Atomic modeling calculations were performed at the quantum mechanical level based on Density Functional Theory and classical force fields based on empirical interatomic potentials. LDHNAP materials were prepared by ion exchange reaction from Mg2Al(OH)6Cl and Zn2Al(OH)6Cl pristine phases. The characterization of the materials confirmed NAP intercalation and also the permanence of the pristine phases in the isolated materials after ion exchange. Crystallographic lattice parameters, elemental analysis, and TGA experimental results were then employed in the calculations, which revealed that NAP anions can completely neutralize the positive charge of the LDH layers both Mg2Al and Zn2Al LDH structures could be optimized with all Cl- anions substituted by NAP. The drug assumed different dispositions in the NaNAP crystal or when intercalated into LDH. The performance of the model was evaluated by studying the adsorption of phenobarbital from aqueous solutions by commercial activated carbon. The amount of water occupying the adsorption sites was estimated by graphical analysis of the 'knee point' of water-vapor adsorption isotherms and was found to correlate well with the relative percentage of hydroxyl and carbonyl surface groups. It was found that one phenobarbital molecule displaces 2-6 water molecules from the adsorption site. It is shown that adsorption enthalpy was not affected by the adjustment for stoichiometry, supporting the notion that the van't Hoff enthalpy is intrinsic and is independent of the stoichiometry of solvent displacement in Langmuir-based binding. The widely reported disparities between the van't Hoff and calorimetric enthalpies are unlikely to be from a stoichiometric origin.Monoclonal antibodies are complex molecules that often require high concentration formulations to meet clinical requirement and bulk drug substance storage. A hydrogel film composed of 20%(w/w) hydroxypropyl methylcellulose (HPMC) irradiated with electron beams had high transparency and elasticity similar to commercially available wound dressings and soft tissues, suggesting that it is a suitable substrate for TRA. The inclusion of TRA was enabled by immersing the HPMC hydrogel film in TRA aqueous solution. The release and skin permeation of TRA from TRA-containing hydrogel films differed depending on the electron beam dose. Moreover, the analgesic effects in mice were confirmed in a dose-dependent manner. This study demonstrated the usefulness of a hydrogel film containing TRA as a new dosage form alternative to the existing oral medication for peripheral neuropathy.The gastrointestinal uptake of macrocyclic compounds is not fully understood. Here we applied our previously validated integrated system based on machine learning and conformal prediction to predict the passive fraction absorbed (fa), maximum fraction dissolved (fdiss), substrate specificities for major efflux transporters and total fraction absorbed (fa,tot) for a selected set of designed macrocyclic compounds (n = 37; MW 407-889 g/mol) and macrocyclic drugs (n = 16; MW 734-1203 g/mole) in vivo in man. Major aims were to increase the understanding of oral absorption of macrocycles and further validate our methodology. We predicted designed macrocycles to have high fa and low to high fdiss and fa,tot, and average estimates were higher than for the larger macrocyclic drugs. With few exceptions, compounds were predicted to be effluxed and well absorbed. A 2-fold median prediction error for fa,tot was achieved for macrocycles (validation set). Advantages with our methodology include that it enables predictions for macrocycles with low permeability, Caco-2 recovery and solubility (BCS IV), and provides prediction intervals and guides optimization of absorption. The understanding of oral absorption of macrocycles was increased and the methodology was validated for prediction of the uptake of macrocycles in man.Aluminum (Al) is known to induce neurotoxicity in both humans and rodents. Recent evidence has indicated that the toxicity of Al Oxide (Al2O3) nanoparticles (Al-NP), one of the most abundantly used engineered nanoparticles, is far greater than that of Al itself. To date, however, no information is available regarding the effect of Al-NP on the stereological parameters of hippocampus. In particular, no stereological studies have evaluated the effect of Al-NP on hippocampal CA1, dentate gyrus volume, and number of pyramidal and granular cells. Thus, the present study aimed to take a multidimensional approach to assess the concomitant cognitive, stereological, and apoptotic changes induced by a five-day Al-NP ingestion (10 mg/kg/day) in mice. The results demonstrated that the five-day Al-NP ingestion elicited a reduced preference to explore a novel object in the novel object recognition test (a hippocampal-dependent task). Perhaps contributing to this memory deficit, Al-NP induced additional alterations in the hippocampus of male NMRI mice in terms of (1) hippocampal volume (decreased the volume of the whole hippocampus, CA1, and dentate gyrus regions), (2) cell number (decreased the number of CA1 pyramidal neurons and dentate gyrus granular cells), and (3) increased cleaved caspase-3 in the whole hippocampus. These results provided new mechanistic insight to understand the impairing effect of AL-NP on the hippocampal function and structure.The extent to which bilingual language control (BLC) is related to domain-general executive control (EC) remains unclear. The present study applied activation likelihood estimation (ALE) meta-analyses to identify commonalities and distinctions in the brain regions across domains reported in neuroimaging studies. We specifically compare results from two experimental tasks language switching, a typical measure of BLC, and task switching, an experiment that measures EC. Conjunction analyses showed a domain-general pattern between language switching and task switching, with convergent activity in the left dorsolateral prefrontal cortex (DLPFC), pre-SMA/dACC complex (pre-supplementary motor area/dorsal anterior cingulate cortex), and left precuneus. Regarding domain-specificity, contrast analyses revealed stronger convergence of activation in the left fusiform gyrus and occipital gyrus in language switching compared to task switching, and conversely, stronger convergence of activation in the left DLPFC in task switching. These patients are at increased risk of viral contraction and have higher chances of severe disease/mortality. The latter is impacted by other factors and is still debated. In contrast to preliminary impressions, the benefits of anti-cancer treatments outweigh their risks and should be continued. Cancer patients generate antibodies in response to vaccination but in lower amounts than healthy people, especially those with hematologic cancers. Boosters, including third doses, have shown increased immune-responses in most patients. Vaccination should be prioritized in these individuals.
