The extracted PHAs were revealed to be polyhydroxybutyrate (PHB) by characterization methods of Fourier-transform infrared spectroscopy (FTIR) and differential scanning calorimetry (DSC). The obtained results proved the possibility of using VFAs from acidogenic fermentation of food waste as a cheap substrate to reduce the cost of PHAs production.The purpose of this study was to investigate the impact and mechanism of microRNA miR-126 on brain injury induced by blood-brain barrier (BBB) damage in septic rats. We used cecal ligation and perforation (CLP) to create a rat model of sepsis. The experimental rats were randomly divided into Control group, CLP group, CLP + miR-NC group, CLP + miR-126 group and CLP + miR-126 + NF-κB pathway agonist (PMA) group. MiR-126 expressed in the brain tissue of CLP rats was down-regulated by qRT-PCR. Upregulation of miR-126 in CLP rats could improve brain injury and BBB marker protein level, reduce brain water content, Evans blue extravasation, inflammation, and excessive oxidative stress. This could also result in an inhibition of NF-κB signaling pathway activity. In conclusion, miR-126 overexpression can prevent brain injury caused by BBB damage via the inhibition of NF-κB signaling pathway activity.Our study seeks to obtain data which help to assess the impacts and related mechanisms of microRNA miR-509-3p in hepatocellular carcinoma (HCC). We found that the expression of miR-509-3p was down-regulated and Twist was up-regulated in HCC tissues and cell lines (HepG2, HCCLM3, Bel7402, and SMMC7721) compared with the adjacent normal tissues and normal human hepatocyte (L02). Moreover, cell proliferation, invasion, migration and epithelial-mesenchymal transition (EMT) in HepG2 and HCCLM3 cells were appeared to be markedly suppressed by overexpressed miR-509-3p. Overexpression of miR-509-3p also performed inhibition of the growth and metastasis in vivo. In addition, miR-509-3p could target and inhibit Twist expression, and it could further reverse the tumor promotion by Twist in HCC. All in all, miR-509-3p overexpression causes inhibition of the proliferation, migration, invasion and EMT of HCC cells by negatively regulating Twist, thereby suppressing HCC development and metastasis.To overcome the problems of frequent leakage accidents during oil exploitation, a superhydrophobic and superoleophilic porous MS/PDA/DT sponge was successfully prepared via mild solvent evaporation method, and a polydopamine assisted surface coating of 1-dodecanethiol (DT) on a melamine sponge (MS) substrate. Surface structure and performance of the MS/PDA/DT sponge were characterized by Scanning Electron Microscope (SEM), Fourier transform infrared spectrometer (FTIR), and Video Optical Contact Angle (CA) metre. The results showed that the as-prepared MS/PDA/DT sponge has a high-water contact angle (WCA) of 147.2°, which is probably attributed to both the rough surface derived from in situ growth and the low surface energy due to grafting of hydrophobic 1-dodecanethiol. The durability of the as-constructed MS/PDA/DT sponge was studied by repeated abrasion tests. After 50 abrasion cycles, the superhydrophobicity of the MS/PDA/DT sponge good mechanical durability. The MS/PDA/DT sponge can effectively absorb oil with an absorption capacity of up to 24 times its weight. The superhydrophobic and superoleophilic MS/PDA/DT sponge has the potential as a promising adsorbent for oil/water separation.The buckling behavior of latticed columns had been widely investigated based on the theory of Euler, Engesser and Timoshenko shear beam. Although these methods had been formulated and proved to be accurate in case of special assumptions, the influences of lacing bars on the buckling behavior of latticed columns were unclear. find more This paper modeled a general four-legged latticed column to study the influence of the cross-section characteristics of lacing bars along with their imperfections on the buckling capacity of latticed columns. Three loading conditions and four geometric imperfect models were built to testify the performance of lacing bars. To calculate the buckling load of latticed columns with imperfections accurately, advanced nonlinear analytical procedures using Newton-Raphson incremental-iterative method (ANAP-NR) and Risk arc-length incremental-iterative method (ANAP-Risk) were developed, and then validated by FE software ABAQUS. The current data in the paper show the maximum variation on the critical buckling load of latticed columns, caused by the cross-section area, the bending moment of inertia outer lacing plane, and the imperfections of lacing bars, could reach 68%, 30%, and 25%. The analytical results indicate the great importance of lacing bars on the buckling capacity of latticed columns.Platelet function assays and global haemostasis assays are essential in diagnosing bleeding tendencies, with light transmission aggregometry (LTA) as golden standard. The Multiple Electrode Aggregation (Multiplate), platelet function assay (PFA) and rotational thromboelastometry (ROTEM) are mostly used as whole-blood screening tests. Currently, patients have to travel to specialized laboratories to undergo these tests, since specific expertise is required. Pre-analytical variables, like storage time and temperature during transport, are still considered to be the most vulnerable part of the process and may lead to discrepancies in the test results. We aim to give a first impression on the stability of blood samples from healthy volunteers during storage and investigate the effect of storage time (1, 3, 6 and 24 hours) and temperature (4°C, room temperature and 37°C) on the Multiplate, PFA, ROTEM and LTA test results. Our data indicated that, for the PFA, whole blood can be stored for 3 hours at room temperature. Whole blood used for the Multiplate and ROTEM can be stored for 6 hours of storage. For LTA, PRP and whole blood were stable up to 3 hours at 4°C or room temperature and 6 hours at room temperature, respectively.Myocardial cell injury caused by myocardial ischemia and reperfusion is one of the main causes of the occurrence and development of heart disease. Recent study has shown that inducing mitophagy of cardiomyocytes is a crucial method to alleviate ischemia-reperfusion injury. While, Polo-like kinase 1 (PLK1) can induce the mitophagy of breast cancer cells. Moreover, PLK1 was able to promote the expression of p-AMPK and FUNDC1, which are the protective factors for myocardium. Therefore, the mouse model of ischemia/reperfusion was established and the effect of PLK1 on ischemia reperfusion induced myocardial damage was investigated. The PLK1 was overexpressed in H9c2 cells and rat model of ischemia/reperfusion. Ischemia reperfusion inhibited the expression of PLK1. While overexpression of PLK1 relieved the myocardial infarction and myocardium apoptosis through inducing mitophagy in rats model of ischemia reperfusion. In vitro, the H9c2 cells overexpressing the PLK1 were treated with the hypoxia and reoxygenation and the apoptosis, survival rate and expression of mitophagy-related proteins of H9c2 cells were detected using the flow cytometry, CCK-8 assay and western blotting. The results reveled that overexpression of PLK1 alleviated the hypoxia and reoxygenation induced apoptosis of H9c2 cells and promoted the expression of mitophagy-related proteins. In addition, enhanced PLK1 expression promoted the expression of p-AMPK and FUNDC1 in H9c2 cells. However, the inhibition of FUNDC1 abolished the positive effect of PLK1 on H9c2 cells mentioned above. In conclusion, PLK1 alleviated the ischemia reperfusion induced myocardial damage by inducing the mitophagy in a p-AMPK/FUNDC1 signaling dependent pathway.Spent drilling mud (DM), a co-product during horizontal directional drilling operations, needs proper disposal to comply with social and environmental sustainability awareness, with one option being its cost-effective reuse as a soil amendment. The possibility was investigated of DM applications to improve the physical quality of loamy sand soil. The soil was treated with increasing DM contents in a laboratory at volumetric DM-to-soil ratios of 0100 (DM0, no DM), 2575 (DM25), 5050 (DM50), 7525 (DM75), and 1000 (DM100, no soil). The mixtures were analyzed for water retention, available water capacity, water storage capacity, S-index, soil aggregate stability (SAS), and chemical quality based on pH, effective electrical conductivity (ECe), sodium adsorption ratio (SAR), and exchangeable sodium percentage (ESP). The optimal ratio of DM and the control (DM0) in the batch experiment were selected for application to three species of forest plant seedlings in the field. The results showed that DM positively influencland requires the installation capacity, transmission, and distribution pipelines. Horizontal directional drilling (HDD) operations are generally used to install underground pipelines, generating large amounts of drilling mud rich in sodium bentonite as a co-product. At present, drilling mud is becoming a more serious solid waste and has been raised in proper management. Tumor lysis syndrome is an oncologic emergency characterized by hyperuricemia. Previous studies have demonstrated that a fixed-dose strategy of rasburicase is as effective as the FDA approved weight-based dose. Albany Medical Center employs rasburicase 1.5 mg in patients with a uric acid (UA) between 8 and 12 mg/dL and 3 mg for UA above12 mg/dL.We aimed to evaluate the UA lowering effectiveness and provider adherence to the institutional protocol, as well as the cost-efficiency of this dosing strategy.
This is a single center, retrospective, cohort study. The electronic medical record was used to identify patients receiving rasburicase and to collect baseline demographic and laboratory data. The fixed-dose strategies of rasburicase 1.5 mg and 3 mg were compared in their degree of UA reduction and clinical outcomes. Cost-savings of fixed-dosing was compared to the FDA-approved weight-based dose.
Mean UA reduction in the 1.5 mg group (n = 49) from baseline to 24 hours was 2.88 ± 0.88 mg/dL (p < 0.0001) the FDA-approved weight-based dose.
