The ABO(H) secretor status is controlled by FUT2-encoded α(1,2)fucosyltransferase (Se enzyme) activity. Three SNPs of FUT2, 302C>T (rs200157007), 385A>T (rs1047781), and 428G>A (rs601338), cause three major variants of nonsecretor (se) or weak-secretor (Se
) alleles. Evidence has been accumulating that suggests the secretor status is associated with various conditions including infectious diseases but a robust multiplex method for assaying relatively large-scale samples to determine the genotype of these three SNPs simultaneously has not been developed yet.
By combined usage of two Eprobes and a dual-labeled fluorescence probe, we developed a real-time PCR, followed by triplex probe-based fluorescent melting-curve analysis (FMCA) for genotyping of 302C>T, 385A>T, and 428G>A of FUT2 in a single tube.
Three genotypes of each of three variants of FUT2 were accurately determined by the triplex probe-based FMCA. We then validated this method using genomic DNA samples of 47 Bangladeshis, and the results obtained by using this method were fully concordant with those by previous Sanger sequencing.
Since the present single triplex probe-based FMCA is robust, fast, and cost-effective, we are able to effectively estimate the secretor status of subjects on a large scale in many populations around the world.

Sulfate and chloride salts commonly used and present in natural wastewater influence the electrochemical degradation process. In this study, the effect of electrolyte composition on the active sulfate species (SO4●⁻ and S2O82⁻) formation, benzotriazole degradation-a model organic compound, and chlorinated by-products distribution have been investigated while using a boron-doped diamond (BDD) anode. Different Na2SO4NaNO3 and Na2SO4NaCl ratios with constant conductivity of 10 mS/cm were used in the experiments and applied anode potential was kept constant at 4.3 V vs. Ag/AgCl. The electrogenerated SO4●⁻ and S2O82⁻ formation were faster in 101 and 21 Na2SO4NaNO3 ratios than in the 10 ratio. The ●OH-mediated SO4●⁻ production has prevailed in 101 and 21 ratios. However, ●OH-mediated SO4●⁻ production has hindered the 10 ratio due to excess chemisorption of SO42⁻ on the BDD anode. Similarly, the faster benzotriazole degradation, mineralization, and lowest energy consumption were achieved in the 101 Na2SO4NaNO3 and Na2SO4NaCl ratio. Besides, chlorinated organic by-product concentration (AOX) was lower in the 101 Na2SO4NaCl ratio but increased with the increasing chloride ratio in the electrolyte. LC-MS analysis shows that several chlorinated organic transformation products were produced in 01 to 21 ratio, which was not found in the 101 Na2SO4NaCl ratio. Herein, water hyacinth [(WH) (Eichhornia crassipes)]-derived metal-free biochar (WHBC) prepared at 300-900 °C was used to promote the calcium peroxide (CP)-mediated remediation of 4-NP-contaminaed sediments. At [CP] = 4.37 × 10-4 M, [WHBC] = 1.5 g L-1, and pH = 6.0, the degradation of 4-NP was 77% in 12 h following the pseudo-first order rate law with rate constant (kobs) of 4.2 × 10-2 h-1. The efficient 4-NP degradation performance and reaction mechanisms of the WHBC/CP system was ascribed to the synergy between the reactive species (HO• and 1O2) at the WHBC surface on which there were abundant electron-rich carbonyl groups and defects/vacancies in the catalyst structure provides active sites, and the ability of the graphitized carbon framework to act as a medium for electron shuttling. According to microbial community analysis based on amplicon sequence variants, bacteria of the genus Solirubrobacter (Actinobacteria phylum) were dominant in WHBC/CP-treated sediments and were responsible for the biodegradation of 4-NP. The results showed great promise and novelty of the hydroxyl radical-driven carbon advanced oxidation processes (HR-CAOPs) that relies on the value-added utilization of water hyacinth for contaminated sediment remediation in achieving circular bioeconomy.Microplastics (MPs) contamination in rivers and lakes is of paramount environmental importance as freshwater systems transport MPs from land to ocean. However, information regarding the spatio-vertical distributions of MPs in rivers, and their associations with surrounding industrial activities, is scarce and unclear. This study investigated MPs in the Taipu River, where there is a highly developed textile industry in Yangtze River Delta, China. Results showed a widespread occurrence of MPs particles with concentrations in the range of 0.65-6.07 items/L and 0.30-3.63 items/L in surface and bottom waters. A higher abundance of MPs was observed in surface waters than in bottom waters (t = 5.423, p = 0.024). The MPs distributions varied markedly in space, with the highest abundances being found in textile manufacturing zones as a consequence of industrial release (F = 14.642, p less then 0.001). Transparent fibers were the major MPs compositions with 100-500 μm in size. Polyethylene terephthalate (PET) accounted for 71.4% and 59.73% of the total MPs identified in surface and bottom water, respectively. These PET polymers were predominantly presented in "fibrous" shapes, further reflecting the point sources of textile wastewater. Moreover, polyvinyl acetate (PVAC), used as fabric coating and resin matrix to form nonwoven fabrics, was firstly highlighted at a watershed scale. Although risk assessments revealed a light to moderate risks of MPs in the Taipu River, textile wastewater appears to cause a high "grey water" footprint and increase the risks of MPs pollution from textile life-cycle production. This study bridged gaps between field data and policy-making for MPs control and shed insight into the cleaner production of the textile industry.Marine aquaculture is considered a potential source of microplastics (MPs). MPs can induce oxidative stress and damage in marine species. In this study we evaluated the impact of MPs intake in the commercial fish, Sparus aurata, from aquaculture facilities and the antioxidant response associated to this MPs ingestion in caged specimens for 120 days. Sampling was carried out at the beginning of the study (T0), at 60 days (T60) and at 120 days (T120). At each sampling stage, gastrointestinal tract, blood, plasma, liver and muscle samples were obtained to analyse MPs intake (gastrointestinal tract), oxidative stress markers (blood, plasma and liver) and plasticizers bioaccumulation (muscle). Fish sampled at T60 presented the highest MPs intake and plasticizers accumulated in muscle over time, but with a different pattern according to type bisphenols and phthalates. This indicates MPs ingestion induces a differential tissue response in S. aurata. Similarly, stress biomarkers presented a differential response throughout the study, depending on the analysed tissue. In the case of oxidative damage markers, for malondialdehyde (MDA) an increase throughout the study was observed both in liver and blood cells but with a progressive decrease in plasma. In the case of phase I detoxifying enzyme activities in liver, 7-ethoxyresorufin O-deethylase (EROD), 7-benzyloxy-4-[trifluoromethyl]-coumarin-O-debenzyloxylase (BFCOD) and carboxylesterases (CE), showed a comparable decrease at T60 with a slight recovery at T120. In contrast, glutathione-S-transferase (GST) activity was significantly enhanced at T60 compared to the other sampling stages. In conclusion, MPs ingestion occurs in aquaculture reared seabream where potentially associated plasticizers accumulate in the muscle and both could be responsible for plasma and liver oxidative stress damage and alterations on detoxifying biomarkers responses.Frequently-occurred secondary organic aerosols (SOAs) under low-NOx conditions contribute to the winter haze episodes and remain unclear in the abundant presence of NH3. Here, the effects of CaCl2 seed particles on the photooxidation of low-molecular-weight C3H6 with co-existing NO2 and NH3 were highlighted and investigated through a chamber-simulation study equipped with high-resolution proton-transfer-reaction time-of-flight mass spectrometer (PTR-ToF-MS). The influences of NH3 are often overestimated to exclusively enhance SOA yields under a low-[NO2]0 condition. Instead, the seeds played a central role in the heterogeneous formation of SOAs in this reaction with two orders of magnitudes higher than that in the absence of seeds at relative humidity (RH) of 82%. Interestedly, the O3 production was unchanged whether the seeds existed or not, small changes in the production of O3 were observed whether the seeds existed or not, indicating that the gas-phase conversions of C3H6 and NOx into C1-C3 oxygenated volatile organic compounds (OVOCs) and nitrogen-containing compounds (NOCs) were not affected by seed particles. Given that the ensuing formation of these low-volatile compounds was condensed into nucleation on the seeds, the explosive growth of C3H6 SOAs was then stimulated in the addition of NH3. Besides NO2 photolysis, the producing O3 was related to the formation of secondary carbonyls such as formaldehyde and then was consumed in the ·OH generation of approximately 3.40 × 10-12 molecules cm-3. This study provides a new insight to better understand the new gas-to-particle formation mechanisms when the haze pollution outbreaks in the complex air mixture.This study assessed the appropriate sampling design required for quantifying variability in metal accumulation in the leaf tissues of A. marina, a dominant mangrove inhabiting Australian estuaries, by applying a hierarchical nested sampling design to sample mangroves at various levels of biological and spatial hierarchies (leaf, branch, tree, site). It was revealed that most variation in metal accumulation occurred among trees and branches, with insignificant variation between sites and among leaves. We also examined the accumulation of metal (loid)s in the leaf tissues collected from six locations across the Georges River estuary in southern Sydney, which differ in metal contamination history. Prospect Creek and Salt Pan Creek were the most contaminated locations, which exceeded sediment quality guideline values for Cu (66.71 ± 2.18 μg/g), Zn (317.14 ± 46.14 μg/g) and Pb (81.02 ± 2.79 μg/g). All metal(loid) concentrations in leaf tissues were much lower than their concentrations in sediment, but essential metals exhibited greater mobility.
