However, current detection methods typically rely on expensive and bulky instrumentation. Here, we developed a simple, sensitive, instrument-free, CRISPR-based diagnostics of SARS-CoV-2 using a self-contained microfluidic system. The microfluidic chip integrates isothermal amplification, CRISPR cleavage, and lateral flow detection in a single, closed microfluidic platform, enabling contamination-free, visual detection. To simplify the operation and transportation of the device, we lyophilized the CRISPR reagents in the reaction chamber and pre-stored the liquid solutions in blisters. We employed a low-cost, portable hand warmer to incubate the microfluidic chip without the need for electricity. The self-contained microfluidic system can detect down to 100 copies of SARS-CoV-2 RNA. Further, we clinically validated our method by detecting 24 COVID-19 clinical nasopharyngeal swab samples, achieving excellent sensitivity (94.1%), specificity (100%), and accuracy (95.8%). This simple, sensitive, and affordable microfluidic system represents a promising tool for point-of-care diagnostics of COVID-19 and other infectious diseases.Monolayers of self-assembled quasi-spherical colloidal particles are essential building blocks in the field of materials science and engineering. More typically, they are used as a template for the fabrication of nanostructures if they serve, for instance, as a mask for deposition of new material on the surface on which particles are assembled or for etching of the material underneath; in this case, they are removed afterwards. This is what occurs in colloidal or nanosphere lithography. In some other cases, they are not used as a sacrificial material but they are incorporated in the final structure because they are inherently interesting for their properties. Independently of their specific use and application, different strategies have been devised in order to modify size and shape of colloidal particles, so as to enrich the variety of attainable patterns and to tailor the properties of the final structures and materials. In this review, we will focus on one of the most widespread methods to shape spherical colloidal particles, i.e. dry etching techniques. We will follow the development of such approaches until recent days, so as to trace an extensive panorama of the diverse parameters that can be harnessed to achieve specific morphological changes and highlight the characteristic features of the variants of this method. We will finally discuss how particles modified via dry etching can be used for patterning or can be resuspended in solvents for very diverse applications.Development of the kiwifruit industry has been severely hindered by the canker disease, which is caused by the bacterium Pseudomonas syringae pv. Actinidiae (Psa). However, endophytic fungi associated with healthy kiwi plants may protect host plants through the production of metabolites with potent anti-Psa activity. In the current study, four undescribed isobenzofuranones, namely sporulactones A‒D, two undescribed isocoumarins, namely sporulactones E and F, together with eight known analogs were isolated from the kiwi endophytic fungus Paraphaeosphaeria sporulosa. The structures with absolute configurations were established by extensive spectroscopic methods, quantum chemistry calculations, and X-ray diffraction experiments. In addition, five of the compounds exhibited anti-Psa activity, with minimum inhibitory concentration (MIC) values ranging from 25 to 100 μg/mL. So far research has revealed that farm animals such as cattle, domestic pigs, goats, sheep and horses contain the necessary receptors on the epithelial surface of the respiratory tract to accommodate binding of the IDV HEF protein of both worldwide clades D/Oklahoma (D/OK) and D/Oklahoma/660 (D/660). More recently, seroprevalence studies have identified IDV-seropositive wildlife such as wild boar, deer, dromedaries, and small ruminants. However, no research has thus far been conducted in wildlife to reveal the distribution of acetylated sialic acid receptors that accommodate binding of IDV. Using our previously developed tissue microarray (TMA) system, we developed TMAs containing respiratory tissues of various wild and domestic species including wild boar, deer, dromedary, springbok, water buffalo, tiger, hedgehog, and Asian elephant. Protein histochemical staining of these TMAs with HEF proteins showed no receptor binding for wild Suidae, Cervidae and tiger. The findings reveal that 60% of papers have been published between 2015 and 2020, and this topic is of global interest. Leading countries are mainly located in Europe, North America and Commonwealth; however, China and Malaysia are also assuming a driving role. Bibliometrics and text mining provide the intellectual configuration of the knowledge on recycling behaviour; co-word analysis individuates conceptual sub-domains in food waste, determinants of recycling behaviour, waste management system, waste electrical and electronic equipment (WEEE), higher-level education, plastic bags, and local government. Overall, waste management and related human behaviour represent a universal challenge requiring a structured and interdisciplinary approach at all levels (individual, institutions, industry, academia). Lastly, this paper offers some suggestions for future research such as smart city design, sensor network system, consumer responsibilisation, the adoption of a more comprehensive view of the areas of investigation through the holistic analysis of all stakeholders.In recent years, combined sewer overflow (CSO) has been identified as a significant contributor to the deterioration of the urban water environment. It is thought that remolding it to a separate sewer system is a thorough and effective method of controlling the CSO in the appropriate area. However, according to current research, the separate stormwater sewer systems will also have overflow pollution due to functional defects, damaged or inappropriately connected with sewage, which has serious consequences for the separate system's operational efficiencies and the urban water environment. The event mean concentration, first flush effect, source apportionment, and correlation analysis of variables in overflow pollution generated in three residential catchments in Nanning, China, were investigated in this study. The results showed that the event mean concentration values in drainage outlets inappropriately connected with sewage were 2-4 times higher than those in stormwater outlets, especially for NH3-N, TN, and TP. Meanwhile, more than 80% of overflow events at outlets inappropriately connected with sewage had a weak first flush or even a weak dilution effect, with peak pollutant concentrations occurring 40-60 min after the overflow began. Besides, the discharge pollution load was primarily derived from the inside of the sewer. When the rainfall was heavy, the contribution rate of sewer sediment erosion exceeded 60%, which was much higher than the contribution rate of rainfall runoff and sewage. The variability in event mean COD and TSS concentrations was primarily attributed to the antecedent dry period and rainfall intensity. The COD concentration increased from 140.7 to 277.1 mg/L with the increase of antecedent dry period from 3 to 10 days. This study could help guide the implementation of targeted measures to treat overflow pollution in urban residential catchments, as well as the development of strategies to mitigate the effects on receiving water bodies.Increasing metal demand is accelerating the mining and processing of minerals, however to ensure sustainable growth innovative approaches are required to better manage associated effluents. Biochar from the fast pyrolysis of residues from fishery and forestry operations has been studied as a low-cost, environmentally and economically friendly method for treating mine tailings and processing effluents. However, the bulk of the studies focus on terrestrial biomass (e.g. wood) and do not include potential inhibition/enhancement of adsorption due to pH controlling compounds. In this work biochar generated from snow crab (Chionoecetes Opilio) processing was studied as an adsorbent for copper solutions containing sulfate (a key compound in sulfide ore mining waters) with the objective of assessing adsorption capacity and the impact of sulfate on copper adsorption. We assessed the impacts of heavy traffic (TON), and the impacts of the same heavy equipment driven over top of access mats (AM), on soil physical, hydrological, and nutrient responses in sandy and loamy soils in the Dry Mixedgrass prairies over a 2-year period. We also assessed how the timing (early vs. late in the growing season) and duration (6 vs. 12 vs. 24 weeks) of AM and TON affected the same metrics. Compared to undisturbed soils, TON increased soil penetration resistance (15 cm depth) up to 93% in loamy and up to 101% in sandy soils, and decreased water infiltration rates from 53 to 71%, respectively.
