In a randomised, placebo-controlled crossover intervention, subjects (n = 41) were given ordinary or hydrolysed high-protein, lactose-free milkshakes (500 mL, 50 g protein) to be consumed daily for ten days. After a washout period of ten days, the other product was consumed for another ten days. Gastrointestinal symptoms were recorded daily during the study periods, and a validated irritable bowel syndrome-symptom severity scale (IBS-SSS) questionnaire was completed at the beginning of the study and at the end of both study periods. Blood and urine samples were analysed for markers of inflammation, intestinal permeability and immune activation. Both the IBS-SSS score (p = 0.001) and total symptom score reported daily (p = 0.002) were significantly reduced when participants consumed the hydrolysed product. Less bloating was reported during both study periods when compared with the baseline (p less then 0.01 for both groups). Flatulence (p = 0.01) and heartburn (p = 0.03) decreased when consuming the hydrolysed product but not when drinking the control product. No significant differences in the levels of inflammatory markers (tumor necrosis factor alpha, TNF-α and interleukin 6, IL-6), intestinal permeability (fatty acid binding protein 2, FABP2) or immune activation (1-methylhistamine) were detected between the treatment periods. The results suggest that the partial hydrolysation of milk proteins (mainly casein) reduces subjective symptoms to some extent in subjects with functional gastrointestinal disorders. The mechanism remains to be resolved.Alternative splicing, a ubiquitous phenomenon in eukaryotes, is a regulatory mechanism for the biological diversity of individual genes. Most studies have focused on the effects of alternative splicing for protein synthesis. However, the transcriptome-wide influence of alternative splicing on RNA subcellular localization has rarely been studied. By analyzing RNA-seq data obtained from subcellular fractions across 13 human cell lines, we identified 8720 switching genes between the cytoplasm and the nucleus. Consistent with previous reports, intron retention was observed to be enriched in the nuclear transcript variants. Interestingly, we found that short and structurally stable introns were positively correlated with nuclear localization. Motif analysis reveals that fourteen RNA-binding protein (RBPs) are prone to be preferentially bound with such introns. To our knowledge, this is the first transcriptome-wide study to analyze and evaluate the effect of alternative splicing on RNA subcellular localization. BLU9931 To develop a novel platform compound, we synthesized a flavonoid derivative, (E)-5-((4-oxo-4H-chromen-3-yl)methyleneamino)-1-phenyl-1H-pyrazole-4-carbonitrile (named DK4023) and characterized its inhibitory effects on the motility and MMP2 and MMP9 expression of highly metastatic MDA-MB-231 breast cancer cells. We found that DK4023 inhibited tumor necrosis factor alpha (TNFα)-induced motility and F-actin formation of MDA-MB-231 cells. DK4023 also suppressed the TNFα-induced mRNA expression of MMP9 through the downregulation of the TNFα-extracellular signal-regulated kinase (ERK)/early growth response 1 (EGR-1) signaling axis. These results suggest that DK4023 could serve as a potential platform compound for the development of novel chemopreventive/chemotherapeutic agents against invasive breast cancer.We numerically and theoretically investigate a highly sensitive and tunable plasmonic refractive index sensor that is composed of a metal-insulator-metal waveguide with a side-coupled nanoring, containing silver nanorods using the finite element method. Results reveal that the presence of silver nanorods in the nanoring has a significant impact on sensitivity and tunability performance. It gives a flexible way to tune the system response in the proposed structure. Our designed sensor has a sensitivity of 2080 nm/RIU (RIU is the refractive index unit) along with a figure of merit and a quality factor of 29.92 and 29.67, respectively. The adequate refractive index sensitivity can increase by adding the silver nanorods in a nanoring, which can induce new surface plasmon polaritons (SPPs) modes that cannot be found by a regular nanoring. Pharmacogenetics aims to identify specific genetic variants that influence pharmacokinetics and pharmacodynamics of drugs, better determining their effectiveness/safety profile. Opioid response is a complex scenario, but some gene variants have shown a correlation with pain sensitivity, as well as with opioid metabolism and clinical efficacy/adverse events. Although questions remain unanswered, some of these gene variants may already be used to identify specific patients' phenotypes that are more prone to experience better clinical response (i.e., better analgesia and/or less adverse events). Once adopted, this approach to opioid prescription may improve a patient's outcome. This review summarizes the available data on genetic variants and opioid response we will focus on basic pharmacogenetic and its impact in the clinical scenario discussing how they may lead to more appropriate opioid prescription in cancer patients.The use of drainage water in the irrigation of agroecosystem is associated with environmental hazards, and can pose threats to human health. Nine heavy metals (Fe, Mn, Zn, Cu, Co, Cr, Ni, Cd and Pb) along three main drains in the middle Nile Delta were measures in the sediments, roots and shoots of three common macrophytes (Echinochloa stagnina, Phragmites australis and Typha domingensis). The physicochemical characteristics, as well as the enrichment factor (Ef), contamination factor (Cf), geoaccumulation index (Igeo), ecological risk factor (Er), degree of contamination (Dc) and potential ecological risk index (PERI), were determined for sediment. The metal bioaccumulation factor (BAF) and translocation factor (TF) were assessed for plants. Data revealed high contents of Cr, Zn and Cd in the upstream of the drains, while Mn, Cu and Ni were recorded in high concentrations in the downstream. Mn, Cr, Co, Cu, Ni and Zn were recorded to be within EU (2002), CSQGD (2007) and US EPA (1999) limits, while Cd and Pb showed high a ecological risk index. This high concentration of pollutants could be attributed to unremitting industrial activities, which can bioaccumulate in the food chains and cause serious problems for humans. The root of P. australis showed the effective accumulation of most of the elements, while T. domingensis revealed the highest accumulation of Pb. However, the highest BAF shoot value was found in T. domingensis for most of the heavy metals, except for Fe and Zn in P. australis and Mn in E. stagnina. Thus, P. australis could be used as a potential phytoextractor of these hazardous metals, as an eco-friendly and cost-efficient method for remediation of the polluted drains. Further, T. domingensis could be integrated as a hyperaccumulator of Pb. Strict laws and regulations must be taken into consideration by the policymaker against unmanaged industrial activities, particularly near the water streams in the Nile Delta.Principal component analysis (PCA)-based method is popular for detecting the damage of bridges under varying environmental temperatures. However, this method deletes some information when the damage features are projected in the direction of nonprincipal components; thus, the effectiveness of PCA-based methods will decrease if the deleted information is related to bridge damage. To address this issue, a hybrid method is proposed to detect the damage of bridges under environmental temperature changes. On one side, the PCA-based method is applied to deal with the nonprincipal components; on the other side, the Gaussian mixture method (GMM) is used to classify all the principal components into different clusters, and then the novel detection method is implemented to detect bridge damage for each cluster. In this way, all the damage feature information is saved and used to detect bridge damage. The numerical example and example of an actual bridge show that the proposed hybrid method is effective in detecting bridge damage under environmental temperature changes. The GMM is effective for classifying the natural monitoring frequency data of actual bridges, and the relationship between the natural frequencies of actual bridges and the environmental temperature is not always linear.Background and Objectives Dehydration might be an issue after hip fracture surgery, but the optimal tools to identify the dehydrated condition have not been determined. The aim of the present study was to compare the characteristics of elderly postoperative patients who were classified as dehydrated according to the methods used in the clinic. Materials and Methods Thirty-eight patients aged between 65 and 97 (mean, 82) years were studied after being admitted to a geriatric department for rehabilitation after hip fracture surgery. Each patient underwent blood analyses, urine sampling, and clinical examinations. Results Patients ingested a mean of 1,008 mL (standard deviation, 309 mL) of fluid during their first day at the clinic. Serum osmolality increased significantly with the plasma concentrations of sodium, creatinine, and urea. Seven patients had high serum osmolality (≥300 mosmol/kg) that correlated with the presence of tongue furrows (p less then 0.04), poor skin turgor (p less then 0.03), and pronounced albuminuria (p less then 0.03). Eight patients had concentrated urine (urine-specific gravity ≥ 1.025) that correlated with a low intake of liquid and with a decrease in body weight during the past month of -3.0 kg (25-75 th percentiles, -5.1 to -0.9) versus +0.2 (-1.9 to +2.7) kg (p less then 0.04). Conclusions Renal fluid conservation of water, either in the form of hyperosmolality or concentrated urine, was found in 40% of the patients after hip fracture surgery. Hyperosmolality might not indicate a more severe fluid deficit than is indicated by concentrated urine but suggests an impaired ability to concentrate the urine.
