Although the citrate biosynthesis is well studied, little is known about the transport mechanism by which citrate is exported. To gain better insight into this mechanism, we set out to identify a transporter involved in citrate export of Y. lipolytica. A total of five proteins were selected for analysis based on their similarity to a known citrate exporter, but neither a citrate transport activity nor any other phenotypic function could be attributed to them. Differential gene expression analysis of two strains with a distinct citrate productivity revealed another three putative transporters, one of which is YALI0D20196p. Disrupting YALI0D20196g in Y. lipolytica abolished citrate production, while extrachromosomal expression enhanced citrate production 5.2-fold in a low producing wildtype. Furthermore, heterologous expression of YALI0D20196p in the non-citrate secreting yeast Saccharomyces cerevisiae facilitated citrate export. Likewise, expression of YALI0D20196p complemented the ability to secrete citrate in an export-deficient strain of Aspergillus niger, confirming a citrate export function of YALI0D20196p. This report on the identification of the first citrate exporter in Y. lipolytica, termed Cex1, represents a valuable starting point for further investigations of the complex transport processes in yeasts.
Diagnostic delay >12 months is frequent in Crohn's disease [CD]. Recently, the International Organization for Inflammatory Bowel Disease [IO-IBD] developed a tool to identify early CD and reduce diagnostic delay. Subjects with an index ≥8 are more likely to have suspected CD (odds ratio [OR] 205, p <0.0001). We aimed to validate this questionnaire at the community level in patients seen by the general practitioners [GPs] in two large areas of Lombardy, Italy.

Consecutive adult patients referring to the GP were screened. The GPs administered the Red Flags [RF] questionnaire to the eligible patients. All patients were referred to the nearest participating centre to confirm or exclude the diagnosis of CD. Sensitivity, specificity, and positive and negative predictive values [PPV, NPV] of the RF index [RFI] were calculated. Patients lost to follow-up after the first gastroenterological visit were analysed using a non-responder imputation, assuming they were negative for CD diagnosis.

From November 2016 to November 2019, 112 patients were included. A total of 66 subjects [59%] completed the study after the first gastroenterological visit. The prevalence of CD was 3.6% in the study population [4/112]. The RF index had 50% sensitivity, 58% specificity, 4% PPV, and 97% NPV. A combined diagnostic strategy with faecal calprotectin [FC] [RFI ≥8 and/or FC >250 ng/g] resulted in significantly improved accuracy sensitivity 100% [29-100%], specificity 72% [55-85%], PPV = 21% [5-51%], NPV = 100% [88-100%].

However, in forensic or archaeological contexts, there is the possibility that the fibula is the only bone available for analysis. The present study applied geometric morphometrics to evaluate the potential of using fibular morphology for assessment of sex and ancestry. Three-dimensional datasets of 1609 fibulae of adult South Africans (412 females, 507 males) were digitized. Datasets were submitted to Generalized Procrustes Analysis for rotation and scaling to a common centroid. Mean centroid sizes were compared using parametric testing, and morphological variation was assessed using multivariate analyses. Discriminant Function Analysis coupled with leave-one-out cross-validation testing was used to assess classification accuracy of sex and ancestry based on these shape variations. Fibulae were smaller in females than males, but show insufficient shape variation to distinguish the sexes. Fibulae of Coloured individuals were relatively and absolutely smaller than those of the Black and White ancestry groups, likely due to contributions of small-bodied groups to the genetic composition of this group. Based on shape variation, ancestry estimations of 72.4-77.2% were obtained. Evaluation of variation according to sex and ancestry combined still produced insufficient distinction between the sexes and decreased the accuracy of ancestry classification. These results show that morphological variation of the fibula may not be useful for sex estimation, but provides reasonable accuracies for ancestry estimations, making it particularly useful in cases where only the fibula is available for analysis.ObjectiveMenarcheal age is the starting point of reproductive life and could be a marker of health and socioeconomic status of a population.The aim of this study was to assess the temporal pattern in variability of menarcheal age from 2001 to 2019 and the relationship with biological factors and educational level of parents. Subjects and methods A cross-sectional anthropometric survey was conducted in three intervals 2001-2004, 2011-2014 and in 2019. In total, 5201 adolescent girls from Vojvodina (North Serbia) aged 11-18 were surveyed, 3272 (62.91%) of them with menarche. Data on the age at menarche was gathered applying status quo method and the age at menarche was calculated by probit analysis. The surveyed traits included height, weight and Body mass index (BMI). check details Results Mean menarcheal age declined from 12.60 ± 1.11 years to 12.33 ± 1.20 years during the period from 2001 to 2019. Within three investigated periods variance decreased after the first decade from 1.23 year2 to 1.09 year2, and then increased to 1.44 year2. On average, postmenarcheal girls were 4.36 cm taller, 8.03 kg heavier, and their BMI was 1.96 kg/m2 higher than in premenarcheal girls of the same calendar age. An inverse effect of BMI and mother's education on the menarcheal age was detected, but the interaction between the variables was not statistically significant. Conclusion These results indicate continuing decrease in menarcheal age in girls from Vojvodina. Menarche is closely associated with BMI and it appears that this factor effects the age at menarche irrespective of the parents' education.This paper focuses on an unusual Pre-Hispanic burial, 11-12th century cal AD, from Fuerteventura Island. The study first uses forensic anthropology techniques to characterise the individual's injuries before turning to archaeological and ethno-historical records of the indigenous populations to identify the potential scenarios explaining the fatal injuries. MicroRNAs (miRNAs) are small noncoding RNAs that play key roles in the regulation of biological pathways, from development to cellular stress responses. In this study, we performed genome-wide miRNA expression profiling in primary human dermal fibroblasts 20 weeks after chronic and acute (non-chronic) treatments to four AuNPs with different shapes and surface chemistries at a low dose. The exposure condition and AuNP surface chemistry had a significant impact on the modulation of miRNA levels. In addition, a network-based analysis was employed to provide a more complex, systems-level perspective of the miRNA expression changes. In response to the stress caused by AuNPs, miRNA co-expression networks perturbed in cells under non-chronic exposure to AuNPs were enriched for target genes implicated in the suppression of proliferative pathways, possibly in attempt to restore cell homeostasis, while changes in miRNA co-expression networks enriched for target genes related to activation of proliferative and suppression of apoptotic pathways were observed in cells chronically exposed to one specific type of AuNPs. In this case, miRNA dysregulation might be contributing to enforce a new cell phenotype during stress. Our findings suggest that miRNAs exert critical roles in the cellular responses to the stress provoked by a low dose of NPs in the long term and provide a fertile ground for further targeted experimental studies.A fully-automated and portable leukocyte separation platform was developed based on a new type of inertial microfluidic device, multi-dimensional double spiral (MDDS) device, as an alternative to centrifugation. By combining key innovations in inertial microfluidic device designs and check-valve-based recirculation processes, highly purified and concentrated WBCs (up to >99.99% RBC removal, ∼80% WBC recovery, >85% WBC purity, and ∼12-fold concentrated WBCs compared to the input sample) were achieved in less than 5 minutes, with high reliability and repeatability (coefficient of variation, CV less then 5%). Using this, one can harvest up to 0.4 million of intact WBCs from 50 μL of human peripheral blood (50 μL), without any cell damage or phenotypic changes in a fully-automated operation. Alternatively, hand-powered operation is demonstrated with comparable separation efficiency and speed, which eliminates the need for electricity altogether for truly field-friendly sample preparation. The proposed platform is therefore highly deployable for various point-of-care applications, including bedside assessment of the host immune response and blood sample processing in resource-limited environments.
