In this study, using a murine model established by intranasal infection with RSV, we found that acute RSV infection induced the production of a neurotransmitter-neuromedin U (NMU) in the lungs of RSV-infected mice and up-regulated the expression of neuromedin U receptor 1 (NMUR1) on ILC2s. Moreover, in vivo administration of NMU exacerbated RSV-induced airway inflammation by promoting the proliferation and activation of pulmonary ILC2s via the NMUR1 pathway, which involved PI3K, MEK, and NFAT signaling proteins. Furthermore, pulmonary neurons responded to the stimulation of RSV infection and secreted NMU in a TLR4- and TLR7-dependent manner. Collectively, our data suggest that NMU is a powerful neuropeptide to activate ILC2s, highlighting the critical regulatory effects of neurotransmitters on antiviral, inflammatory, and tissue homeostasis at the mucosal barrier during a viral respiratory infection.
We investigated the impact of changes in lifestyle habits on colorectal cancer (CRC) risk in a multicountry European cohort.

We used baseline and follow-up questionnaire data from the European Prospective Investigation into Cancer cohort to assess changes in lifestyle habits and their associations with CRC development. We calculated a healthy lifestyle index (HLI) score based on smoking status, alcohol consumption, body mass index, and physical activity collected at the 2 time points. HLI ranged from 0 (most unfavorable) to 16 (most favorable). We estimated the association between HLI changes and CRC risk using Cox regression models and reported hazard ratios (HR) with 95% confidence intervals (CI).

Among 295,865 participants, 2,799 CRC cases were observed over a median of 7.8 years. The median time between questionnaires was 5.7 years. Each unit increase in HLI from the baseline to the follow-up assessment was associated with a statistically significant 3% lower CRC risk. Among participants in the top tertile at baseline (HLI > 11), those in the bottom tertile at follow-up (HLI ≤ 9) had a higher CRC risk (HR 1.34; 95% CI 1.02-1.75) than those remaining in the top tertile. Among individuals in the bottom tertile at baseline, those in the top tertile at follow-up had a lower risk (HR 0.77; 95% CI 0.59-1.00) than those remaining in the bottom tertile.

In this work, we tailored a hybrid functional layer consisting of Bi/MgCl2/polytetrahydrofuran (PTHF) by an in situ THF polyreaction during the reaction of the Mg anode with BiCl3 solution. The introduction of PTHF inhibits the growth of Bi particles and fills the layer interstice with MgCl2-containing PTHF, improving the structural integrity of the functional layer and insulation between the electrolyte and Mg anode. As a result, compared to a simply modified Bi/MgCl2 layer, the Bi/MgCl2/PTHF functional layer exhibits a lower polarization voltage of 0.25 V and longer cycling life of more than 2000 h at 0.1 mA cm-2. Mechanism analysis shows that Mg is plated on the surface of Bi particles within the layer. In the bleomycin model, nintedanib drove expression of IL-4/IL-13 associated genes important for tissue regeneration and repair at early and late time points in lung macrophages. These findings were replicated in vitro in mouse primary bone marrow derived macrophages exposed to IL-4/IL-13 and nintedanib. In addition, nintedanib promoted expression of IL-4/IL-13 pathway genes in human macrophages in vitro. The molecular mechanism was connected to inhibition of the CSF1 receptor (CSF1R) in both human and mouse macrophages. Moreover, nintedanib counterbalanced the effects of TNF on IL-4/IL-13 in macrophages to promote expression of IL-4/IL-13 regulated tissue repair genes in fibrotic contexts in vivo and in vitro.
