Hepatitis B virus (HBV) infection and its related liver diseases are important health problems worldwide, particularly in the Asia-Pacific region. The alloys were prepared by vacuum induction melting, followed by centrifugal casting. The as-cast samples were subjected to hot isostatic pressing and heat treatment consisting of solution annealing in β (Ti-based solid solution) phase field, cooling at a constant rate and stabilization annealing. The microstructure of the alloys consists of α2 (Ti3Al) + γ (TiAl) lamellar grains, single γ phase, coarse Ti2AlC particles, and irregular shaped α2 phase. The increase in the content of C at the expense of decreasing Al in the studied alloys affects solid-state phase transformation temperatures and leads to a decrease in size of grains and primary Ti2AlC particles, increase in the volume fraction of reinforcing carbide particles, decrease in the volume fraction of lamellar colonies, and widening of the grain boundaries. Long-term ageing at 800 °C has no effect on the grain size but leads to the formation of Ti4Al3Nb particles and increase in interlamellar spacing. The Vickers hardness, microhardness of lamellar grains, indentation nanohardness, and elastic modulus of the boundary γ phase decrease during ageing. The Ti-42.6Al-8.7Nb-0.3Ta-2.0C alloy shows improved creep resistance compared to that of Ti-41.0Al-8.7Nb-0.3Ta-3.6C and some reference TiAl-based alloys at a temperature of 800 °C and applied stress of 200 MPa.Genomic instability is a hallmark of cancer cells which results in excessive DNA damage. To counteract this, cells have evolved a tightly regulated DNA damage response (DDR) to rapidly sense DNA damage and promote its repair whilst halting cell cycle progression. TRIM24, TRIM28, TRIM33 and TRIM66 make up the transcriptional intermediary factor 1 (TIF1) family of chromatin-binding proteins, a subfamily of the large tripartite motif (TRIM) family of E3 ligases. All four TIF1 proteins are aberrantly expressed across numerous cancer types, and increasing evidence suggests that TIF1 family members can function to maintain genome stability by mediating chromatin-based responses to DNA damage. This review provides an overview of the TIF1 family in cancer, focusing on their roles in DNA repair, chromatin regulation and cell cycle regulation.Alzheimer's disease (AD) is the most common type of dementia and is currently incurable, and mitogen-activated protein kinase (MAPK) p38 is implicated in the pathogenesis of AD. p38 MAPK inhibition is considered a promising strategy against AD, but there are no safe inhibitors capable of penetrating the blood-brain barrier. Earlier, we have shown that mitochondria-targeted antioxidant plastoquinonyl-decyltriphenylphosphonium (SkQ1) at nanomolar concentrations can prevent, slow down, or partially alleviate AD-like pathology in accelerated-senescence OXYS rats. Here we confirmed that dietary supplementation with SkQ1 during active progression of AD-like pathology in OXYS rats (aged 12-18 months) suppresses AD-like pathology progression, and for the first time, we showed that its effects are associated with suppression of p38 MAPK signaling pathway (MAPKsp) activity. Transcriptome analysis, western blotting, and immunofluorescent staining revealed that SkQ1 suppresses p38 MAPKsp activity in the hippocampus at the level of expression of genes involved in the p38 MAPKsp and reduces the phosphorylation of intermediate kinases (p38 MAPK and MK2) and a downstream protein (αB-crystallin). Thus, the anti-AD effects of SkQ1 are associated with improvement in the functioning of relevant signaling pathways and intracellular processes, thus making it a promising therapeutic agent for human AD.Researchers typically assume constant fur and hair growth for primates, but the few studies that have investigated growth explicitly suggest this may not be the case. link3 Instead, growth may vary considerably among individuals and across seasons. One might expect this variability to be most pronounced for species that have seasonally variable activity patterns (e.g., Madagascar's Cheiorogaleidae). In particular, dwarf lemurs (Cheirogaleus spp.) undergo considerable changes in their daily activity levels (torpor) in the austral fall, when nights get shorter. I monitored regrowth of shaved fur patches for eight adult captive fat-tailed dwarf lemurs (Cheirogaleus medius) and gray mouse lemurs (Microcebusmurinus) on a bi-weekly basis for 21 months in total. Regrowth varied considerably both within and among individuals. Overall, fur regrew in spurts and was faster for mouse lemurs (0-14 to 215-229 days) than dwarf lemurs (27-40 to 313-327 days). There were significant differences between species and an obvious influence of season for dwarf lemurs, but no clear influence of shave location, age, or sex. Similar trends have been previously reported for captive lemurids, suggesting that seasonal fur growth may be widespread across Lemuroidea. Researchers are cautioned against using primate fur or hair to investigate variables confounded by seasonality (such as diet and body condition) until patterns of growth are better understood.Actigraphy, an objective measure of motor activity, reliably indexes increased movement levels in attention-deficit/hyperactivity disorder (ADHD) and may be useful for diagnosis and treatment-monitoring. However, actigraphy has not been examined in complex neurodevelopmental conditions. This study used actigraphy to objectively measure movement levels in individuals with a complex neurodevelopmental genetic disorder, tuberous sclerosis (TSC). Thirty participants with TSC (11-21 years, 20 females, IQ = 35-108) underwent brief (approximately 1 h) daytime actigraph assessment during two settings movie viewing and cognitive testing. Multiple linear regressions were used to test associations between movement measurements and parent-rated ADHD symptoms. Correlations were used to examine associations between actigraph measures and parent-rated ADHD symptoms and other characteristics of TSC (symptoms of autism spectrum disorder (ASD), intellectual ability (IQ), epilepsy severity, cortical tuber count). Higher movement levels during movies were associated with higher parent-rated ADHD symptoms. Higher ADHD symptoms and actigraph-measured movement levels during movies were positively associated with ASD symptoms and negatively associated with IQ. Inter-individual variability of movement during movies was not associated with parent-rated hyperactivity or IQ but was negatively associated with ASD symptoms. There were no associations with tuber count or epilepsy. Our findings suggest that actigraph-measured movement provides a useful correlate of ADHD in TSC.Liquid biopsy-based methods to test biomarkers (e.g., serum proteins and extracellular vesicles) may help to monitor brain tumors. In this proteomics-based study, we aimed to identify a characteristic protein fingerprint associated with central nervous system (CNS) tumors. Overall, 96 human serum samples were obtained from four patient groups, namely glioblastoma multiforme (GBM), non-small-cell lung cancer brain metastasis (BM), meningioma (M) and lumbar disc hernia patients (CTRL). After the isolation and characterization of small extracellular vesicles (sEVs) by nanoparticle tracking analysis (NTA) and atomic force microscopy (AFM), liquid chromatography -mass spectrometry (LC-MS) was performed on two different sample types (whole serum and serum sEVs). Statistical analyses (ratio, Cohen's d, receiver operating characteristic; ROC) were carried out to compare patient groups. To recognize differences between the two sample types, pairwise comparisons (Welch's test) and ingenuity pathway analysis (IPA) were performed. According to our knowledge, this is the first study that compares the proteome of whole serum and serum-derived sEVs. From the 311 proteins identified, 10 whole serum proteins and 17 sEV proteins showed the highest intergroup differences. Sixty-five proteins were significantly enriched in sEV samples, while 129 proteins were significantly depleted compared to whole serum. Based on principal component analysis (PCA) analyses, sEVs are more suitable to discriminate between the patient groups. Our results support that sEVs have greater potential to monitor CNS tumors, than whole serum.Celastrol (CL), a compound isolated from Tripterygium wilfordii, possesses various bioactivities such as antitumor, anti-inflammatory and anti-obesity effects. In previous studies, we developed CL-encapsulated silk fibroin nanoparticles (CL-SFNP) with satisfactory formulation properties and in vitro cancer cytotoxicity effect. For further in vivo oral bioavailability evaluation, in this study, a simple and reliable LC-MS/MS method was optimized and validated to determine CL concentration in rat plasma. The separation of CL was performed on a C18 column (150 by 2 mm, 5 µm) following sample preparation using liquid-liquid extraction with the optimized extraction solvent of tert-butyl methylether. The assay exhibited a good linearity in the concentration range of 0.5-500 ng/mL with the lower limit of quantification (LLOQ) of 0.5 ng/mL. The method was validated to meet the requirements for bioassay with accuracy of 91.1-110.0%, precision (RSD%) less than 9.1%, extraction recovery of 63.5-74.7% and matrix effect of 87.3-101.2%. The developed method was successfully applied to the oral bioavailability evaluation of CL-SFNP. The pharmacokinetic results indicated the AUC0-∞ values of CL were both significantly (p less then 0.05) higher than those for pure CL after intravenous (IV) or oral (PO) administration of equivalent CL in rats. The oral absolute bioavailability (F, %) of CL significantly (p less then 0.05) increased from 3.14% for pure CL to 7.56% for CL-SFNP after dosage normalization. This study provides valuable information for future CL product development.New molybdenum trioxide-incorporated ZnO materials were prepared through the electrospinning method and then calcination at 500 °C, for 2 h. The obtained electrospun ZnOMoO3 hybrid materials were characterized by X-ray diffraction, scanning and transmission electron microscopies, ultraviolet (UV)-diffuse reflectance, UV-visible (UV-vis) absorption, and photoluminescence techniques. It was observed that the presence of MoO3 as loading material in pure ZnO matrix induces a small blue shift in the absorption band maxima (from 382 to 371 nm) and the emission peaks are shifted to shorter wavelengths, as compared to pure ZnO. Also, a slight decrease in the optical band gap energy of ZnOMoO3 was registered after MoO3 incorporation. The photocatalytic performance of pure ZnO and ZnOMoO3 was assessed in the degradation of rhodamine B (RhB) dye with an initial concentration of 5 mg/L, under visible light irradiation. A doubling of the degradation efficiency of the ZnOMoO3 sample (3.26% of the atomic molar ratio of Mo/Zn) as compared to pure ZnO was obtained.
