Purpose Trauma is recognized as a significant health concern across the world. The increase in the mortality rate of multiple trauma (MT) patients serves as a major issue for health-care providers. The study investigates the relationship between the initial arterial blood gases and coagulation profiles among the multiple injured/trauma patients. Patients and Methods A retrospective study design was used, and 137 acute trauma patients' data were included from the King Abdullah University Hospital (KAUH) from 1 January 2014 to 1 October 2017. Results The results showed that patients were mainly male (79.6%) and had RTA (71.5%). They had a normal pH (50.4%) and pCO2 (68.6%) while decreasing HCO3 (51.1%). An increasing level of pCO2 (45%) along with PT (28%), PTT (17%) and INR (23%) is observed. It also showed a decreasing level of platelets (39%). The overall findings showed a close association of the blood gases and coagulation indicators. Conclusion The study concludes that to control the coagulation markers, the blood gases of the patients should be screened and monitored. © 2020 Raffee et al.Background Preterm birth is one of the most common problems in pregnancy globally, often leading to neonatal death. The rate of preterm birth is increasing every year. The etiology of preterm birth is multifactorial; preventable maternal psychosocial stress is one of the causes of preterm birth. There has not yet been a report of maternal psychosocial stress and preterm birth in Thailand. This study aimed to explore the relationship between maternal perceived stress and preterm birth. Objective To study the effects of maternal perceived stress on preterm birth. Patients and Methods An unmatched case-control study was conducted in postpartum women, between January 17, 2019, and September 30, 2019, at the Department of Obstetrics and Gynecology, Faculty of Medicine, Vajira Hospital. A total of 200 participants were enrolled and divided into case and control groups in a 11 ratio. The case group consisted of postpartum women with preterm birth and the control group consisted of consecutive postpartum women with term birth. Perceived stress was measured by using the Thai Perceived Stress Scale-10 (T-PSS-10). Results Maternal perceived stress during pregnancy was significantly greater in preterm birth compared with term birth in both means score (19.43±4.48 vs 14.08±4.06, p less then 0.001) and moderate to high stress level (92 vs 51, p less then 0.001). Perceived stress score and stress level were significant predictive factors for preterm birth (AOR 1.31, 95% CI 1.20-1.44, P less then 0.001 and AOR 8.66, 95% CI 3.64-20.63, P less then 0.001, respectively). Conclusion Maternal perceived stress during pregnancy was statistically greater in preterm birth. Perceived stress score and stress level were significant predictive factors for preterm birth. © 2020 Tanpradit and Kaewkiattikun.Pycnodysostosis (PYCD) is a rare autosomal recessive skeletal dysplasia arising from a change in the production of the cathepsin K enzyme and it is also known as Maroteaux-Lamy Syndrome. The main changes in the buccal and maxillofacial region are modifications of the facial bones (leading to the loss of the mandibular angle due to micrognathia), deep palate, premature and/or delayed tooth eruption, hypopneumatization of the maxillary sinuses, chin hypoplasia, hypercementosis, enamel hypoplasia, hyperdontia, incomplete dentin calcification, root hypoplasia, pulp chamber obliteration, and increased free functional space. This paper aims to report a case of a patient with PYCD, in which are highlighted the oral manifestations of this syndrome and present an option of oral rehabilitative treatment through a removable overdenture made on natural tooth of the patient. The technique of dental coating through overdenture is a simple, viable and conservative alternative for oral rehabilitation of patients with large maxillomandibular discrepancies, as in cases of pycnodysostosis, especially in the presence of increased free space - characteristic of patients with this syndrome. The result was quite satisfactory both aesthetically and functionally and provided a better life quality for the patient. © 2020 Costa et al.Background Out-of-hours (OOH) health care services are often divided into emergency medical services (EMS) and OOH primary care (OOH-PC). EMS and many OOH-PC use telephone triage, yet the patient still makes the initial choice of contacting a service and which service. Sociodemographic characteristics are associated with help-seeking. Yet, differences in characteristics for EMS and OOH-PC patients have not been investigated in any large-scale cohort studies. Such knowledge may contribute to organizing OOH services to match patient needs. Thus, in this study we aimed to explore which sociodemographic patient characteristics were associated with utilizing OOH health care and to explore which sociodemographic characteristics were associated with EMS or OOH-PC contact. Methods A population-based observational cohort study of inhabitants in two regions (North Denmark Region and Capital Region of Copenhagen) with or without contact to OOH services during 2016 was conducted. Associations between sociodemographic chae service outside office hours and with contacting EMS rather than OOH-PC. Old age, low income, low education and low socioeconomic status were of greatest importance. © 2020 Søvsø et al.Background and Aim Hepatocellular carcinoma (HCC) is a type of cancer with high mortality rates. The overexpression of microRNA-519d (miR-519d) has been explored in different types of cancers, which could significantly help suppress cancer development. This study aimed to investigate the interaction of miR-519d with its target gene, Rab10, as well as its effects on cell proliferation and autophagy in HCC cells through modulation of the AMPK signaling pathway. Methods Microarray analysis was used to analyze the differentially expressed genes in HCC, and the target genes of the screened-out miRNA were predicted and verified. The expression of miR-519d and Rab10, AMPK signaling pathway-related proteins, apoptosis- and autophagy-related proteins was determined by RT-qPCR and Western blot analysis in HCC tissues and cell lines. Lastly, the effects of miR-519d and Rab10 in HCC cell proliferation, apoptosis, and mouse tumour xenograft in vivo were examined through gain- and loss-of-function experiments. Results MiR-519d was down-regulated and Rab10 was upregulated in HCC tissues and cell lines. Overexpression of miR-519d decreased the expression of Rab10, mTOR, and Bcl-2, but increased the expression of Bax, Beclin1, Atg5, and p53. Upregulated miR-519d and downregulated Rab10 expression suppressed cell proliferation and induced cell apoptosis and autophagy in HCC cells. Finally, upregulation of miR-519d inhibited tumour growth in vivo. Conclusion The result obtained in this study indicates that up-regulation of miR-519d inhibits proliferation and promotes apoptosis and autophagy of HCC cells through activation of the AMPK signaling pathway via downregulating Rab10, which provides a potential target for the treatment of HCC. © 2020 Zhang et al.Background Osteosarcoma (OS), an aggressive malignant neoplasm, exhibits osteoblastic differentiation. Cisplatin (DDP) and taxanes are among the most effective drugs for OS patients. Nevertheless, the drug resistance remains a main limitation to efficacious chemotherapy in OS. The current report sets to explore the biological function of microRNA-584 (miR-584) and the potential mechanism underlying OS cells resistance to these two drugs. Materials and Methods The expression profiles of miR-584 and connective tissue growth factor (CTGF, CCN2) in OS tissue samples and cell lines were tested by means of reverse transcription-quantitative polymerase chain reaction and Western blot. U2OS and MG63 cell lines were delivered with miR-584 mimic alone or plus CCN2 to excavate theirs functions by cell counting kit-8 and EdU, flow cytometric analysis, as well as transwell assay, severally. Western bot analysis was conducted to examine the expression of IκBα, pIκBα, NF-κB and pNF-κB. Dual-luciferase reporter gene assay was carried out to assess the targets of miR-584. Results The downregulation of miR-584 was identified in OS tissues and cells, which was closely linked to the dismal prognosis of OS patients. Overexpression of miR-584 repressed cell viability, migration as well as invasion, potentiated apoptosis and sensitized OS cells to DDP and taxanes. Mechanism investigation specified a direct targeting relationship between CCN2 and miR-584 in OS. Conclusion In conclusion, miR-584 has the potency to act as a therapeutic maneuver for OS mainly by inducing the chemosensitivity of OS cells to DDP and taxanes. © 2020 Li et al.Objective The aim of this study is to assess the diagnostic and screening performance of a standardized methylation-specific real-time PCR assay targeting SOX1 and PAX1 genes for cervical cancer in a Chinese cohort. Methods Genomic DNA was extracted from cervical exfoliated cells and converted by sodium bisulfite and then analyzed by qMSP assay. Ct values were collected for PAX1 and SOX1 as target genes and β-actin as an endogenous reference gene. The samples included 295 cervicitis, 111 LSIL (low-grade squamous intraepithelial lesion), 51 HSIL (high-grade squamous intraepithelial lesion) and 30 cervical cancer. Results The Ct values decreased with the progression of cervical cancer from cervicitis, through LSIL and HSIL to cancer. The difference in Ct values between cytological grades was highly significant (p≤0.01) between grades either for PAX1 or for SOX1 except the difference between cervicitis and LSIL of SOX1. RZ-2994 With the Ct cut-off values of PAX1 gene and SOX1 gene 38.6 and 38 and with the PAX1/SOX1 in combination, the positive rate of methylation in invasive cancer tissues was 100%, in contrast to 11.5% (95% CI 8.67%-14.33%) in cervicitis tissues, 45.1% (95% CI 40.68%-49.52%) in LSIL tissues, and 68.5% (95% CI 64.37%-72.63%) in HSIL tissues. The specificity and sensitivity of differentiating tumors from cervicitis were 0.957 (95% CI 0.939-0.975) and 1.00, respectively. The specificity and sensitivity of differentiation between cervicitis+LSIL and HSIL+cervical cancer were 0.881 (95% CI 0.852-0.91) and 0.748 (95% CI 0.709-0.787), respectively. Conclusion PAX1/SOX1 methylation could be translated into clinical practice for cervical neoplasia detection. © 2020 Zhang et al.Introduction Colorectal cancer (CRC) is one of the most common illnesses that seriously threatens human health; many papers have reported that microRNAs (miRNAs) are promising biomarkers for cancer detection. However, miRNAs have not been used in clinical practice even though they are superior to the currently used screening tools, such as the fecal occult blood test (FOBT) and carcinoembryonic antigen (CEA) measurement. Methods In this study, we focused on the usefulness of a panel of miRNAs and the combination of miRNAs with the FOBT and CEA measurement, the currently used general diagnosis methods, to improve the accuracy of CRC diagnosis. Results The results showed that the miRNA panel has great potential value as a diagnostic biomarker with high specificity and sensitivity, and further analysis demonstrated that the miRNA panel had higher sensitivity and specificity than the FOBT and CEA measurement, even when these methods were combined. More importantly, although the miRNA panel is superior to the FOBT and CEA measurement, it cannot replace them.
