To test the regulation of NH3 on the apoptosis and autophagy of mammary epithelial cells, we selected NH4Cl as NH3 donor in vitro model. MTT and CCK-8 assay kits were employed to detect cell activity. Real-time quantitative PCR and western blot methods were used to detect the abundance of inflammatory molecules, apoptosis markers, and autophagy genes. We selected TUNEL kit and the Annexin-FITC/PI method to detect apoptosis. TEM analysis was used to detect autophagic vesicles, and MDC stain evaluated the formation of autophagosome. The results indicated that NH4Cl reduced cell viability in a concentration-dependent manner and promoted cell inflammatory response, apoptosis, and autophagy. NH4Cl stimulation notable increased the autophagosomes number. Interestingly, we also detected that the addition of LY294002 and Rapamycin inhibited the PI3K/Akt pathway and the mTOR pathway, respectively, resulting in changes in both apoptosis and autophagy. Therefore, we draw a conclusion that NH3 may regulate the apoptosis and autophagic response of bovine mammary epithelial cells through the PI3K/Akt/mTOR signaling pathway. Further investigations on ammonia's function in other physiological respects, will be critical to provide theoretical help for the improvement of production performance. It will be also helpful for controlling the harmful gas ammonia concentration in the livestock house to protect the health of dairy cows.Data on endoparasitic infections in dogs from dog shelters in Southeastern Europe are limited; thus, this study aimed to add to the existing knowledge on this topic by reporting on the prevalence of intestinal parasites in dogs from public dog shelters in the Republic of Serbia. In 2017 and 2018, individual and pooled fecal samples, were collected from 1267 dogs from six shelters. All samples were qualitatively examined for parasites using flotation tests. Seven taxa of intestinal parasites were identified Cystoisospora spp., ascarids Toxocara canis and Toxascaris leonina, hookworms, Trichuris vulpis, taeniids and Dipylidium caninum. The overall prevalence of intestinal parasites was 58. 3 % (78. 1 % in young dogs and 53.1 % in adult dogs). The parasites detected in both young (1 year old) were Cystoisospora spp. (20 % and 4.9 %), T. canis (33.5 % and 14.7 %), T. leonina (7.7 % and 2.3 %), and hookworms (16.9 % and 15 %), respectively. However, T. vulpis (9.6 %), taeniids (1.3 %), and D. caninum (5.4 %) were detected only in adult dogs. In the Belgrade shelter, young dogs had a higher prevalence of endoparasitic infections (18.9 %, 49/260) than adult dogs (14.8 %, 149/1007). In the Subotica, Jagodina and Niš shelters, young dogs had significantly higher (p less then 0.001 and p less then 0.05, respectively) prevalence of endoparasitic infections (10 %, 12.3 % and 14.6 %) than adult dogs (5.3 %, 8 % and 7.2 %). These results will be useful for establishing health care programs in dog shelters and implementing effective strategies for the control of intestinal parasites.Blastocystosis is an interesting parasitosis, since the parasitic infection is still seriously neglected and a considerable zoonotic evidence is emerging. Blastocystis sp. infects the intestinal lumen of humans and a wide range of animals, while there is a lack of comprehensive information on Blastocystis epidemiology in cattle worldwide. Thus, the present systematic review and meta-analysis were performed by exploring four electronic databases (PubMed, Web of Science, Scopus, and Google scholar) for relevant published papers up to 7th November 2020, utilizing a random-effects model to pool estimations and assign 95 % confidence intervals (CIs). Results of 28 studies (29 datasets) on cattle showed a 24.4 % (95 % CI 16.9-33.9 %) prevalence for Blastocystis infection. Also, 16 out of 26 reported subtypes (STs) were isolated from cattle, with ST10 (18 datasets) as the highest-reported [32.3 % (95 % CI 21.6-45.3)] as well as ST24 and ST25 (one study each) as the lowest-reported STs [1.4 % (95 % CI 0.2-9.1)]. Additionally, among nine well-known zoonotic STs (ST1-ST8 and ST12), all STs except for ST8 were reported from cattle worldwide, demonstrating this animal species as a potential reservoir for human infections. Meanwhile, the overall prevalence of Blastocystis in various subgroups (publication year, WHO regions, countries, continents, and age groups) was analyzed separately. The finding of the present review article highlights the cattle as a significant source of zoonotic transmission of Blastocystis infection to humans, which must be considered for preventive measures.Rabies is a highly fatal viral infection of the central nervous system affecting all warm-blooded animals including humans. To implement the preventive and control measures, it is important to decide the status of anti-rabies antibodies in dogs. Out of 120 serum samples, 47 (39.2 %) serum samples, showed an antibody titre equal to or above the cut off value of 0.5 IU/ml. The maximum number of dogs showed anti-rabies antibody titres equal to or above the cut-off value of 0.5 IU/ml after less then 1 month pre-exposure to the rabies vaccine. In 15 serum samples of pet dogs, we observed 13 (86.66 %) dogs with protective anti-rabies antibody titre. Statistical analysis suggests that the age of the animal had no significant effect on anti-rabies antibody titre in vaccinated pet dogs. The overall low seroprevalence of anti-rabies antibody in stray dogs indicates their susceptibility to rabies infection and thus posing a risk of rabies to other animals and humans.Melatonin (MT), an important antioxidant existing in plants and animals, has been widely reported to participate in the process of plants coping with stress. In this study, we demonstrated the mechanism of MT enhancing photosynthesis in rice under salt stress. The results showed that MT treatment increased relative water content, sucrose and starch content of rice under salt stress. This was mainly owing to the fact that MT enhanced the net photosynthetic rate and enhanced the absorption and transmission of light energy. The effect of MT on photosynthesis of rice under salt stress conditions was mainly due to the regulation of three processes maintaining low ROS status by improving the total antioxidant capacity, promoting the xanthophyll cycle and increasing the xanthophyll pool size to dissipate excess light energy, increasing the activities of key photosynthetic enzymes. Taken together, these results provide a mechanism for MT to improve the photosynthetic capacity of rice under salt stress.RNA has long been an enticing therapeutic target, but is now garnering increased attention, largely driven by clinical successes of RNA interference-based drugs. In this review, we focus on several recently developed bifunctional molecular technologies that both interact with and act upon a target RNA. These new approaches for programmable RNA knockdown, editing, splicing, translation, and chemical modifications stand to provide impactful new modalities for therapeutic development in the coming decades.
To explore the efficacy and tolerability of adjuvant perampanel (PER) and their associated risk factors in late add-on drug-resistant epilepsy.
Retrospective multicenter 'real-world' observational study. Consecutively identified patients commenced on PER, with mixed epilepsy syndromes, from nine Australian epilepsy centers. Primary efficacy endpoints were at least 50% reduction in seizure frequency (responders), seizure freedom, and retention at 6 and 12 months, following a 3-month titration period. Tolerability endpoints were cessation of PER for any reason, cessation of PER due to treatment-emergent adverse events (TEAE), or cessation due to inefficacy. Outcomes were assessed for a-priori risk factors associated with efficacy and tolerability.

A large body of research has documented that females have less positive attitudes and more negative affectivity to math than males. This study examined emotional valence ratings of math-related verbal stimuli among adults and performed a pioneering investigation of gender differences in emotional perceptions. A random sample of 290 adults completed a battery of online affect questionnaires designated to measure the relations of various math-related words to the field of mathematics (i.e., math loading) and compared the emotional valence of these words to words known to have negative and neutral valence. Results revealed that (1) math-related words were rated as less threatening than words with negative valence, but more threatening than neutral words; (2) math loading ratings were the strongest and most significant predictor of the emotional valence ratings of math-related words; and (3) females rated math-related words and words with negative, but not neutral, valence as more threatening than males. The study concludes that negative affective valence is linked with math-related information, especially among females, and this finding has implications for researchers, parents, and educators.The spread of African swine fever (ASF) poses a grave threat to the global swine industry.
