BACKGROUND Screening borderline Spitz tumors with p16 immunohistochemistry has known utility. The applicability to other melanocytic neoplasms is not well defined. METHODS Cases (N = 104) of blue, cellular blue, epithelioid blue, congenital pattern, deep penetrating, desmoplastic, desmoplastic Spitz, acral, "epithelioid" nevi, nevoid melanoma, melanoma with a precursor nevus, and non-nevoid melanoma with Breslow thickness > 0.5 mm were stained for p16. RESULTS Lesions showed either a single uniform pattern of expression (single/homogeneous pattern positive, checkerboard, rare, or lost) or multiple regionally distributed patterns (multiple/heterogeneous pattern). Most (78%, n = 81) demonstrated single pattern expression. Within single pattern cases, total loss was restricted to melanoma (7/81/9%). Multiple patterns were more common in melanoma (12/23, 52%). Within multiple pattern (22%, n = 23) lesions, those with a total loss component (7/23; 30%) were malignant. Total p16 loss (diffuse or regional) was not seen in a subset of nevoid melanomas (1/8; 12.5%), melanomas arising in nevi (2/6; 33%), and non-nevoid melanomas (6/9; 66%). Total p16 loss (single pattern or part of multiple patterns), captured 61% (14/23) of melanomas and no nevi. CONCLUSION p16 IHC may be useful in dermal based melanocytic lesions. Total p16 loss is seen only in melanoma. Multiple pattern expression should prompt careful evaluation. This article is protected by copyright. All rights reserved. However, the chemical composition and the content accumulation of main secondary metabolites of D. moldavica in different natural environments were remaining unclear. The study aimed to conduct a qualitative and quantitative analysis of main secondary metabolites to explore the quality variation of D. moldavica in market. The evaluation of space-time accumulation of main secondary metabolites in D. moldavica was carried out from different growth period and geographical location. A total of 35 ingredients were detected out and 24 identified, including 21 flavonoids, 2 phenolic acids, and 1 coumarin by UPLC-Qtof-MS method. Furthermore, a simple and convenient HPLC method was successfully developed for the simultaneous determination of lutelin-7-O-glucuronide and tilianin and rosmarinic acid in D. moldavica. The results of space-time accumulation analysis showed the distinct variation of secondary metabolites of D. moldavica with the growth period and geographical location. Finally, the current study provided a meaningful and useful approach for comprehensively evaluating the quality of D. moldavica. This article is protected by copyright. All rights reserved.Currently, Rhizoma Dioscoreae Makino (RDM) has obtained a fine effect in treating gouty arthritis (GA) and hyperuricacidemia, especially in promoting uric acid excretion and reducing the inflammatory reaction. Bioactive constituents in RDM are mainly steroidal saponins such as dioscin, trillin, protodioscin and protogracillin. However, the mechanism of its anti-GA is still unclear, owing to the complex pathological and physiological characteristics of GA, and integration of RDM with multiple components, multiple targets and multiple pathways. Herein, GA rat model was induced with monosodium urate (MSU), and RDM could reduce inflammation of rat synovium tissue. Through metabolomics analysis, 35 potential biomarkers with significant changes involved in the pathogenesis of GA induced by MSU were identified, and their perturbations could be restored after RDM-treatment. It was observed that the most correlated pathways involved in d-galactose, d-mannose, d-glucose, myoinositol, PC (160/160), LysoPC (150) , PA (181(9Z)/181(11Z)) and glutathione induced by MSU were as following Galactose metabolism, Inositol phosphate metabolism, Glycerophospholipid metabolism and Glutathione metabolism, and the derivations of all those biomarkers could be regulated by RDM treatment.
