We suggest that patients on the wait-list for a deceased human kidney graft who are unlikely to receive one due to long waiting times are those for whom kidney xenotransplantation might first be considered. The potential risk of infection, public attitudes to xenotransplantation, and ethical, regulatory, and financial aspects are briefly addressed.Genome-wide sequencing (GWS) is increasingly being used in neonatal intensive care units. While studies have explored its clinical utility, little is known about parental experiences with this testing post-return of results. We conducted a qualitative study, using an interpretive description framework and thematic analysis, to gain further insight into parents' perceptions of the value and utility of GWS for their infant. We sought to explore whether parents' perceptions differ if their child received a diagnosis or not, and whether their child is living or deceased. Semi-structured, telephone interviews were conducted with parents of infants who had rapid exome sequencing while in the neonatal intensive care unit at BC Women's Hospital in Vancouver, Canada. Interviews addressed perceived benefits and harms of GWS and included an evaluation of decisional regret. Parents of 27 probands were approached and 14 (52%; 13 mothers and 1 father) participated in interviews. On average, 26 months had elapsed from the time of results to the interview. Six themes were identified. Firstly, parents had a positive regard for GWS. The results of GWS helped provide context for their child's admission to the NICU, and all parents experienced relief following receiving the results. A diagnosis by GWS enabled parents to picture the future, form connections with other parents, and coordinate their child's care. Lastly, some parents experienced discomfort with the concept of a genetic diagnosis, and interestingly felt lack of a genomic diagnosis indicated a reduced severity of their infant's condition. Decisional regret post-results was found to be low. Our results highlight how parents cope with the results of GWS and suggest that a genetic counselor can have an important role in helping families understand and adjust to these results in the neonatal intensive care unit.Antimicrobial peptides (AMPs) are important components of the innate immune system and are involved in skin protection against environmental insults and in wound healing. GSK-LSD1 Herein, we assessed the gene expression of chemerin (Rarres2), cathelicidin CRAMP (Camp), and three β-defensins (Defb1, Defb3, and Defb14) in mouse skin during light/dark cycle (LD 1212) and constant darkness (DD). Next, we examined the survival of bacteria applied on the skin at specific times during the day. We found that the expression of Rarres2, Camp, and Defb1 was the highest at 4 h after the beginning of darkness, during high activity of mice. These rhythms, however, were not maintained under DD in the skin but were present in the liver. This indicated that in the case of skin, a circadian input was masked by daily changes of light in the environment. In contrast, Defb3 and Defb14 showed the highest mRNA levels when the mice slept, and these rhythmic mRNA oscillations were maintained under DD. This shows that Rarres2, Camp, and Defb1 levels in the skin are correlated with high locomotor activity in mice and they are controlled by daily changes of light and dark. Alternatively, oscillations in the mRNA levels of Defb3 and Defb14 seem to protect skin and heal wounds during sleep. These rhythms are maintained under DD, indicating that they are regulated by a circadian clock. Our study suggests that daily AMP expression affects the survival of bacteria on the surface of skin, which depends on the phase of AMP cycling.Chemically enhanced primary treatment (CEPT) with ferrate(VI), a multifunctional treatment agent, was investigated for the treatment of municipal wastewater in a laboratory-scale study. The treatment performance was evaluated at different ferrate(VI) doses (0.0-9.0 mg/L as Fe) and pH (6.0 and 7.5). The optimal removals of total suspended solids (TSS) (52%), total chemical oxygen demand (COD) (34%), and total phosphorus (47%) were achieved at the highest ferrate(VI) dose (9.0 mg/L as Fe) and the weakly alkaline condition (pH 7.5). The pollutant abatements principally ascribed to the formation of large-sized aggregate and ensuing sedimentation fell within the reported ranges of CEPT with traditional coagulants. However, different from conventional CEPT, ferrate(VI) appreciably removed recalcitrant dissolved organic phosphorus (49%) and simultaneously inactivated total coliform (3.30 log removal) and Escherichia coli (3.67 log removal) at 9.0 mg/L Fe(VI) and pH 7.5. The CEPT with ferrate(VI) offers an innovative alternative for improving municipal wastewater treatment. PRACTITIONER POINTS Ferrate(VI) represents a promising agent for chemically enhanced primary treatment (CEPT) of municipal wastewater. CEPT with ferrate(VI) can effectively alleviate TSS, total COD, and total P via the formation of large-sized aggregates and ensuing sedimentation. Ferrate(VI) can substantially remove recalcitrant dissolved organic phosphorus in municipal wastewater. Different from other CEPT coagulants, ferrate(VI) can appreciably inactivate bacterial indicators during CEPT. Higher ferrate(VI) dose and weakly alkaline pH favor the performance of ferrate(VI) CEPT.Bread wheat (Triticum aestivum) is an allohexaploid that was formed via two allopolyploidization events. Growing evidence suggests histone modifications are involved in the response to 'genomic shock' and environmental adaptation during polyploid formation and evolution. However, the role of histone modifications, especially histone H3 lysine-27 dimethylation (H3K27me2), in genome evolution remains elusive. Here we analyzed H3K27me2 and H3K27me3 profiles in hexaploid wheat and its tetraploid and diploid relatives. Although H3K27me3 levels were relatively stable among wheat species with different ploidy levels, H3K27me2 intensities increased concurrent with increased ploidy levels, and H3K27me2 peaks were colocalized with massively amplified DTC transposons (CACTA family) in euchromatin, which may silence euchromatic transposons to maintain genome stability during polyploid wheat evolution. Consistently, the distribution of H3K27me2 is mutually exclusive with another repressive histone mark, H3K9me2, that mainly silences transposons in heterochromatic regions.
