Pathologic retinal neovascularization is a potentially blinding consequence seen in many common diseases including diabetic retinopathy, retinopathy of prematurity, and retinal vaso-occlusive diseases. This study investigates epithelial membrane protein 2 (EMP2) and its role as a possible modulator of angiogenesis in human retinal pigment epithelium (RPE) under hypoxic conditions. To study its effects, the RPE cell line ARPE-19 was genetically modified to either overexpress EMP2 or knock down its levels, and RNA sequencing and western blot analysis was performed to confirm the changes in expression at the RNA and protein level, respectively. Protein expression was evaluated under both normoxic conditions or hypoxic stress. Capillary tube formation assays with human umbilical vein endothelial cells (HUVEC) were used to evaluate functional responses. EMP2 expression was found to positively correlate with expression of pro-angiogenic factors HIF1α and VEGF at both mRNA and protein levels under hypoxic conditions. Mechanistically, EMP2 stabilized HIF1α expression through downregulation of von Hippel Lindau protein (pVHL). EMP2 mediated changes in ARPE-19 cells were also found to alter the secretion of a paracrine factor(s) in conditioned media that can regulate HUVEC migration and capillary tube formation in in vitro functional angiogenesis assays. This study identifies EMP2 as a potential mediator of angiogenesis in a human RPE cell line. EMP2 levels positively correlate with pro-angiogenic mediators HIF1α and VEGF, and mechanistically, EMP2 regulates HIF1α through downregulation of pVHL. This study supports further investigation of EMP2 as a promising novel target for therapeutic treatment of pathologic neovascularization in the retina.This study compared the ability of Nanostructured Lipid Carrier (NLC), Solid Lipid Nanoparticles (SLN), and Cream systems in delivering para Methoxycinnamic Acid (PMCA) to the dermis layer of the skin. Wistar rats were used as research subjects. NLC and SLN were made by applying the high shear homogenization method. Nile red was used as a penetration indicator based on its fluorescence. The interaction between fluorescence labeled NLC, SLN, or Cream and rat skin was visualized by fluorescence microscopy. Observations were made after 2 and 4.5 h of smearing the test sample. From the observations, it was known that the system/lipid base could penetrate the stratum corneum for delivering drugs. Penetration speed differs among systems as does the number of PMCAs that can be delivered. In this study, it can be concluded that the NLC system is able to deliver PMCA more quickly and in greater quantities to the dermis than SLN and Cream.Mechanical precision corn seed-metering planter has a compact structure, missed and repeated seeding advantages during high-speed operation. In this regard, the current research study focuses on the development of a corn seed planter that features an inclined seed-metering device. The spatial layout of the seed-metering device is optimized to change the seed-filling mode to meet the needs of high-speed operation. Firstly, the mechanical characteristics and seeds in the metering device chamber were analyzed, and then the seed-filling stress model was established. Secondly, a mechanical model for corn seed particles was developed for virtual simulation tests and numerical analysis using the discrete element method (DEM) and EDEM software. Moreover, a quadratic rotating orthogonal center combination test was implemented by setting the inclination angle of seed-metering device θ(A), machine ground speed v(B), and rotation speed of metering disc n(C) as the influence factors, with the missed seeding rate M and the seed-filling stress S as the evaluation indices. The results indicated that the most significant factors affecting the missed seeding rate, seed-filling stress, S, were the rotation speed of the metering disc (n) > machine ground speed (v) > inclination angle of the metering disc (θ) and inclination angle of the metering device (θ) > rotation speed of the metering disc (n) > machine ground speed (v), respectively. However, the field verification test shows that the optimized corn seed-metering planter achieved mean values of M = 4.33, Q(qualified seeding rate) = 92.83%, and R(repeated seeding rate) = 2.84%, with average relative errors of 1.17% compared to the simulation tests and the accuracy and effectiveness of the DEM simulation model was verified. Therefore, the developed corn seed-metering device meets the industry standards and operation requirements for precise corn sowing, and technical support can be given for future studies of similar precision seeding equipment.Microfabricated chip-edge microcantilevers are commonly used as surface probes, e.g. in near-field microscopy. Such probes normally function in the low-deflection regime, where their behaviour is very well understood and documented. In contrast, when microcantilevers are used for applications such as electrical testing probes, their deflection can be somewhat higher, taking them into the less well understood high-deflection regime of microelectromechanical systems (MEMS). Here, a scalable model for the relationship between the skate, overtravel, and resulting tip contact force in tilted triangular cantilevers-which are bending with high deflection and in contact with a flat surface-is presented. The model is tested experimentally using macroscopic triangular cantilevers-the experimental results agree well with the proposed model. The findings enable a practical solution for zero-skate in tapered MEMS probes to be suggested. It is hoped that the findings may be of use for probe engineers involved with on-wafer testing and designers of emerging MEMS micro cantilever-based probes.Placentas of obese women have low mitochondrial β-oxidation of fatty acids (FA) and accumulate lipids in late pregnancy. This creates a lipotoxic environment, impairing placental efficiency. We hypothesized that placental FA metabolism is impaired in women with obesity from early pregnancy. Maternal fasting triglyceride and insulin levels were measured in plasma collected at the time of procedure. The loss of the magnetic coupling system usually takes dominance in the whole system. This paper built the calculation model of magnetic field strength and coil loss, the proposed calculation model can effectively consider the mutual influence between the transmitter and receiver coil and accurately calculated the AC loss of WPT coils. Then, the effect of turn spacing on the AC resistance of coil is analyzed. It reveals that the proximity effect loss is greater when the coil is tightly wound, and the AC loss can be optimized by designing the turn spacing. Based on the above analysis, a double-layer coil method is proposed. This method can reduce the AC loss and improve the quality factor (Q) without changing the mutual inductance and footprint of coil at high frequency. The AC resistance of the double-layer coil method can be greatly reduced compared with the general method through simulation and experiment. The work efficiency of WPT system is increased by 4.3%, which verifies the accuracy and flexibility of theoretical analysis.We aimed to explore the relationship between cortisol response to psychosocial stress, mental distress, fatigue and health related quality of life (HRQoL) in individuals with coronary artery disease (CAD) after recent acute coronary syndrome (ACS). A cross-sectional study initially included 113 subjects (88% men, 53 ± 7 years) 1-3 weeks after ACS. Cortisol response was assessed by measuring salivary cortisol during Trier Social Stress Test. Mental distress was measured with Hospital Anxiety and Depression Scale, State-Trait Anxiety Inventory, and Type D Scale-14. Fatigue symptoms were evaluated using Multidimensional Fatigue Inventory 20-items, while HRQoL was assessed with 36-Item Short Form Medical Outcome Questionnaire. After conducting multivariable linear regression analyses, diminished cortisol response sampled after Public speech (T3-T1, + 15 min) was significantly associated with higher anxiety symptoms (β = -0.224; p = 0.035), while diminished cortisol response sampled after preparation time (T2-T1, + 10 min) was significantly linked with the presence of Type D personality (β = -0.290; p = 0.006; β = -0.282; p = 0.008 respectively), even after controlling for confounders (i.e., sex, age, education, New York Heart Association functional class, beta-blockers and baseline levels of cortisol measures). We found that mental distress, but not fatigue and HRQoL, was linked with blunted cortisol response during anticipation time of psychosocial stress, independently of potential covariates.Neural oscillations in the gamma frequency band have been identified as a fundament for synaptic plasticity dynamics and their alterations are central in various psychiatric and neurological conditions. Transcranial magnetic stimulation (TMS) and alternating electrical stimulation (tACS) may have a strong therapeutic potential by promoting gamma oscillations expression and plasticity. Here we applied intermittent theta-burst stimulation (iTBS), an established TMS protocol known to induce LTP-like cortical plasticity, simultaneously with transcranial alternating current stimulation (tACS) at either theta (θtACS) or gamma (γtACS) frequency on the dorsolateral prefrontal cortex (DLPFC). We used TMS in combination with electroencephalography (EEG) to evaluate changes in cortical activity on both left/right DLPFC and over the vertex. We found that simultaneous iTBS with γtACS but not with θtACS resulted in an enhancement of spectral gamma power, a trend in shift of individual peak frequency towards faster oscillations and an increase of local connectivity in the gamma band. Furthermore, the response to the neuromodulatory protocol, in terms of gamma oscillations and connectivity, were directly correlated with the initial level of cortical excitability. These results were specific to the DLPFC and confined locally to the site of stimulation, not being detectable in the contralateral DLPFC. We argue that the results described here could promote a new and effective method able to induce long-lasting changes in brain plasticity useful to be clinically applied to several psychiatric and neurological conditions.
