Microbiota influenced quality formation of ripened Pu-erh tea. To understand the effect of each tea-derived fungal strain, tea-leaves were fermented by Aspergillus pallidofulvus PT-3 (ApaPT), Aspergillus sesamicola PT-4 (AsePT) and Penicillium manginii PT-5 (PmaPT), respectively. 14 Phenolic compounds, 3 purine alkaloids, 19 free amino acids and γ-aminobutyric acid contents were determined by HPLC and amino acid analyzer analysis. Additionally, UHPLC-Q-TOF/MS method was developed for LC-MS metabolomics analysis. Multivariate statistical analyses, such as PCA and HCA, exhibited that the chemical profile of PmaPT fermentation was similar to biocidal treatment, but had significant differences with ApaPT and AsePT fermentation. The differentiated metabolites (VIP > 1, p 1.50 or less then 0.66) and one-way ANOVA revealed the impact of three fungal strains in tea-leaves fermentation. APaPT and AsePT contributed to biosynthesis of gallic acid and several flavonoids, such as kaempferol, quercetin and myricetin in the metabolism of phenolic compounds. This study aimed to assess the feasibility of identifying multiple chemical constituents in matcha using visible-near infrared hyperspectral imaging (VNIR-HSI) technology. Regions of interest (ROIs) were first defined in order to calculate the representative mean spectrum of each sample. Subsequently, the standard normal variate (SNV) method was applied to correct the characteristic spectra. Competitive adaptive reweighted sampling (CARS) and bootstrapping soft shrinkage (BOSS) were used to optimize the models. They were built based on partial least squares (PLS), creating two models referred to as CARS-PLS and BOSS-PLS. The BOSS-PLS models achieved best predictive accuracy, with coefficients of determination predicted to be 0.8077 for caffeine, 0.7098 for tea polyphenols (TPs), 0.7942 for free amino acids (FAAs), 0.8314 for the ratio of TPs to FAAs, and 0.8473 for chlorophyll. These findings highlight the potential of VNIR-HSI technology as a rapid and nondestructive alternative for simultaneous quantification of chemical constituents in matcha. In this study, the ochratoxin A (OTA) interaction with some metal cations exhibits a quenching effect of the Cu2+, Co2+, and Ni2+ in the OTA intrinsic fluorescence intensity. find more The property of the OTA fluorescence change in complexation with the Cu2+ has been applied in developing a label-free and selective fluorimetric sensor for fast detection of the OTA trace amounts in wheat flour samples. The achieved color difference map (CDM) based on the spectral profile provides the technical support for the rapid visual sensing of OTA in the wheat flour samples. The feasibility of the hybrid fluorimetric sensor and visual OTA detection in wheat flour samples has been confirmed with the high-performance liquid chromatography (HPLC) method as a standard method. The calculated LOD (0.4 ng g-1) and LOQ (1.2 ng g-1) values of the proposed method are much lower than the maximum permissible limit of OTA value reported by the European Union (5 ng g-1).We report allele frequencies and standard population genetics parameters for 23 forensic autosomal STR loci tested among White British population. In addition, we use generated allele frequency data to compare the extent of genetic variation with 19 other European populations.Hypercholesterolemia has been linked to neurodegenerative disease development. Previously others and we demonstrated that high levels of plasma cholesterol-induced memory impairments and depressive-like behavior in mice. More recently, some evidence reported that a hypercholesterolemic diet led to motor alterations in rodents. Peripheral inflammation, blood-brain barrier (BBB) dysfunction, and neuroinflammation seem to be the connective factors between hypercholesterolemia and brain disorders. Herein, we aimed to investigate whether treatment with gold nanoparticles (GNPs) can prevent the inflammation, BBB disruption, and behavioral changes related to neurodegenerative diseases and depression, induced by hypercholesterolemic diet intake in mice. link2 Adult Swiss mice were fed a standard or a high cholesterol diet for eight weeks and concomitantly treated with either vehicle or GNPs by the oral route. At the end of treatments, mice were subjected to behavioral tests. After that, the blood, liver, and brain structures were collected for biochemical analysis. find more The high cholesterol diet-induced an increase in the plasma cholesterol levels and body weight of mice, which were not modified by GNPs treatment. Hypercholesterolemia was associated with enhanced liver tumor necrosis factor- α (TNF-α), BBB dysfunction in the hippocampus and olfactory bulb, memory impairment, cataleptic posture, and depressive-like behavior. Notably, GNPs administration attenuated liver inflammation, BBB dysfunction, and improved behavioral and memory deficits in hypercholesterolemic mice. Also, GNPs increased mitochondrial complex I activity in the prefrontal cortex of mice. It is worth highlight that GNPs' administration did not cause toxic effects in the liver and kidney of mice. find more Overall, our results indicated that GNPs treatment potentially mitigated peripheral, brain, and memory impairments related to hypercholesterolemia.Poly (amidoamine) (PAMAM) with 3rd and 5th generation was covalently grafted as the contact active biocidal agent on the surface of polyethylene terephthalate (PET) with the help of UV induced carbene chemistry (PAMAM-g-PET). The graft density and the surface roughness were controlled by turning UV irradiation time and the PAMAM generation. The PAMAM graft monolayer was characterized via the contact angle, XPS, nanoIR, SEM and AFM. The antibacterial ability of PAMAM-g-PET was evaluated ex-vivo with the help of laser scanning confocal microscope (CLSM), and the results indicated that the decorated PET was able to kill both S. aureus and E. coli in the aqueous environment. Increasing the surface graft concentration and using the dendrimer with higher generation enhanced the lethality towards the bacterial. link2 The decorated film was still able to kill the contact bacterial strain when the cationic primary amine groups were shielded by acetyl chloride, however, the bacterial in the suspension was hardly affected in this case. The un-selectivity and instantaneity of carbene chemistry endowed this grafting strategy the potential to be extended to other organic substances. The present work designed to improve the skin delivery of arbutin niosome (arbusome) was prepared via an ultrasonic technique. The arbusome formulations were optimized by investigating the effects of the cholesterolsurfactants ratio. To characterize the morphology and solid-state of arbutin in arbusome, differential scanning calorimetry, photon correlation spectroscopy, powder x-ray diffractometer, scanning electron microscopy (SEM), and attenuated total reflectance-Fourier transform infrared spectroscopy were utilized. link3 The findings indicated that adding cholesterol incremented the arbusome's particle size. link2 Further studies proved that the zeta potential and the size of nanoparticles can be modulated by the alterations in the ratio of cholesterol surfactant. When the cholesterol concentration was high in the formulation, the highest entrapment efficiency was found to be approximately 44 %. Solid-state analysis showed that arbutin in the niosome was in the amorphous state. The skin permeation test indicated the greater quantities of the arbutin in skin layers and the receptor chamber for arbusome gel compared to arbutin simple gel. Furthermore, in vitro cytotoxicity test indicated no cytotoxicity for the improved formulation of niosome containing arbutin. link3 The cell viability (HFF cell line) for niosomal formulation of arbutin was reported to be about 86 %. The formulations were examined in terms of skin irritation on Wistar rats, and non-irritancy of arbutin niosomal gels was indicated. The findings of this work discovered that the manufactured arbusome could be utilized as possible nano-vehicle for the arbutin topical delivery and might open new approaches for the treatment of hyperpigmentation complaints. This study was aimed towards the development of a novel microfluidic approach for the preparation of (co)polymeric and hybrid nanoparticles (NPs) composed of (co)polymers/tannic acid (TA) in the microfluidic flow-focusing glass-capillary device. The MiliQ water was used as water phase, whereas the organic phase was composed of poly(ε-caprolactone) (PCL) and poly(trimethylene carbonate) (PTMC) homopolymers and (co)polymers with different proportion of comonomers which were prepared via enzymatic polymerization that allows avoiding the usage of potentially toxic catalyst. To prepare hybrid NPs, TA was additionally added to the organic phase. Subsequently, as a result of mixing between these distinct phases in microfluidic channels, the nanoprecipitation in the form of spherical NPs occurs. The size of NPs was tuned over the range of 140-230 nm by controlling phase flow rates and the composition of NPs. Moreover, the release studies of the encapsulated anticancer drug doxorubicin (DOX) demonstrated that the drug release is greatly influenced by the (co)polymers composition, their molecular weight, NPs size, and the presence of TA. link3 The antitumor activities of the (co)polymeric and hybrid NPs toward breast cancer cells (MCF-7) were tested in vitro. Among all tested formulation, the NPs composed of PCL/TA most efficiently inhibit the cell proliferation of MCF-7 cells, most importantly, their efficiency was higher than free DOX. The proposed strategy may provide an efficient alternative for the construction of nanocarriers with great potential in anticancer therapy. The disease burden due to nontuberculous mycobacteria is growing worldwide, consequently to improved diagnostic abilities and an increase in the individuals at risk. Uncertainties exist about the right moment on which start treatment. We investigated the clinical features associated with starting an antimicrobial treatment in patients with nontuberculous mycobacterial lung disease among Italian physicians involved in the field. We found that in real life predictors of starting antimicrobial treatment are quite adherent to international guidelines, with some uncertainties regarding the implication of immunosuppressive drugs.
The temporal dynamics of cavity formation in patients with the noncavitary nodular bronchiectatic (NC-NB) form of Mycobacterium avium complex pulmonary disease (MAC-PD) have not yet been well described. We aimed to investigate the development of new cavities in the NC-NB form of MAC-PD.

Of the patients diagnosed with NC-NB-type MAC-PD between 2002 and 2013 and followed-up until July 2018at a tertiary referral center in South Korea, we identified 589 patients who underwent follow-up chest computed tomography at least once after the diagnosis and retrospectively analysed their medical records.

The patients' mean age was 62.0 years, 64.7% were women. During the median follow-up of 3.8 years (interquartile range [IQR] 1.7-5.9), new cavity formation was noted in 51 (8.7%) patients. The median interval between the diagnosis of NC-NB MAC-PD and cavity formation was 3.7 years (IQR 1.8-5.4), with a constant occurrence over time. The Cox regression analysis showed that a history of pulmonary tuberculosis (adjusted hazard ratio [HR] 1.
