In the group overexpressing all cytoplasmic tail exons (ICD-FL), 1401 differentially expressed genes (DEGs) were identified; overexpressed of ICD constructs lacking exon-35 (ICD-△35) identified 413 DEGs; and overexpression of ICD constructs lacking exon-35 and exon-36 (ICD-△35 + 36) identified 22 DEGs. The DEGs were enriched in immunity and metabolism-related pathways. The expression of selected genes was confirmed using quantitative real-time reverse transcription PCR. The transcriptomes of Drosophila S2 cells overexpressing different ICDs were then determined. We identified key immune, metabolic, and cell proliferation-regulated genes and gene networks, providing insights into the membrane-to-nuclear signaling pathway of Dscam.N-myc-interactor (Nmi) belongs to interferon (IFN) stimulated genes (ISGs) and is involved in the regulation of physiological processes including viral infection, inflammatory response, apoptosis and tumorigenesis in mammals. However, the function of Nmi in teleost fish remains to be explored. In this study, an Nmi homologue was characterized from mandarin fish Siniperca chuatsi. selleck The mandarin fish Nmi shares two conserved functional Nmi/IFP35 homology domains (NIDs) with mammalian Nmi protein in its C-terminal domain and a coiled coil region (CC) in its N-terminal domain, with its genomic DNA sequence consisting of nine exons and eight introns. Subcellular localization analysis shows that mandarin fish Nmi is a cytoplasmic protein and that its localization is dependent on the CC and NID1 regions. High and constitutive mRNA level of Nmi was observed in all examined tissues, with the highest level being observed in blood. In addition, the Nmi gene was significantly induced in various organs/tissues following the infection of infectious spleen and kidney necrosis virus (ISKNV), and its mRNA and protein level was also significantly induced in vitro after the treatment of IFNh, IFNc, as well as IFN-γ. The dual luciferase activity analysis indicated that the Nmi promoter was activated by the three type I IFNs through interferon-stimulated response element (ISRE) sites, and it can be also transcriptionally activated by IFN-γ via IRF1 which can activate the expression of Nmi through ISRE. Taken together, it is demonstrated in this study that the transcription of Nmi in mandarin fish can be regulated by type I and type II IFNs, thus confirming that Nmi in fish is also an ISG, and is involved in antiviral and IFN-induced innate immunity.Tinnitus, the auditory phantom percept, is a well-known heterogenous disorder with multiple subtypes. Researchers and clinicians have tried to classify these subtypes according to clinical profiles, aetiologies, and response to treatment with little success. The occurrence of overlapping tinnitus subtypes suggests that the disorder exists along a continuum of severity, with no clear distinct boundaries. In this perspective, we propose a neuro-mechanical framework, viewing tinnitus as a dimensional disorder which is a complex interplay of its behavioural, biological and neurophysiological phenotypes. Moreover, we explore the potential of these dimensions as interacting networks without a common existing cause, giving rise to tinnitus. Considering tinnitus as partially overlapping, dynamically changing, interacting networks, each representing a different aspect of the unified tinnitus percept, suggests that the interaction of these networks determines the phenomenology of the tinnitus, ultimately leading to a dimensional spectrum, rather than a categorical subtyping. We verify that the formulas for the basic reproduction numbers correctly specify the invasibility conditions.Tire wear (TW) constitutes a significant source of microplastic in terrestrial ecosystems. It is known that particles emitted by roads can have an effect up to 100 m into adjacent areas. Here, we apply for the first-time thermal extraction desorption gas chromatography-mass spectrometry (TED-GC/MS) to determine TW in soil samples by detection of thermal decomposition products of styrene-butadiene rubber (SBR), without additional enrichment. Additionally, zinc contents were determined as an elemental marker for TW. Mixed soil samples were taken along three transects along a German motorway in 0.3, 2.0, and 5.0 m distance from the road. Sampling depths were 0-2, 2-5, 5-10, and 10-20 cm. Four fine fractions, 1 000-500, 500-100, 100-50, and less then 50 μm, were analyzed. TW contents based on SBR ranged from 155 to 15 898 mg kg-1. TW contents based on zinc were between 413 and 44 812 mg kg-1. Comparison of individual values of SBR and zinc reveals SBR as a more specific marker. Results confirm that most TW ends up in the topsoil within a 2 m distance. The sampling strategy resulted in representative data for a larger area. Standard deviations of quadruple TED-GC/MS determination of SBR were less then 10% for all grain size fractions. TED-GC/MS is a suitable analytical tool for determining TW in soil samples without the use of toxic chemicals, enrichment, or special sample preparation.We measured concentrations of 19 trace elements and mercury speciation in grey seals (Halichoerus grypus) from the Gulf of St. Lawrence (GSL), Canada. With interest growing in commercializing grey seal products for human consumption in this region, our goal was to measure essential and non-essential trace elements in grey seals to evaluate health concerns and nutritional benefits. From 2015 to 2019, 120 grey seals were sampled by hunters and researchers at 4 sites in the GSL. Muscle, liver, heart and kidney samples were analyzed for 10 non-essential elements (Sb, As, Be, B, Cd, Pb, Hg, Ni, Tl, Sn) and 9 essential elements (Co, Cr, Cu, Fe, Mg, Mn, Mo, Se, Zn). Both total mercury (THg) and methylmercury (MeHg) were analyzed for a subset of samples. Results showed a two-step bioaccumulation pattern with lower element concentrations in the muscle (Fe, Mg, Se) and livers (Cd, Cr, Hg, Mn, Mo, Se) of young-of-the-year harvested in the winter ( less then 6 wks old) compared to older animals feeding at sea. We did not observe progressive age-dependent bioaccumulation for older seals (∼5 mos-29 yrs). Sex-specific differences were not very pronounced, but a few elements were 30-70% higher in the muscle (THg, MeHg) and liver (Mn, Zn) of male seals. Comparison to Canadian dietary reference intakes shows that a weekly portion of liver from young-of-the-year ( less then 6 wks old) is a good source of essential elements (Cu, Fe) and that muscle and liver from this age category do not exceed reference values for toxic elements (As, Cd, Pb, MeHg). Discussions with regional public health professionals are on-going to develop dietary recommendations for the consumption of older grey seals.Ionic liquids (ILs) are composed of only anions and cations and are liquid solvents at room temperature. Different functional groups were introduced into the ILs, conferring them with specific functions or purposes and thus forming special ILs, namely task-specific ILs (TSILs). Imidazolium-based ILs are the most widely used ILs in industrial production. To date, there have been some studies on the toxic effects of ILs on different organisms. However, the effect of functionalized groups on the toxicity of ILs is still unclear. In the present study, zebrafish were used as model organisms to study the toxic effects of 1-ethyl-3-methylimidazolium nitrate ([C2mim]NO3) and 1-hydroxyethyl-3-methylimidazolium nitrate ([HOC2mim]NO3). The results showed that both promoted an increase in reactive oxygen species (ROS) contents, leading to lipid peroxidation and DNA damage. Furthermore, integrated biological response analysis showed that [HOC2mim]NO3 was less toxic to zebrafish than [C2mim]NO3, which indicated that adding functional groups decreased the toxicity of ILs to organisms. The influence of chemical structure on IL toxicity was also reported. These results could provide a scientific basis for better synthesis and utilization of ILs in the future.Aquatic insects within glacial-melt streams are adapted to low dissolved inorganic ion concentrations. Increases in ion concentrations in glacial-melt streams are predicted with increasing air temperatures, which may impact future aquatic insect survival in these streams. We hypothesized that stonefly (Plecoptera) naiads from glacial-melt streams acclimated to different conductivity would differ in survival, median lethal concentrations, and chloride cell responses to elevated conductivity above that expected in our study streams. We conducted field bioassays in remote glacial-melt streams in southwestern China in 2015 and exposed representative stonefly naiads (Chloroperlidae, Nemouridae, Taeniopterygidae) from stream sites differing in conductivity to experimental conductivity ranging from 11 to 20,486 μS/cm for up to 216 h. We examined survivorship, calculated 96-h median lethal concentrations, and measured chloride cell responses with scanning electron microscopy.
