reveal the relationship between CA and cold stress resistance, and the mechanism of CA-induced plant resistance to cold stress. This study examined the effects of CA treatment (at 8-10℃ for 2 d) on the cold tolerance of tobacco leaves under 4°C cold stress treatment using seedlings without CA treatment as the control (NA). In both CA and NA leaves, cold stress treatment resulted in a decrease in maximum photochemical efficiency of PSII (Fv/Fm), increase in relative variable fluorescence (VJ) at 2 ms on the standardized OJIP curve, inhibition of PSII activity, and impairment of electron transfer on the acceptor side. Besides increasing the malondialdehyde (MDA) content and electrolyte leakage rate, the cold stress exacerbated the degree of membrane peroxidation. The CA treatment also induced the accumulation of reactive oxygen species (ROS), including superoxide anion (O2·-) and H2O2, and increased the activities of antioxidant enzymes, such as superoxide dismutase (SOD), peroxidase (POD), catalase (CAT) and ascorbic acid peroxidase (APX). The CA treatment also enhanced the accumulation of soluble sugar (SS) and soluble protein (SP), cyclic electron flow (CEF), and the proportion of regulatory energy dissipation Y(NPQ). Moreover, CA+ cold stress treatment significantly reduced CEF and Y(NPQ) in tobacco leaves than under NA+ cold stress treatment, thus significantly alleviating the degree of PSII photoinhibition. In conclusion, CA treatment significantly alleviated PSII photoinhibition and oxidative damage in tobacco leaves under cold stress treatment. Improvement in cold resistance of tobacco leaves is associated with the induction of antioxidant enzyme activity, accumulation of osmoregulation substances, and initiation of photoprotective mechanisms.

The aim of this work is to explore the subjective experience of adolescent suicidal behavior to determine the aspects that are common to adolescent girls and boys and those that are specific to one gender.

The study included 22 adolescents receiving care for suicidal behavior. The semi-structured interviews were analyzed according to the methods of Interpretative Phenomenological Analysis.

Three themes of experience emerged the experience of suffering, connection to others, and courage. The individual and interpersonal dimensions highlight feelings that they are not understood by others and are isolated. Here, we isolate 9 neutralizing mouse MAbs raised against the spike protein of a SARS-CoV-2 prototype strain and evaluate their neutralizing potency towards a panel of variants, including B.1.1.7, B.1.351, B.1.617.1, and B.1.617.2. By using a combination of biochemical, virological, and cryo-EM structural analyses, we identify three types of cross-variant neutralizing MAbs, represented by S5D2, S5G2, and S3H3, respectively, and further define their epitopes. S5D2 binds the top lateral edge of the receptor-binding motif within the receptor-binding domain (RBD) with a binding footprint centred around the loop477-489, and efficiently neutralizes all variant pseudoviruses, but the potency against B.1.617.2 was observed to decrease significantly. S5G2 targets the highly conserved RBD core region and exhibits comparable neutralization towards the variant panel. S3H3 binds a previously unreported epitope located within the evolutionarily stable SD1 region and is able to near equally neutralize all of the variants tested.
