Gram-positive pathogens remain a major cause of healthcare- and community-associated infections. In particular, the dissemination of methicillin-resistant staphylococci, such as methicillin-resistant Staphylococcus aureus (MRSA) and vancomycin-resistant enterococci (VRE), have significantly reduced the therapeutic options, making the management of these infections even more challenging. Dalbavancin is a second-generation lipoglycopeptide approved for the treatment of moderate to severe acute bacterial skin and skin structure infections (ABSSSIs) caused by Gram-positive organisms, showing a bactericidal effect and a low propensity towards the selection of resistance over time.

To evaluate the antimicrobial activity of dalbavancin and other comparators against recent clinical isolates of Gram-positive pathogens obtained from different sources and from several European countries, including countries of Southern and Eastern Europe, and Russia, where resistance rates are typically high. This study also aimed tnote, few coagulase-negative staphylococci (CoNS) isolates were resistant to dalbavancin and susceptible to vancomycin, highlighting the importance of testing for susceptibility to dalbavancin before its administration for CoNS infections.Most in vitro studies examine the effects of a single ED or a mixture of EDs on granulosa cells using short-term exposure; however, this approach is unlikely to reflect long-term, real-life exposures that are common in humans. We established an in vitro model that mimics long-term exposure of granulosa cells to real-life ED mixture. Human granulosa cells, HGrC1, were exposed to the mixture consisting of bisphenol A, polychlorinated biphenyl 153, benzo[a]pyrene, and perfluorooctanesulfonate in concentrations found in human follicular fluid (MIX) for 48 h and 4 weeks. Only long-term exposure to MIX decreased estradiol production after 2 and 3 weeks, and CYP19A1 protein after 2 weeks of exposure. By week 4, the cells restored estradiol production and CYP19A1 protein level. MIX increased basal progesterone production after 3 and 4 weeks of exposure but did not affect STAR and CYP11A1 mRNA. Cells that had been exposed to MIX for 4 weeks showed augmentation of forskolin-stimulated progesterone production. These results demonstrate that only long-term exposure to MIX alters steroidogenesis in HGrC1. This study also revealed that adverse effects of MIX on steroidogenesis in HGrC1 occurred a few weeks into MIX exposure and that this effect can be transient.Circular RNA (circRNA) is a non-coding RNA molecule that lacks polyadenylated tails and is highly stable, abundant, and conserved in human cells. CircRNAs can serve as a competing endogenous RNA (ceRNA) to sponge microRNAs (miRNA) and block their effects on target mRNA expression. CircRNAs also have possible relevance to cancer and therefore may be considered as ideal biomarkers for monitoring cancer progression. Of the about 300,000 predicted human circRNAs, only a few have validated biological functions related to cancer. To better understand the ceRNA role of circRNAs in colorectal cancer (CRC), we performed genome-wide circRNA-based RNA-sequencing (RNA-Seq) on nine CRC tumor samples and their paired histologically normal adjacent tissue samples. By profiling the mRNA expression in the same patients, we further explored the expression correlation between circRNAs and mRNAs generated from the same parental gene. Focusing on the concordant differential expression between circRNAs and mRNAs, we substantially diagnostic biomarker for CRC.Leucine-rich repeat kinase2 (LRRK2) influences the host immune responses and correlates with the pathogenesis of inflammation, cancer as well as Parkinson' Disease. Furthermore, scRNA-seq and co-culture experiments demonstrated that LRRK2 deficiency impaired macrophage functions, and influenced the neoplastic progression in a cancer type-specific manner. Therefore, the present study provided a therapeutic strategy for LGG based on the interference with LRRK2 expression and activity to prevent macrophage recruitment and promote tumor eradication.Extremely low coverage whole genome sequencing (lcWGS) is an economical technique to obtain high-density single nucleotide polymorphisms (SNPs). Here, we explored the feasibility of constructing a haplotype reference panel (lcHRP) using lcWGS and evaluated the effects of lcHRP through a genome-wide association study (GWAS) and genomic prediction in pigs. A total of 297 and 974 Duroc pigs were genotyped using lcWGS and a 50 K SNP array, respectively. We obtained 19,306,498 SNPs using lcWGS with an accuracy of 0.984. With the help of lcHRP, the accuracy of imputation from the SNP array to lcWGS was 0.922. Compared to the SNP array findings, those from the imputation-based GWAS identified more signals across four traits. With the integration of the top 1% imputation-based GWAS findings as genomic features, the accuracies of genomic prediction was improved by 6.0% to 13.2%. This study showed the great potential of lcWGS in pigs' molecular breeding.Environment-ubiquitous low-molecular-weight organic acids (LMWOAs) can interact with heavy metal ions and thus affect their mobility in subsurface aquifers. Herein, the effects of LMWOAs (including acetic acid, tartaric acid, malonic acid, oxalic acid, and citric acid) on the mobility of heavy metal ions (including Cd2+, Zn2+, Ni2+, Mn2+, and Co2+) in porous media were investigated to reveal the role of the stability constants of metal-LMWOA complexes in the mobility of heavy metal ions in porous media. The results showed that the mobility of different metal ions followed the order of Cd2+ acetic acid. An interesting finding was that there was a significantly positive correlation between the enhanced abilities of LMWOAs to metal mobility and the complex stability constants (log K) (R2 = 0.801-0.961, p less then 0.05), indicating that the complex stability of metal-LMWOA was the dominant factor responsible for the enhanced transport of heavy metal ions. Meanwhile, the linear slope indicated the intensity of enhancement of LMWOAs on heavy metal mobility was heavy metal type-dependent. This study proposed that the complex stability of metal-LMWOA could be an indicator to quantify and predict the impact of LMWOAs on the mobility of heavy metals.The eastern coastal region of China is the area with the highest emission of PAHs in China. Therefore, understanding the sources and health risk of parent polycyclic aromatic hydrocarbons (PPAHs) and their derivatives in eastern coastal cities of China is the main basis for air pollution control. In this study, we measured the concentrations of 18 parent PAHs, 17 nitrated PAHs, 7 oxygenated PAHs, 8 chlorinated PAHs, and 13 brominated PAHs in PM1.0 and PM2.5 samples collected at an urban coastal city site and a coastal background site in 2019. We analyzed the temporal distribution, molecular composition, and sources and performed health risk assessments for both winter and summer samples. The average concentration of the PPAHs and their derivatives (all 63 compounds combined) in the PM1.0 samples accounted for 75.57% of the PAHs concentration in PM2.5 samples. The average concentration of PM2.5- and PM1.0- bound PPAHs in winter was 114.70 times higher than in summer, and their derivatives was 27.51 times. Both the combined concentrations of the 18 PPAHs and the combined concentrations of the 45 derivatives were higher in the coastal city compared to the background site during the winter (1.90 and 1.48 times, respectively), but they were comparable during the summer. The positive matrix factorization analysis indicated that the compounds mainly originated from coal/biomass combustion, industrial sources, vehicle emissions, and secondary formation. In addition, the concentration-weighted trajectories model revealed that the PAHs were mainly emitted locally in Shandong Province and surrounding areas, such as Hebei Province, Henan Province, and Bohai Sea. The compounds 1-NPYR, 2+9-BrPHE, 9,10-Cl2PHE, and 1-ClPYR dominantly contributed to the derivatives of TEQ during the winter due to their high concentrations or the high TEFs of these compounds.We present a superhydrophobic material based on commercially melamine sponge (MS) with great durability, recyclability, and excellent sorption performance. The fabrication process of this sponge is facile without using toxic reagents or sophisticated equipment and therefore it is simple to scale up. The CuO layer utilized to give a rough surface of the substrate (MS) was successfully prepared in a commercial microwave to seed copper nucleuses in an alkaline medium. Stearic acid (SA) plays a role as the self-assembled monolayer on the surface of the sponge skeletons. Throughout this study, the properties of the modified sponge were fully characterized, and the changes in wettability were carefully examined. Water contact angle (WCA) measurements revealed the excellent superhydrophobicity of the material with high static WCA of 165.1° and low dynamic WCA of 8°. However, only 58% of cases have been associated with known risk factors (reproductive, hormonal, lifestyles, and genetic), and the rest to unknown causes. Nevertheless, growing evidence suggests that exposure to environmental contaminants is an important risk factor for BC. Polycyclic aromatic hydrocarbons (PAHs) are formed during organic matter combustion, including smoking, grilled meat, and fuels, and are important carcinogenic constituents of environmental pollution. We examined the information generated by epidemiological studies evaluating the association between BC and PAHs exposure from multiple sources. Our work was conducted according to Conducting Systematic Reviews and Meta-Analyses of Observational Studies of Etiology (COSMOS-E) guidelines. We searched PubMed, Web of Science, and Scopus from January 2000 to December 2019. A total of 124 records were identified, and only 23 articles met all inclusion criteria. Occupational and/or environmental exposure to PAHs was significantly associated with BC, irrespective of exposure being assessed by direct or indirect methods.
