This article discusses the development of 68Ga cold kit radiopharmacy, including technical issues, and regulatory aspects.
The aim of this study was to describe clinical features in different subtypes of chronic pulmonary aspergillosis (CPA)-simple aspergilloma (SA), chronic cavitary pulmonary aspergillosis (CCPA), chronic fibrosing pulmonary aspergillosis (CFPA), aspergillus nodule (AN), and subacute invasive aspergillosis (SAIA), respectively, and identify long-term prognosis of CPA.
We reviewed patients diagnosed with different subtypes of CPA from 2002 to 2020 at Nanjing Jinling Hospital, China. We analyzed the clinical and survival information of five different subgroups. A Cox regression model was used to explore proper antifungal duration and long-term survival factors of CCPA and SAIA.
A total of 147 patients with CPA were included, consisting of 11 SA, 48 CCPA, 5 CFPA, 12 AN, and 71 SAIA. The most common underlying pulmonary disease was pulmonary tuberculosis (
= 49, 33%), followed by bronchiectasis (
= 46, 31.3%) and chronic obstructive pulmonary disease (COPD) or emphysema (
= 45, 30.6%), while in SAIA and gnosis factors for SAIA and CCPA group, and BMI ≤ 20.0 kg/m
was independently associated with increased mortality (hazard ratio (
) 5.311, 95%
1.405-20.068,
= 0.014). Multivariable Cox regression indicated that surgery (
0.093, 95%
0.011-0.814,
= 0.032) and antifungal duration >6 months (
0.204, 95%
0.060-0.696
= 0.011) were related to improved survival.
The clinical features and laboratory test performance are different among SA, CCPA, CFPA, AN, and SAIA. Low BMI was an independent risk factor for survival. Selective surgery and antifungal duration over 6 months were associated with improved survival.

There are growing concerns regarding the emerging variants escaping vaccines or therapeutic neutralizing antibodies. In this study, we conducted an epidemiological survey to identify SARS-CoV-2 variants that are sporadically proliferating in vaccine-advanced countries. Subsequently, we created HiBiT-tagged virus-like particles displaying spike proteins derived from the variants to analyze the neutralizing efficacy of the BNT162b2 mRNA vaccine and several therapeutic antibodies. We found that the Mu variant and a derivative of the Delta strain with E484K and N501Y mutations significantly evaded vaccine-elicited neutralizing antibodies. This trend was also observed in the Beta and Gamma variants, although they are currently not prevalent. Although 95.2% of the vaccinees exhibited prominent neutralizing activity against the prototype strain, only 73.8 and 78.6% of the vaccinees exhibited neutralizing activity against the Mu and the Delta derivative variants, respectively. A long-term analysis showed that 88.8% of the vaccinees initially exhibited strong neutralizing activity against the currently circulating Delta strain; the number decreased to 31.6% for the individuals at 6 months after vaccination. Notably, these variants were shown to be resistant to several therapeutic antibodies. Our findings demonstrate the differential neutralization efficacy of the COVID-19 vaccine and monoclonal antibodies against circulating variants, suggesting the need for pandemic alerts and booster vaccinations against the currently prevalent variants. Both medical errors and suicidal behaviors are important problems for medical staff. Although the association between them was implied in previous studies, their association has not been built until now. In this study, the first aim was to build the association between perceived medical errors and suicidal ideation, and we also want to explore the mediating role of depression in the association between perceived medical errors and suicidal ideation among Chinese medical staff. In this study, we interviewed 3,338 medical staff in Chinese general hospitals. Questions about suicidal ideation (SI) and perceived medical errors were interviewed for medical staff. Depressive symptoms were evaluated by the Chinese version of Center for Epidemiologic Studies Depression Scale (CES-D). Social-demographic and occupation-related variables were also assessed in the data collection. This study found that the prevalence of suicidal ideation among medical staff was 9%. After the adjustment for controlling variables, suicidal ideation was significantly associated with a higher level of depression (OR = 1.10, p less then 0.001) and perceived medical errors (OR = 2.41, p less then 0.001). The other associated factors were female (OR = 2.21, p less then 0.001), religious belief (OR = 2.66, p less then 0.001), and weekly work hours (OR = 1.02 p less then 0.001). The mediating effect of depressive symptoms on the association between perceived medical errors and suicidal ideation was also supported in this study, and it can explain the 38.73% of the total effects of the perceived medical errors on suicidal ideation. The medical staff, with perceived medical errors, were in higher risk of suicidal ideation, and depressive symptoms can partially mediate the association between perceived medical errors and suicidal ideation. Infigratinib inhibitor For the medical staff who are experiencing medical errors, some scanning on their suicidal ideation and depressive symptoms are necessary to promote their mental health.
Gastric cancer (GC) is the fifth leading cancer around world. And prognosis of patients with GC is still undesirable. Our study aimed to explore potential prognostic biomarkers for patients with GC.
The clinical samples were collected from the Qinghai University Affiliated Hospital, which were subjected to the whole exome sequencing (WES). The other GC-related data were obtained from The Cancer Genome Atlas (TCGA) database. Cross analyses were done to determine the candidate genes. And the final mutated genes were determined by survival analyses, univariate and multivariate Cox regression analyses. CIBERSORT and GSEA were used for immune cell infiltration analysis and functional enrichment, respectively.
After cross analyses, 160 candidate-mutated genes were identified. And mutated ELP6 and PLIN5 were significantly independently correlated with the overall survival (OS) of patients with GC. Patients with GC with ELP6 and PLIN5 mutations had worse and better prognosis, respectively. Totally 5 types of immune cells were significantly differentially infiltrated in wild-type and mutated ELP6 and PLIN5 GC samples. In mutated ELP6 and PLIN5 GC samples, totally 7 and 11 pathways were significantly enriched, respectively.
The ELP6 and PLIN5 mutations were probably prognostic biomarkers for patients with GC.

Sarcopenia, an age-related degenerative disease, seriously affects the health and quality of life of the elder. The research of sarcopenia has changed dramatically around the world. This article aims to analyze global trends in this field over the past 20 years.
"Sarcopenia" was used as the search term to retrieve relevant publications from the WOS and PubMed databases. Co-occurrence, literature coupling, co-citation, and co-author analysis were performed by using the software VOS viewer. We analyzed the trends of sarcopenia research over the last 20 years from different aspects, such as the number of papers, total citations, average citations per item, h-index, research area, article types, institutions, country, journals, and funding.
We retrieved 13,421 research articles published on sarcopenia between 2001 and 2020. The results showed that the USA made the highest contributions to this field. Geriatrics gerontology is the most study classification of sarcopenia. Basic research on sarcopenia in geriaiseases will be the next research hotspot.Endocytosis is a mechanism that internalizes and recycles plasma membrane components and transmembrane receptors via vesicle formation, which is mediated by clathrin-dependent and clathrin-independent signaling pathways. Podocytes are specialized, terminally differentiated epithelial cells in the kidney, located on the outermost layer of the glomerulus. These cells play an important role in maintaining the integrity of the glomerular filtration barrier in conjunction with the adjacent basement membrane and endothelial cell layers within the glomerulus. An intact podocyte endocytic machinery appears to be necessary for maintaining podocyte function. De novo pathologic human genetic mutations and loss-of-function studies of critical podocyte endocytosis genes in genetically engineered mouse models suggest that this pathway contributes to the pathophysiology of development and progression of proteinuria in chronic kidney disease. Here, we review the mechanism of cellular endocytosis and its regulation in podocyte injury in the context of glomerular diseases. A thorough understanding of podocyte endocytosis may shed novel insights into its biological function in maintaining a functioning filter and offer potential targeted therapeutic strategies for proteinuric glomerular diseases.
Absolute blood volume (ABV) is a critical component of fluid status, which may inform target weight prescriptions and hemodynamic vulnerability of dialysis patients. Here, we utilized the changes in relative blood volume (RBV), monitored by ultrasound (BVM) upon intradialytic 240 mL dialysate fluid bolus-infusion 1 h after hemodialysis start, to calculate the session-specific ABV. With the main goal of assessing clinical feasibility, our sub-aims were to (i) standardize the BVM-data read-out; (ii) determine optimal time-points for ABV-calculation, "before-" and "after-bolus"; (iii) assess ABV-variation.
We used high-level programming language and basic descriptive statistics in a retrospective study of routinely measured BVM-data from 274 hemodialysis sessions in 98 patients.
Regarding (i) and (ii), we automatized the processing of RBV-data, and determined an algorithm to select the adequate RBV-data points for ABV-calculations. Regarding (iii), we found in 144 BVM-curves from 75 patients, that the averpartment modeling and comparison with reference methods of ABV-determination. Hopes are high that clinicians will be able to use ABV to inform target weight prescription, improving hemodynamic stability.
