PCoA showed that the methanogen community in H2/CO2 and acetate amendments exhibited greater differences than those in other treatments. Lower temperature (10 °C) limits hydrogenotrophic and acetoclastic methanogenesis, but methylotrophic methanogenesis is much less affected. The response of methanogen diversity to the incubation temperature varied among the different substrate-amended slurries. The multitrophic methanogen Methanosarcina became increasingly abundant in H2/CO2- and acetate-amended sediment slurries when the temperature increased from 10 to 30 °C.The wide use of chlorinated organophosphate esters (Cl-OPEs) as additive flame retardants has aroused concern about their potential risks on ecosystem and human health. However, knowledge about the toxicity of Cl-OPEs on soil organisms remains limited. In this study, earthworms, Eisenia fetida, were exposed to three representative Cl-OPEs, i.e., tris(2-chloroethyl) phosphate (TCEP), tris(2-chloro-1-methylethyl) phosphate (TCPP), and tris(1,3-dichloro-2-propyl) phosphate (TDCPP) in artificial soil. Using a combination of biochemical indicators (biomarkers), transcriptomics, and metabolomics, we compared the Cl-OPE-induced toxicity to E. fetida and provide new insight into the related molecular mechanism. All three Cl-OPEs elicited immune defense by the earthworms, as evidenced by increased acid phosphatase and alkaline phosphatase activities, and the genes involved in immune-related pathways (e.g., lysosomal and interleukin-17 signaling pathways). Furthermore, no effects on acetylcholinesterase activity were observed among the three Cl-OPEs. However, the TCPP and TDCPP treatments significantly decreased the neurotransmitter serotonin, suggesting the potential neurotoxicity of Cl-OPEs. Although TCEP affected the genes involved in carbohydrate and amino acid metabolism, the changes in the corresponding metabolites were not statistically significant. In contrast, exposure to TCPP and TDCPP induced oxidative stress, and affected xenobiotic metabolism and energy metabolism, leading to the decreased body weight in E. fetida. Based on these toxic effects, TCPP and TDCPP were more severely toxic than TCEP, despite their structural similarity. Given that the use of TCEP has been tightly regulated, our results suggest the potentially toxic effects of TCPP and TDCPP should not be ignored in future risk assessments of flame retardants.Rising concern about emerging and already persisting pollutants in water has urged the scientific community to develop novel remedial techniques. A new group of remediation methods is based on the modification of nanoscale zero-valent iron particles (nZVI), which are well known for treating volatile organic compounds and heavy metals. The properties of nZVI may be further enhanced by modifying their structure or surface using "green" polymers. Herein, nZVI was modified by a β-cyclodextrin polymer (β-CDP), which is considered an environmentally safe and inexpensive adsorbent of contaminants. This composite was used for the first time for the degradation of sulfamethoxazole (SMX). Coating by β-CDP not only enhanced the degradation of SMX (>95%, under 10 min) by the nanoparticles in a wide pH range (3-9) and enabled their efficient reusability (for three cycles) but also made the coated nZVI less toxic to the model bioindicator microalga Raphidocelis subcapitata. Moreover, degradation products of SMX were found to be less toxic to Escherichia coli bacteria and R. subcapitata microalga, contrary to the SMX antibiotic itself, indicating a simple and eco-friendly cleaning process. This research aims to further stimulate and develop novel remedial techniques based on nZVI, and provides a potential application in the degradation of antibiotics in a wide pH range. Moreover, the wealth of available cyclodextrin materials used for surface modification may open a way to discover more efficient and attractive composites for environmental applications.Analogous to flow regime, this study proposed a new statistical framework to assess inter-annual and intra-annual terrestrial water storage (TWS) regime and its changes from the aspects of magnitude, variability, duration and components. The framework was applied to two endorheic basins, Inner Basin (IB) and Qaidam Basin (QB), in the Tibetan Plateau and their eight sub-regions. Our major findings are as follows (1) TWS in the IB (2.09-2.35 mm/a, P 50 mm), accompanied by a longer duration and higher variability in the IB and its northeastern sub-regions. There was a large reduction in low storage (-18.25 mm) combined with higher variability in southeastern IB; (3) SWA change dominated the storage surplus in summer (82%) and storage deficit in autumn (-78%) and winter (-51%) in the IB, while GLPIA change dominated the storage surplus in spring (57%). In the QB, TWS change was mainly contributed by SWA change in spring (94%) and by GLPIA change in summer (73%), autumn (-62%) and winter (-58%). Component contribution rates showed a significant change in spring and winter but not much change in summer and autumn, indicating that the TWS components were more sensitive to climate change in the cold season.Triclosan (TCS) and copper (Cu2+) were exposed to aerobic granular sludge (AGS) system treating wastewater containing environmental tetracycline, respectively, to explore the different biochemical responses, more importantly, the fates of resistance genes (RGs) in AGS system. The results showed that TCS and Cu2+ could significantly inhibit the N and P removal in AGS system by reducing several key functional genes, including amoA gene of ammonia-oxidizing bacteria, Nitrospira and phosphorus accumulating organisms 16S rRNA genes. TCS caused higher degree of RGs' enrichment than Cu2+, which made the average total relative abundance of RGs of 1.38 ± 0.73 and 0.78 ± 0.24 in TCS and Cu system, respectively. Cu2+ could induce a wider range of horizontal gene transfer than TCS, leading to the detections of more potential hosts harboring RGs in Cu system. Cu system seemed to have stronger repair, immunity and defense ability than TCS system, which enabled it to have sufficient ability to trigger protection mechanism to realize self-protection, eventually the RGs also were controlled. Integron (intI1 and intI3) and plasmids (trb-C and IncQ) might cooperate with microorganisms and water quality parameters to enhance the enrichment of RGs in TCS system, however this interaction among various environmental factors was not obvious in Cu system, which might be responsible for the lower abundance of RGs. The increasing levels of TCS and Cu2+ in wastewater should be paid more attentions during the treatment of wastewater containing environmental tetracycline by AGS system. Selleck AGI-24512 Especially for TCS, it had the ability to enrich RGs more easily than Cu2+, which should be prevented from entering wastewater treatment plants as far as possible, to avoid more serious proliferation and dissemination of various RGs.The chemistry of dissolved organic matter (DOM) in soil determines its bioavailability and is crucial to soil nutrient cycling and vegetation restoration. However, the response of DOM to revegetation in degraded ecosystems is not well understood. This study analyzed soil chronosequences from semifixed (SF) sand in the Mu Us Desert, China that was afforested with arbor and shrub 23-54 years ago. The DOM organic composition and chemical properties were investigated by fluorescence excitation-emission matrix spectrophotometry and UV-visible spectroscopy. As the revegetation progressed, DOM concentration at 0-20 cm continually increased. In arbor land, the DOM was mainly influenced by dissolved organic carbon, and the average concentration significantly increased from 67.5 to 203.2 mg C•kg-1. In the shrubland, changes in DOM were mainly influenced by dissolved organic nitrogen, with the average concentration significantly increasing from 3.7 to 8.9 mg N•kg-1. Arbor afforestation significantly increased the proportion of hydrophobic humic-like components (C1), with a decline in bioactive tryptophan-like components (C3) and higher DOM molecular complexity (i.e., higher molecular weight, aromaticity, and degree of humification). However, in shrubland, the proportions of C1 and C3 were significantly lower than those in arbor forest soil, with DOM molecular complexity slightly increased, and DOM tended to become more bioavailable over time. Linear regression analysis indicated that DOM rich in carbon had a high molecular complexity, while DOM rich in nitrogen is more bioavailable. These findings reveal a dissimilar evolution in the DOM composition and chemistry in the two forest types throughout the SF sand revegetation period. The DOM under arbor forest was more stable and conducive to soil organic matter sequestration, while the DOM under shrub forest contained more bioactive substances and was more favorable for nutrient cycling. As a broad-spectrum antibacterial agent, triclosan (TCS) has been confirmed to possess potential immunotoxicity to organisms, but the underlying mechanisms remains unclear. Herein, with the aid of transgenic zebrafish strains Tg (coro1A EGFP) and Tg (rag2 DsRed), we intuitively observed acute TCS exposure caused the drastic differentiation, abnormal development and distribution of innate immune cells, as well as barriers to formation of adaptive immune T cells. These abnormalities implied occurrence of the cytokine storm, which was further evidenced by expression changes of immune-related genes, and functional biomarkers. Based on transcriptome deep sequencing, target gene prediction and dual luciferase validation, the highly conservative and up-regulated miR-19a was chosen as the research target. Under TCS exposure, miR-19a up-regulation triggered down-regulation of its target gene socs3b, and simultaneously activated the downstream IL-6/STAT3 signaling pathway. Artificial over-expression and knock-down of miR-19a was realized by microinjecting agomir and antagomir, respectively, in 1-2-cell embryos. The miR-19a up-regulation inhibited socs3b expression to activate IL-6/STAT3 pathway, and yielded abnormal changes in the functional cytokine biomarkers, along with the sharp activation of immune responses. These findings disclose the molecular mechanisms regarding TCS-induced immunotoxicity, and offer important theoretical guidance for healthy safety evaluation and disease early warning from TCS pollution. This study investigated the removal of selected per- and polyfluoroalkyl substances (PFAS) from water via two green sorption media (IFGEM-7 and AGEM-2). Both selected green sorption media recipes contain sand (85-91%) and clay (3-4%), in addition to recycled iron (Fe) (5-7.5%) or aluminum (Al) (4.5% in AGEM-2 only). Batch and column studies were integrated and performed using the prescribed green sorption media recipes to determine their efficiencies in removing two most targeted PFAS, including perfluorooctanesulfonic acid (PFOS) and perfluorooctanoic acid (PFOA). In the batch test, while the removal efficiencies of PFOS ranged from 27 to 46% and 23 to 42%, those for PFOA ranged from 6 to 16% and 5 to 18% when using IFGEM-7 and AGEM-2, respectively. The higher removal of PFOS than PFOA observed in both IFGEM-7 and AGEM-2 batch tests could be attributed to higher media affinity for sulfonate groups of PFOS when compared to the carboxylate groups of PFOA. In the column study, the initial removal (within 1 h) by IFGEM-7 was greater than 99% for PFOS and 28% for PFOA.
