Cardiac hypertrophy (CH) forms the main pathological basis of chronic heart failure (CHF). Mitigating and preventing CH is the key strategy for the treatment of ventricular remodeling in CHF. Yi-Xin-Shu capsule (YXS) has been commonly applied in the clinical treatment of CHF in Asian countries for several decades. However, the underlying mechanism of YXS has not been revealed yet.

To assess the efficiency of YXS in CH and identify its potential therapeutic targets for the managing of CH.

Ultrasonic cardiogram was used to evaluate the cardiac function of CH rats. Hematein Eosin (HE)-staining, Masson-staining and transmission electron microscope were used to measure the morphological changes, cardiac fibrosis degree and ultrastructure characteristics of cardiomyocytes, respectively. ELISA was used to detect the myocardial injury biomarkers. Then, the potential targets regulated by YXS were screened out via proteomic analysis and mass spectrometry image analysis. Finally, the targets were validated by realtion of the efficacy of YXS against CH. These data demonstrate that YXS may protect against CH by regulating the RB/HDAC1/GATA4 signaling pathway.
Liver fibrosis can be easily developed into irreversible liver cirrhosis or even liver cancer. Lysosomal acid lipase (LAL), encoded by the lipase A (Lipa) gene, is a critical enzyme involved in liver fibrosis development. Morroniside, aniridoidglycoside isolated from Cornus officinalis Sieb. et Zucc., exerts hepatic protective effects. However, the mechanism of action underling the anti-liver fibrosis effects of morroniside have not been fully elucidated.

To explore whether Lipa served as a biomarker for liver fibrosis and investigate the anti-liver fibrosis effects of morroniside and the underlying action mechanism in liver fibrosis cell models.

Besides, morroniside exerts its anti-liver fibrosis effects by targeting GATA3 and LAL and hence inhibits HSC activation. These findings provide strong scientific evidence to support the development of morroniside as novel alternative or complementary therapeutics for liver injury prevention and treatment.
Whether a brachymorphic conformation may lead to electrocardiographic peculiarities diverging from generic reference intervals (RIs) cited in textbooks remains to be established. Therefore, this study aimed at evaluating electrocardiographic variables in one of the most common brachymorphic breed, namely the French Bulldog (FB).

Medical records were retrospectively reviewed to identify healthy FBs ≥1-year-old that underwent an electrocardiogram. The Mann-Whitney U test or the Student's t-test was used to compare continuous variables among male and female dogs, while the χ
test was used for categorical variables. Spearman's correlation coefficients between electrocardiographic measurements and age and body weight were also calculated. Moreover, electrocardiographic RIs were determined.

Eighty healthy FBs were enrolled; their electrocardiographic variables fitted with generic RIs, with the exception of the QRS-complex mean electrical axis (QRS-MEA). The median QRS-MEA value was 43.5° (from -18° to +90°). A QRS-MEA shift was found in 24/80 (30%) dogs; in all cases, it was a left shift (median 26°, from -18° to +39°). Specifically, 20 dogs showed a slight left shift (from +39° to +14°), while four dogs showed a marked one (from +9° to -18°). No meaningful differences were found when comparing electrocardiographic variables between males and females. No significant correlations between electrocardiographic variables and age or body weight were found, with the exceptions of the P-wave amplitude, PQ-interval duration and QT-interval duration.

The burden of gout is substantial in the United States (US). Most gout cases can be attributed to modifiable risk factors, while systematic evidence-based assessment of gout cases and incidence attributable to the risk factors in the US is limited.

The estimated cases and incidence of gout in the US was obtained from global burden of disease (GBD) study 2019. We calculated the exposure rate of risk factors from National Health and Nutrition Examination Survey (NHANES) 2009-2010, since we estimated an average induction time of 10 years for risk factors and gout. We also conducted a meta-analysis to evaluate the associations of modifiable risk factors and gout in the US population. Furthermore, the population attributable fraction (PAF) was calculated using based on the prevalence of risk factors and relative risk (RR) from the meta-analysis.

The weighted prevalence of hypertension was 14.37%, and the average body mass index (BMI) and alcohol consumption was 28.50kg/m
and 7.14g/d, respectively. Meta-analysis showed that individuals with hypertension had a higher risk of gout (2.12, 95% CI 1.88 to 2.40). For every five units of increase in BMI, the risk of gout increased by 1.48-fold (95% CI 1.26 to 1.75). The pooled RR was 1.21 (95% CI 1.13 to 1.29) for every 10g/day increment of alcohol consumption. Mechanisms by which microbes drives outcomes are not fully elucidated. We hypothesize that Gardnerella vaginalis induces matrix metalloproteinases through TLR-2, leading to epithelial barrier dysfunction and premature cervical remodeling. Cervicovaginal cells were treated with live Gardnerella vaginalis or Lactobacillus crispatus or their bacteria-free supernatants for 24 h. For TLR-2 experiments, cells were pretreated with TLR-2 blocking antibody. A Luminex panel was run on cell media. For human data, we conducted a case-control study from a prospective pregnancy cohort of Black individuals with spontaneous preterm (sPTB) (n = 40) or term (n = 40) births whose vaginal microbiota had already been characterized. Standard analytical approaches were used to determine differences across in vitro conditions, as well as MMP-9 and associations with clinical outcomes. Gardnerella vaginalis induced MMP-1 in cervical cells (p = 0.01) and MMP-9 in cervical and vaginal (VK2) cells (p ≤ 0.001 for all). TLR-2 blockade mitigated MMP-9 induction by Gardnerella vaginalis. This is achieved by adding a compliance term for each vascular segment based on 1D NS equations. The results of the proposed method are compared against 1D Navier-Stokes and 3D fluid-structure interaction (FSI) modeling in idealized and patient-specific models. We show that 1D Navier-Stokes blood flow modeling can be highly inaccurate in predicting flow and pressure dynamics in diseased cases, while in comparison the DLP approach produces consistently accurate flow and pressure waveforms as compared to 3D FSI modeling. The relative accuracy and computational efficiency of the proposed DLP approach offer the possibility to replace or augment 1D or 3D modeling to study hemodynamics in a variety of applications.To effectively control the ongoing outbreaks of fast-spreading SARS-CoV-2 variants, there is an urgent need to add rapid variant detection and discrimination methods to the existing sewage surveillance systems established worldwide. We designed eight assays based on allele-specific RT-qPCR for real-time allelic discrimination of eight SARS-CoV-2 variants (Alpha, Beta, Gamma, Delta, Omicron, Lambda, Mu, and Kappa) in sewage. In silico analysis of the designed assays for identifying SARS-CoV-2 variants using more than four million SARS-CoV-2 variant sequences yielded ∼100% specificity and >90% sensitivity. All assays could sensitively discriminate and quantify target variants at levels as low as 10 viral RNA copy/µL with minimal cross-reactivity to the corresponding nontarget genotypes, even for sewage samples containing mixtures of SARS-CoV-2 variants with differential abundances. Integration of this method into the routine sewage surveillance in Hong Kong successfully identified the Beta variant in a community sewage. Complete concordance was observed between the results of viral whole-genome sequencing and those of our novel assays in sewage samples that contained exclusively the Delta variant discharged by a clinically diagnosed COVID-19 patient living in a quarantine hotel. Our assays in this method also provided real-time discrimination of the newly emerging Omicron variant in sewage two days prior to clinical test results in another quarantine hotel in Hong Kong. These novel allelic discrimination assays offer a rapid, sensitive, and specific way for detecting multiple SARS-CoV-2 variants in sewage and can be directly integrated into the existing sewage surveillance systems.Why would concepts seem to grow when their instances become rare? Human observers can respond to decreases in stimulus prevalence by expanding their conceptual boundaries of those stimuli. This prevalence-induced concept change may have serious social consequences, since many real-world detection tasks demand consistent judgments over time. The current work aims to identify the computational process that produces prevalence-induced concept change. I review some plausible models from the cognitive and social sciences that could account for this phenomenon, and then use trial-level computational modeling to see how well each model predicts actual human data, finding that they are best explained as a range-frequency compromise in judgment. Finally, I test an intervention that successfully eliminates prevalence-induced concept change by making stimuli more intense as they become rare.
