Our results indicate that siRNA uptake was not uniform across 3D multicellular spheroids. The electrophoretic migration of nucleic acids upon delivery of unipolar electric field pulses could explain the asymmetry of siRNA uptake. Moreover, a gradient was observed from external layers toward the center, leading to siRNA silencing of GFP positive cells located in the outer rim. While siRNA delivery experiments on spheroids may differ from intratumoral injections, the levels of transfection in spheroids are comparable to levels observed in published studies in vivo. Taken together, our results provide fundamental information about siRNA 3D distribution during electrotransfer, indicating that multicellular spheroids remain a relevant alternative to animal experimentation.During fermentation in Escherichia coli succinate is transported via Dcu transporters, encoded dcuA, dcuB, dcuC and dcuD although the role of DcuD protein has not been elucidated yet. Total H± efflux was decreased ~ 40% while K± uptake was absent in dcuD mutant. DCCD-sensitive H± flux was absent in dcuD nevertheless it was increased ~ 3 fold in dcuACB. K± uptake in dcuACB was stimulated ~ 30% compared to wild type but in DCCD assays K± ions were effluxed with the rate of 0.15 mmol/min per 109 cells/ml. In dcuACB mutant membrane potential (ΔΨ) was ~ 30 mV higher than in wild type. dcuD gene expression was increased in the dcuACB mutant respect to wild type at pH 7.5 (~120%), suggesting that an increment of DcuD activity compensates the lack of DcuA, DcuC and DcuB carriers. It can be concluded that active DcuD is important for H± efflux via the FOF1-ATPase and K± uptake at pH 7.5. In addition, DcuA, DcuB and DcuC transporters are crucial for regulating DCCD-sensitive K± transport and ΔΨ in E. coli.
Opioid agonist treatment (OAT) for opioid use disorders may be delivered at treatment clinics or dispensed from pharmacies, however the type of delivery may be associated with different risks and benefits. The aim of the study was to investigate whether dispensing of methadone or buprenorphine at pharmacies during treatment for opioid use disorders was associated with adverse outcomes.

Retrospective cohort study using a national, linked, population-level data set from Denmark. Patients included were between 18 and 75 years, living in Denmark, and admitted for treatment for opioid use disorders during 2000-2016 (n = 9299). Cox proportional hazards regression was estimated for convictions, non-fatal overdoses, and death, after the first dispensing of either methadone or buprenorphine from a pharmacy after starting treatment.

Of all patients, 68 % had methadone and 31 % had buprenorphine dispensed at a pharmacy. Compared with the time prior to pharmacy dispension, the risk of criminal convictions increasedle and safe provision of OAT may often require more clinical follow-up than what is typically provided when medication is dispensed at pharmacies.Recently, nanomaterials synthesized ecologically using microorganisms have attracted much interest. In the present study, the ability of Gloeophyllum striatum to synthesize silver nanoparticles is described for the first time. Nanoparticles were formed in an eco-friendly extracellular manner and characterized by UV-Vis, FT-IR, MADLS and SEM techniques. The obtained nanoparticles showed excellent activity against gram-positive and gram-negative bacteria. The MIC values for gram-negative bacteria were 15 µM, while for gram-positive strains they reached 30 µM. The haemolytic and cytotoxic activities of the synthesized nanoparticles towards mammalian cells were also determined. The addition of AgNPs at the concentrations above 30 µM caused 50% haemolysis of red blood cells after they 24-hour incubation. A decrease in the viability of fibroblasts by over 50% was also found in the samples treated with nanoparticles at the concentrations above 30 µM. The ecotoxicological risk of silver nanoparticles was assessed using A. franciscana and D. magna crustaceans as well as L. sativum plants. The EC50 values for A. franciscana and D. magna were 61.97 and 0.275 µM, respectively. Photo- and biodegradation were the main degradation mechanisms of these plastics in the air and soil, respectively. Poly(p-dioxanone) (PPDO) exhibited the highest weight loss potentials in both air (54.7 ± 9.1%) and soil (56.8 ± 4.8%), due to its special ether bond and the rich and diverse microorganisms on its biofilms. The microbiota on PPDO was distinct and enriched with Chloroflexi and Firmicutes that responsible for carbon cycle and organic degradation. The weight loss was only 1.1-8.0% for poly(lactic acid), and 0.8-6.8% for poly(butylene adipate-co-terephthalate), and other plastics are basically non-degradable. Of note, numerous microplastics were formed after PPDO degradation, with 441 ± 326 and 2103 ± 131 item/g plastic in the air and soil, respectively. In this study, a novel permeable electrochemical reactive barrier (PERB) was fabricated for underground water remediation using a TiO2/graphite composite (TiO2/C) as the heterogeneous electrocatalyst. The as-prepared FeSx particles appeared as a honeycomb-like and highly porous structure. The microwave-assisted FeSx reduction process was able to rapidly reduce the TCLP-based reachability of Cr(VI) from 391.8 to 2.6 mg·L-1. The dosage of FeSx, S/Fe molar ratio, initial moisture content, microwave power, and irradiation time can all affect the treatment effectiveness. After 500 days curing under atmospheric conditions, the TCLP-leached concentration of Cr remained below the regulatory limit of 5 mg·L-1, while other treatments failed to meet the goal. Sx2- or S2- served as the primary electron donors, whereas Fe facilitated the microwave absorption and the formation of the stable final product of FeCr2O4. S and Fe are mostly precipitated in soil. The microwave-assisted FeSx reduction was shown to be an effective approach to rapidly reduce the leachability of Cr(VI) in contaminated soil, especially in heavily contaminated soil.We investigated the performance and encapsulation mechanisms of novel biochar-supported nanoscale zero-valent iron (nZVI-BC) used for the remediation of soil co-contaminated with arsenic (As), cadmium (Cd), and lead (Pb) via incubation and column experiments. Compared with the control, 0.50% of nZVI-BC significantly decreased the leakage of As, Cd, and Pb by 97.94-98.45%, 42.86-81.12%, and 82.14-92.49%, respectively. In addition, 0.50% of nZVI-BC could transform the fraction of unstable heavy metals into a stable form, which substantially decreased the availability, leachability, and bioaccessibility of the heavy metals and hence greatly reduced the human health exposure risk. Column experiments showed that 0.50% of nZVI-BC effectively restrained the leaching of As, Cd, and Pb by 95.60-99.84%, 70.82-84.18%, and 91.68-99.81%, respectively. The predominant encapsulation mechanisms of nZVI-BC included complexation, precipitation/co-precipitation, reduction, and the formation of ternary surface complexes. Based on these insights, we can devise new strategies for the remediation of soil co-contaminated with As, Cd, and Pb.Much attention has been paid to developing methods capable of synchronous removal of pollutants from swine wastewater. Due to the natural symbiotic interactions between microalgae and bacteria, the microalgae-bacteria symbiosis (ABS) system has been found to have potential for treating wastewater. However, the corresponding biological mechanisms in the ABS system and the role of dynamic microbial community evolution in pollutant removal systems remain poorly understood. Therefore, we investigate the potential of an ABS system for pollutant removal applications and analyze the bacterial consortium symbiotically combined with Chlorella sp. MA1 and Coelastrella sp. KE4. The NH4+-N and PO43--P removal efficiencies were significantly increased from 12.79% to 99.52% and 35.66% to 96.06% due to biotic interactions between the microalgae and bacteria. The abundance of bacterial taxa and genes related to oxidative stress, cell growth and nitrogen transfer were found to increase in response to photosynthesis, respiration and NH4+-N uptake.
