In this article, we (i) explain the rationale for considering anticoagulation therapy in patients with kidney failure to reduce atherothrombotic events, (ii) highlight the limitations of current anticoagulants in this patient population, (iii) explain the potential benefits of FXI inhibitors, and (iv) summarize ongoing studies investigating FXI inhibition in patients with kidney failure on dialysis.Additive manufacturing (AM) or 3D printing is enabling new directions in product design. The adoption of AM in various industrial sectors has led to major transformations. Similarly, AM presents new opportunities in the field of drug delivery, opening new avenues for improved patient care. In this review, we discuss AM as an innovative tool for drug product design. We provide a brief overview of the different AM processes and their respective impact on the design of drug delivery systems. We highlight several enabling features of AM, including unconventional release, customization, and miniaturization, and discuss several applications of AM for the fabrication of drug products. This includes products that have been approved or are in development. As the field matures, there are also several new challenges to broad implementation in the pharmaceutical landscape. We discuss several of these from the regulatory and industrial perspectives and provide an outlook for how these issues may be addressed. The introduction of AM into the field of drug delivery is an enabling technology and many new drug products can be created through productive collaboration of engineers, materials scientists, pharmaceutical scientists, and industrial partners.Previous studies show that intermittent social defeat (ISD) stress increases self-administration of psychostimulants, which suggests that ISD promotes reward-seeking behavior and, ultimately, increases vulnerability to develop drug abuse. The present study investigates whether ISD alters cost/benefit evaluations to promote reward-seeking behavior and whether these alterations are time-dependent. Male rats performed two different tasks that assessed their motivation to seek and consume food rewards. An effort-discounting task in which rats chose between less and more effortful options (i.e., 1 lever-press versus 2, 5, 10 or 20 lever-presses) associated with low- and high-reward (i.e., 1 sugar pellet versus 3 sugar pellets), respectively; and a progressive ratio task in which rats had to increase their effort (more lever presses) to obtain a sugar pellet. ISD consisted of exposing animals to social defeat once every three days for ten days (4 stress episodes). Rats were tested 24-48 h after stress episodes, and 1 week and 6 weeks after the last stress episode. In the effort-discounting task, stressed animals showed a decrease in their preference for high rewards associated with more effort (i.e., 10 and 20 lever-presses). These effects were transient and not maintained one week after stress. In the progressive ratio task, stressed animals showed an increase in the number of lever presses to obtain rewards that emerged six weeks after the last stress episode. These results suggest different short- and long-term effects on the motivation for rewards after ISD and indicate temporal dynamic adaptations in the function of the brain reward system.Theories of consciousness diverge on the functional requirement that a conscious state need be reportable. Some maintain that the perceptual system's capacity for consciousness exceeds that of its capacity for access. Others contend that what is accessed is all there is to consciousness. Here, we suggest a compelling case for access-free consciousness cannot be made reliant on experimental evidence where access is necessarily invoked. However, a bona fide empirical separation of consciousness and report could counter the claim that reportability, and hence access, is all there is to consciousness. We first overview recent neurophysiological findings from no-report tasks, before examining a series of studies in which participants were unable to report features of clearly visible items. These new data present a challenge for a hard "access-only" view of consciousness, as they appear to demonstrate that properties of our visual experience can remain unreportable. In so doing, we highlight the utility and underappreciated value of so-called failure to report tasks.This single-center, randomized, single-blind, parallel-controlled study aimed to analyze the changes in resting-state functional connectivity (RSFC) in young patients with a suicide attempt caused by depression before and after cognitive-behavioral therapy (CBT) combined with fluoxetine or fluoxetine alone by functional magnetic resonance imaging (fMRI). Before treatment, functional connectivity of the right subgenual anterior cingulate cortex (R-sgACC), left subgenual anterior cingulate cortex (L-sgACC) and right precuneus (R-PCu) was lower in depressed patients with a suicide attempt than that of healthy controls. After treatment, compared with the fluoxetine group, functional connectivity between the R-sgACC and left posterior cerebellar lobe in the CBT group was increased, while this group also showed increased RSFC between the L-sgACC and right anterior cingulate cortex/ medial prefrontal cortex. On the contrary, the functional connectivity between the R-PCu and right parietal lobe was reduced (P less then 0.001). It was also found there were some changes in different brain regions in pre- and post-treatment within both the CBT and MG group. The functional connectivity of the R-sgACC and the left posterior cerebellum lobe was negatively correlated with the SSI score. The functional connectivity of the R-PCu and right middle frontal cortex was negatively correlated with the HAMD score before treatment. Data from the 2018 National Cancer Institute Health Information National Trends Survey was used to assess noticing menu calorie information, using menu calorie information to change menu ordering behavior, and knowledge of daily calorie needs. Regression analysis of weighted data tested associations between individual characteristics and noticing and using menu calorie information. Nearly half of adults (44%) reported noticing menu calorie information. Women, younger individuals, those who seek health information, individuals with a BMI ≥ 30, and those with higher education or higher income were more likely to report noticing menu calorie information. Among adults who reported noticing menu calorie information, three-quarters responded by ordering less (e.g., fewer calories), which equates to about one-third of the population. About 36% of women and 42% of men lacked calorie knowledge. Men with, versus without, calorie knowledge were twice as likely to report noticing menu calorie information (adjusted OR 2.23 95% CI 1.51, 3.29). Findings suggest behavioral response to menu calorie information varies and most individuals who notice the information respond by ordering less in ways that could reduce caloric intake. Future analyses could compare noticing and using menu calorie information before and after menu labeling implementation to assess the effect of policy on population behaviors.The combination of ultrasound and chemotherapy has been proposed as a promising strategy to achieve a better anticancer therapeutic efficacy. Here we present a facile strategy to construct novel bifunctional nanodroplets as smart vehicles for ultrasound and pH responsive delivery of anticancer agents. PFH is used as core and chitosan/alginate complexes are used as the stable shells of the nanodroplets. The effects of alginate/chitosan ratio, and the amount of surfactant as well as PFH on the size, size distribution, and encapsulation efficiency of nanodroplets are systematically investigated with the optimized formulation identified. The release of the encapsulated doxorubicin hydrochloride can be triggered by changing the pH value of the surrounding environment and the exposure to ultrasound. The nanodroplets also show strong ultrasound contrast via droplet-to-bubble transition as demonstrated by B-mode ultrasound imaging. The hemolytic activity and cytotoxicity are further studied, revealing the biocompatibility of the nanodroplets. The in vivo antitumor results demonstrate that the prepared droplets show excellent antitumor therapeutic efficacy and outstanding tumor-targeting ability. The proposed alginate/chitosan stabilized PFH nanodroplets represent an important advance in fabricating multifunctional therapeutic materials with great promises in the applications of combined antitumor therapies.The presence of dyes in the aquatic environment as a result of anthropogenic activities, especially textile industries, is a critical environmental challenge that hinders the availability of potable water. Different wastewater treatment approaches have been used to remediate dyes in aquatic environments; however, most of these approaches are limited by factors ranging from high cost to the incomplete removal of the dyes and contaminants. Thus, the use of adsorption as a water treatment technology to remove dyes and other contaminants has been widely investigated using different adsorbents. This study evaluated the significance of chitosan as a viable adsorbent for removing dyes from water treatment. We summarised the literature and research results obtained between 2009 and 2020 regarding the adsorption of dyes onto chitosan and modified chitosan-based adsorbents prepared through physical and chemical processing, including crosslinking impregnation, grafting, and membrane preparation. Furthermore, we demonstrated the effects of various chitosan-based materials and modifications; they all improve the properties of chitosan by promoting the adsorption of dyes. Hence, the application of chitosan-based materials with various modifications should be considered a cutting-edge approach for the remediation of dyes and other contaminants in aquatic environments toward the global aim of making potable water globally available.Due to its low molecular weight and abundant functional groups, water-soluble lignin (WSL) is considered as a more potent antioxidant than traditional industrial lignin in biofields. However, few studies have been conducted to evaluate its intracellular and endogenous reactive oxygen species (ROS)-scavenging ability, especially for the intervention of ROS-related disease in vivo. In this work, WSL in bamboo autohydrolysate (WSL-BM) and wheat stalk autohydrolysate (WSL-WS) were isolated and characterized to comparably analyze their bioactivities. The composition analysis and NMR characterization showed that both WSL-BM and WSL-WS contained relatively similar components and substructures, but WSL-BM contained higher contents of phenolic OH groups. Both WSL samples exhibited excellent biocompatibility with the concentration below 50 μg/mL, while WSL-BM exhibited superior ROS-scavenging ability and ROS-related ulcerative colitis treatment potential at same concentration. In addition, WSL-BM also showed better performance in ameliorating inflammation and oxidative stress in RAW 264.
