We identified the homozygous BNIP1 intronic variant c.84+3A>T in the apparently unrelated patients 1 and 2 with disproportionate short stature. Radiographs showed abnormalities affecting both the axial and appendicular skeleton and spondylo-epiphyseal dysplasia. We detected ~80% aberrantly spliced BNIP1 pre-mRNAs, reduced BNIP1 mRNA level to ~80%, and BNIP1 protein level reduction by ~50% in patient 1 compared to control fibroblasts. The BNIP1 ortholog in Drosophila, Sec20, regulates autophagy and lysosomal degradation. We assessed lysosome positioning and identified a decrease in lysosomes in the perinuclear region and an increase in the cell periphery in patient 1 cells. Immunofluorescence microscopy and immunoblotting demonstrated an increase in LC3B-positive structures and LC3B-II levels, respectively, in patient 1 fibroblasts under steady-state condition. Treatment of serum-starved fibroblasts with or without bafilomycin A1 identified significantly decreased autophagic flux in patient 1 cells. Our data suggest a block at the terminal stage of autolysosome formation and/or clearance in patient fibroblasts. BNIP1 together with RAB33B and VPS16, disease genes for Smith-McCort dysplasia 2 and a multisystem disorder with short stature, respectively, highlight the importance of autophagy in skeletal development.Placebo analgesia (PA) is defined as a psychobiological phenomenon triggered by the information surrounding an analgesic drug instead of its inherent pharmacological properties. PA is hypothesized to be formed through either verbal suggestions or conditioning. The present study aims at disentangling the neural correlates of expectations effects with or without conditioning through prior experience using the model of PA. We addressed this question by recruiting two groups of individuals holding comparable verbally-induced expectations regarding morphine analgesia but either (i) with or (ii) without prior experience with opioids. We then contrasted the two groups' neurocognitive response to acute heat-pain induction following the injection of sham morphine using electroencephalography (EEG). Topographic ERP analyses of the N2 and P2 pain evoked potential components allowed to test the hypothesis that PA involves distinct neural networks when induced by expectations with or without prior experience. First, we confirmed that the two groups showed corresponding expectations of morphine analgesia (Hedges' gs less then .4 positive control criteria, gs = .37 observed difference), and that our intervention induced a medium-sized PA (Hedges' gav ≥ .5 positive control, gav = .6 observed PA). We then tested our hypothesis on the recruitment of different PA-associated brain networks in individuals with versus without prior experience with opioids and found no evidence for a topographic N2 and P2 ERP components difference between the two groups. Our results thus suggest that in the presence of verbally-induced expectations, modifications in the PA-associated brain activity by conditioning are either absent or very small.
Electrospray ionization mass spectrometry (ESI-MS) is one of the most popular techniques for obtaining structural information, which is commonly used in bioanalysis and clinical diagnostics. However, for the detection of complicated samples with high reactivities (such as reactive sulfur species, RSS), traditional ESI-MS usually suffers from overlapped and inaccurate signals. In this study, based on the multiphase flow of extractive electrospray ionization (MF-EESI), an ambient MS technique of online derivatization was proposed to detect thiols without any other sample pretreatment.
RSS molecules and the derivatization reagent of 4-chloro-7-nitro-1,2,3-benzoxadiazole (NBD-Cl) were introduced into the internal and innermost capillary of the MF-EESI system, respectively. By a high-velocity nebulizing stream of N
gas through an external capillary, both flows of innermost biothiols and internal NBD-Cl were electrosprayed and mixed for online reactions. Therefore, the fast derivatization of thiols was used to generate stable ionized derivatives for MS detection.
By evaluating the changes in MS signals before and after the derivatization, the ions of RSS were identified simply and correctly. Without any sample pretreatment, the fast detection of cysteine, homocysteine, and glutathione has been achieved in the complicated samples.

This presented a potential pathway for the fast identification of thiols in complicated samples.Endoscopic population-based screening for gastric cancer began in April 2016, and the use of transnasal ultrathin esophagogastroduodenoscopy (UT-EGD) has rapidly become popular. With UT-EGD, discomfort associated with an examination is reduced, patient satisfaction is high, and adverse effects on cardiopulmonary function are fewer. Consequently, UT-EGD is a good option for gastric screening in an aging society. Because of the narrower diameter of the endoscope, however, image quality is inferior to that obtained using transoral conventional esophagogastroduodenoscopy (C-EGD). As a result, lesions observed by UT-EGD must be viewed at close proximity and chromoendoscopy should be used concurrently, which is burdensome for the endoscopist. Recent innovations by endoscope manufacturers have enabled dramatic improvements in transnasal UT-EGD and facilitated Hi-Vision imaging. Furthermore, image enhancement that allows for observation on a par with transoral C-EGD is now feasible. In the future, UT-EGD will be equipped with functions that permit magnified endoscopic image. It is anticipated that a multiroute transition between transnasal and transoral UT-EGD will become possible.In response to the COVID-19 outbreak, the British government introduced a lockdown resulting in country wide restrictions on movement and socialisation. This research sought to explore individuals' experience of the first lockdown in the UK. A qualitative online survey was conducted between April and June 2020. Using a convenience sample, 29 individuals participated in the study. Data were analysed using thematic analysis. Four themes were identified 'health and well-being', 'social connectedness and belonging', 'employment and finances' and 'personal and collective values'. Participants' experiences involved both challenges and opportunities. Participants reported challenges to their physical health, mental health, sense of connection to others as well as their employment and finances. Arboviruses are responsible for sporadic diseases or epidemics which cause serious public health issues. Due to the high number of asymptomatic infections and high viremia, blood donors may pass on these viruses by transfusion.
This study used blood bags that would be discarded after evaluation and certification of the absence of infections. The blood products obtained by centrifuging a unit of whole blood were called blood components. All blood bags were infected with viable viruses (previously quantified) compatible with Chikungunya and Mayaro viremia.
Blood bags inoculated with both Chikungunya and Mayaro viruses were able to keep infective viruses during the processing of blood products (red blood cell concentrate, platelet concentrate and fresh frozen plasma) and also after the recommended storage for each component, which may infect individuals transfused with those.
The results indicate that in order to prevent infections by Mayaro and Chikungunya viruses in blood products it is necessary to stimulate the development and use of diagnostic tests for these pathogens in donated blood.

Chronic disease is a growing problem affecting approximately half of all Australian adults. In response to growing calls for action on chronic disease, the My health for life program was created, aimed at improving the health of individuals at high risk of developing preventable chronic disease. The preventive health program is multi-modal, cross-culturally tailored and contains complex social marketing, community engagement, risk assessment and health promotion components. Therefore, a multi-component evaluation framework is essential to understand the effectiveness of the My health for life program. This brief report details the evaluation.
The evaluation design uses non-randomised, longitudinal analysis using repeated measures, observational, program goal-based and pretest-posttest design features to assess the program, its specific modalities and its program adaptations. To ensure timely and credible evaluation, different evaluative implementation frameworks and methods are considered. Quantitative and qualitative methods collect an array of program data at differing levels to assess the processes, outcomes and impacts of My health for life.
