Hs3st3b1-/- mice have reduced intact SMG branching morphogenesis and reduced 3-O-sulfated HS in the basement membrane. The overall glycosaminoglycan composition of adult control and KO mice were similar, although HS disaccharide analysis showed increased N- and non-sulfated disaccharides in Hs3st3a1-/- HS. Analysis of adult KO gland function revealed normal secretory innervation, but without stimulation there was an increase in frequency of drinking behavior in both KO mice, suggesting basal salivary hypofunction, possibly due to myoepithelial dysfunction. Understanding how 3-O-sulfation regulates myoepithelial progenitor function will be important to manipulate HS-binding growth factors to enhance tissue function and regeneration.The disorganized and inefficient tumor vasculature is a major obstacle to the delivery and efficacy of antineoplastic treatments. Antiangiogenic agents can normalize the tumor vessels, improving vessel function and boosting the distribution and activity of chemotherapy. The type III repeats (T3R) domain of thrombospondin-1 contains different potential antiangiogenic sequences. We therefore hypothesized that it might affect the tumor vasculature. Ectopic expression of the T3R domain by the tumor cells or by the host, or administration of recombinant T3R, delayed the in vivo growth of experimental tumors. Tumors presented marked reorganization of the vasculature, with abundant but smaller vessels, associated with substantially less necrosis. Mechanistically, the use of truncated forms of the domain, containing different active sequences, pointed to the FGF2/FGFR/ERK axis as the target for T3R activity. Along with reduced necrosis, the expression of T3R promoted tumor distribution of chemotherapy (paclitaxel), with a higher drug concentration and more homogeneous distribution, as assessed by HPLC and MALDI imaging mass spectrometry. T3R-expressing tumors were more responsive to paclitaxel and cisplatin. This study shows that together with its known role as a canonical inhibitor of angiogenesis, thrombospondin-1 can also remodel tumor blood vessels, affecting the morphological and functional properties of the tumor vasculature. And most practitioners believe that the plant protein is most potential for FM surrenal. While high plant protein feed caused some common problems in aquatic livestock the absorption rate and growth rate are decreased, and even caused digestive tract inflammation. In present study, the inflence of high plant protien feed in Trachinotus ovatus was investigated using illumina HiSeqTM2000 based RNA-Seq. By comparing the two groups of cDNA libraries developed from high plant protien based diet or FM based diet fed T. ovatus livers, 836 unigenes were significantly upregulated, and 345 were significantly down regulated. KEGG analysis indicated that the differentially expressed genes (DEGs) are mainly metabolic-related genes. It was found that more than 28 DGEs beloned to the protein metabolism and absorption, lipid biosynthesis or other metabolic pathways. It indicated that high plant protein based diet had broad effects on metabolism on T. ovatus. There were also more DEGs belong to immune-related signaling pathways, include genes were involved in pathpathogen resistance and genes related to immunity system. These DEGs provided useful clues to explore the mechanisms that high plant protein based diet caused side effects on T. ovatus. These results improved our current understanding of the response of high plant protein based diet in T. ovatus, and outstanding the reasons of the side effect caused by high protein based diet.The present study aimed to investigate the effects of a mixture of three medicinal plants extracts (COP common mallow (Malvae sylvestris), oregano (Origanum vulgare), and Persian shallot (Allium hirtifolium boiss)) on growth, hematological indices, immunological parameters, liver antioxidant, and digestive enzyme activity of common carp (Cyprinus carpio). Accordingly, 540 common carp fingerlings were randomly allotted to 18 fiberglass tanks, including six treatments each in triplicate. Fish were fed with experimental diets supplemented with 0, 0.5, 1, 2, 3, and 5% of the herbal mixture. After 60 days, the effects on growth performance, antioxidant, and immunological parameters were evaluated. According to the results, the highest final weight was observed in the fish fed 2 and 3% of the herbal mixture. Also, the feed conversion ratio (FCR) showed a significant decrease in fish treated with the herbal mix, so that the lowest FCR was observed in the treatment 1%. Hematological examinations such as red blood celgher mucosal and intestinal immunity parameters, including total immunoglobulin (total Ig), lysozyme, alternative complement activities (ACH50), protease, and ALP. According to the results, supplementing the diet with a mixture of the COP can significantly improve growth parameters, stimulate the innate immune system, and improve antioxidant defenses and liver health of common carp.The use of yeasts as a dietary additive for fish can act as a source of nutrients and as an immunostimulant. This work aimed to evaluate the effects of the fermented biomass of the yeast Yarrowia lipolytica as a food additive on zootechnical and hematological parameters, and on immune response in the plasma and kidney of Nile Tilapia (Oreochromis niloticus). After supplementation with 3, 5, and 7% of the yeast biomass for 35 days, the blood and tissues of the animals of each experimental group were collected for analysis. The addition of this biomass in the feed promoted an improvement of zootechnical parameters in tilapia. There was also a rise in the number of neutrophils (groups with 3, 5, and 7%) and monocytes (group 3, 5, and 7%) compared with the control group. Moreover, there was an increase in the levels of lysozyme, myeloperoxidase, and nitrite/nitrate content in the blood of animals fed with yeast biomass. On the other hand, there were no observed alterations in survival and hematological parameters of animals fed with yeast biomass. In the analysis of the kidney, the addition of biomass in feed promoted an increase in levels of myeloperoxidase (group with 3%) but did not alter the levels of lysozyme and nitrite/nitrate content. In conclusion, this study demonstrated that Y. lipolytica had growth and immunostimulatory effects on Nile tilapia. These findings strongly suggest the potential application of a Y. lipolytica-based immunostimulant for tilapia aquaculture.ATG12, a core autophagy protein, forms a conjugate with ATG5 to promote the formation of autophagosome membrane, and plays an important role in antiviral immunity. However, little is known about the function of ATG12 in fish. Here, we cloned the open reading frame (ORF) of large yellow croaker (Larimichthys crocea) ATG12 (LcATG12), which is 354 nucleotides long and encodes a protein of 117 amino acids. The deduced LcATG12 possesses a conserved APG12 domain (residues 31 to 117), and shares 91.45% identities with ATG12 in orange-spotted grouper (Epinephelus coioides). LcATG12 was constitutively expressed in all examined tissues, with the highest level in intestine. Its transcript was also detected in primary head kidney granulocytes (PKG), primary head kidney macrophages (PKM), primary head kidney lymphocytes (PKL), and large yellow croaker head kidney (LYCK) cell line, and was significantly up-regulated by poly(IC). LcATG12 was regularly distributed in both cytoplasm and nucleus of LYCK and epithelioma papulosum cyprinid (EPC) cells. Overexpression of LcATG12 in EPC cells significantly inhibited the replication of spring viremia of carp virus (SVCV). Further studies reveled that LcATG12 could induce the occurrence of autophagy in LYCK cells. Furthermore, overexpression of LcATG12 in LYCK cells increased the expression levels of large yellow croaker type I interferons (IFNs, IFNc, IFNd, and IFNh), IFN regulatory factors (IRF3 and IRF7), and IFN-stimulated genes (PKR, Mx, and Viperin). All these data indicated that LcATG12 plays a role in the antiviral immunity possibly by inducing both autophagy and type I IFN response in large yellow croaker.This study aims to evaluate the effects of dietary α-lipoic acid (α-LA) on bioaccumulation, oxidative stress, apoptosis, and inflammation in Channa argus after 28 d of lead (Pb) exposure. A total of 300 fish were divided into five groups the first group was the control group and the other four groups were exposed to waterborne Pb (800 ppb) and fed α-LA diets supplemented with 0, 300, 600, and 900 mg/kg. The results demonstrated that dietary α-LA effectively reduced the Pb accumulation in the liver, kidney, gill, intestine, and muscle of C. argus after exposure to Pb. Meanwhile, dietary α-LA reversed alterations in the biochemical parameters (Alanine aminotransferase (ALT), lactate dehydrogenase (LDH), aspartate aminotransferase (AST), blood urea nitrogen (BUN), cortisol (COR), and creatinine (CRE)) and immunity parameters (myeloperoxidase (MPO), complement 3 (C3), lysozyme (LYS), complement 4 (C4), C-reactive protein (CRP), and immunoglobulin M (IgM)) in the serum of fish caused by Pb. Pb-induced reduction of antioxidant enzyme activities (Catalase (CAT), glutathione reductase (GR), superoxide dismutase (SOD), glutathione (GSH), glutathione peroxidase (GSH-Px)) was inhibited by dietary α-LA.
