Here, we utilized the small molecule 6-bromoindirubin-3'-oxime (BIO), an activator of Wnt/β-catenin signaling, and investigated whether Wnt/β-catenin signaling regulated the proliferation and acinus-like structure formation of goat mammary epithelial cells (GMECs). We showed that isolated GMECs displayed the typical epithelial cobblestone morphology and expressed specific markers of mammary epithelial cells. BIO inhibited the proliferation of GMECs and decreased the expression of proliferation marker c-myc and cell cycle protein cyclin D1. However, the ability of GMECs to form spheroids was accelerated, and the level of E-cadherin mRNA was upregulated with BIO treatment. E-cadherin showed a bright cytomembrane with DMSO treatment, yet E-cadherin was present in cytomembrane and cytoplasm in GMECs with BIO treatment. Meanwhile, BIO increased the protein level of β-catenin and enhanced the translocation of β-catenin into the nucleus in GMECs. Furthermore, the mRNA level of Axin2 was also upregulated. The objective of this study was to investigate the exosomal miRNAs that regulate lipopolysaccharide (LPS)-induced inflammation in dairy cow mammary alveolar (Mac-T) cells. We found two exosome miRNAs upregulated and five exosomal miRNAs downregulated, respectively, in the LPS-stimulated Mac-T cells. MiR-193b-5p was upregulated 6.3-fold in the LPS-stimulated cell-derived exosome. Target prediction results showed that nuclear factor kappa B (NF-κB) inhibitor delta (NFKBID), transforming growth factor-beta 1 induced transcript 1 (TGFB1I1), interleukin 22 (IL-22), TNF receptor superfamily member 11b (TNFRSF11B), and Janus kinase 3 (JAK3) might be the main target genes of miR-193b-5p. After treatment of Mac-T cells with the miR-193b-5p mimic, the phosphorylation levels of inhibitor of nuclear factor-kappa Bα (IκBα) and p65 were upregulated, the level of IL-6 mRNA was upregulated, and IL-1β, TNF-α, and TGF-β mRNA levels were downregulated. After treatment of Mac-T cells with miR-193b-5p inhibitor, the phosphorylation levels of IκBα and p65 were downregulated. In summary, these findings provide strong evidence that exosomal miR-193b-5p could be a regulator of LPS-induced inflammation in Mac-T cells and reveal a new role of exosomal miRNAs in regulating dairy cow mastitis.
Ampullary neuroendocrine tumors (NETs) make up < 1% of all gastroenteropancreatic NETs, and information is limited to case series. This study compares patients with ampullary, duodenal, and pancreatic head NETs.

The National Cancer Database (2004-2016) was queried for patients with ampullary, duodenal, and pancreatic head NETs. Survival was evaluated using Kaplan-Meier analysis and Cox regression.

Overall, 872, 9692, and 6561 patients were identified with ampullary, duodenal, and pancreatic head NETs, respectively. Patients with ampullary NETs had more grade 3 tumors (n=149, 17%) than patients with duodenal (n=197, 2%) or pancreatic head (n=740, 11%) NETs. Patients with ampullary NETs had more positive lymph nodes (n=297, 34%) than patients with duodenal (n=950, 10%) or pancreatic head (n=1513, 23%) NETs. On multivariable analysis for patients with ampullary NETs, age (hazard ratio [HR] 1.03, p<0.0001), Charlson-Deyo score of 2 (HR 2.3, p=0.001) or ≥3 (HR 2.9, p=0.013), grade 2 (HR 1.9, p=0.007) or grade 3 tumors (HR 4.0, p<0.0001), and metastatic disease (HR 2.0, p=0.001) were associated with decreased survival. At 5years, the overall survival (OS) for patients with ampullary, duodenal, and pancreatic head NETs was 59%, 71%, and 50%, respectively (p<0.0001), whereas the 5-year OS for patients with ampullary, duodenal, and pancreatic head NETs who underwent surgery was 62%, 78%, and 76%, respectively (p<0.0001).

Hepatitis B virus (HBV) infection is a significant global health problem and > 42-52% of patients are infected during perinatal period. Tenofovir alafenamide fumarate (TAF) and tenofovir disoproxil fumarate (TDF) have been widely recognized as the main compounds used for antiviral treatment of hepatitis B. The present study evaluated the efficacy and safety of TAF in reducing HBV vertical transmission.

A total of 72 pregnant women, who met the inclusion criteria, were randomly divided into the TDF (300mg/day, n = 36) and TAF (25mg/day, n = 36) groups. Clinical and laboratory data were analyzed and compared between the two groups.

No significant differences in alanine aminotransferase, total bilirubin, blood creatinine and blood urea nitrogen levels were noted between the two groups after treatment. The serum HBV DNA viral load and hepatitis B e antigen (HBeAg) levels of the two groups were significantly decreased following treatment, whereas the difference between the two groups was not statistically significant. The levels of urine retinol-binding protein and β2-microglobulin had no significant change after TAF treatment (p > 0.05), but increased significantly after TDF treatment (p < 0.05). All drug concentrations were undetectable in umbilical cord blood (UCB) and breast milk samples of the TAF group, while the drug concentration of UCB and breast milk samples in the TDF group was 2.98 ± 1.44 and 19.16 ± 15.26ng/ml, respectively. All infants were tested negative for serum hepatitis B surface antigen, HBV DNA, and HBeAg.

Papillary thyroid cancer (PTC) is the predominant histological type of thyroid cancer, accounting for 80% of thyroid cancers. MiR-181a is a novel microRNA that is usually upregulated in multiple cancers. link2 This study aims to explore the role and underlying mechanism of miR-181a in PTC.

CCK8 and Transwell assays were performed to evaluate cell viability and migration. The mRNA level of miR-181a and KLF15 was calculated by qRT-PCR. The protein level of E-Cadherin, N-Cadherin and GAPDH was evaluated by western blot. Dual luciferase assay was conducted to validate that miR-181a directly targeting the 3'-UTR of KLF15 mRNA in TPC-1 cells.

We observed that miR-181a was overexpressed and KLF15 was low expressed in PTC tissues and cell lines. We hypothesized that L2ers would make use of semantic and orthographic information parafoveally. Using the gaze contingent boundary paradigm, we manipulated six parafoveal masks in a sentence (Mark found th*e wood for the fire; * indicates the invisible boundary) identical word mask (wood), English orthographic mask (wook), English string mask (zwwl), German mask (holz), German orthographic mask (holn), and German string mask (kxfs). We found an orthographic benefit for L1ers and L2ers when the mask was orthographically related to the target word (wood vs. wook) in line with previous L1 research. English L2ers did not derive a benefit (rather an interference) when a non-cognate translation mask from their L1 was used (wood vs. holz), but did derive a benefit from a German orthographic mask (wood vs. holn). While unexpected, it may be that L2ers incur a switching cost when the complete German word is presented parafoveally, and derive a benefit by keeping both lexicons active when a partial German word is presented parafoveally (narrowing down lexical candidates). To the authors' knowledge there is no mention of parafoveal processing in any model of L2 processing/reading, and the current study provides the first evidence for a parafoveal non-cognate orthographic benefit (but only with partial orthographic overlap) in sentence reading for L2ers. We discuss how these findings fit into the framework of bilingual word recognition theories.
