DEGs specifically upregulated in basal-like TN tumours were involved in antigen processing and presentation, in normal-like TN tumours encoded G protein-coupled receptors (GPCRs), and in LB fHER2- tumours were associated with lysosomes, phagosomes, and endosomes formation. Downregulated DEGs in CLs were associated with structural and signalling cell surface components. Hence, our results suggest that upregulation of genes enhancing proliferation and metabolism is a common feature among FMCs and derived CLs. In contrast, the dissimilarities observed in dysregulation of membrane components highlight CLs' disconnection with the tumour microenvironment. Furthermore, recurrent and exclusive DEGs associated with dysregulated pathways might be useful for the development of prognostically and therapeutically-relevant targeted panels.In highly urbanized and industrialized areas, the demand for construction land is expanding, which should have an impact on the water environment. Taking the Yangtze River Delta (YRD) and considering chemical oxygen demand (COD) and ammonia nitrogen (NH3-N) as characteristic pollutants, this study investigated the spatial-temporal characteristics of water pollutant emissions at the county level, optimized the spatial lag model (SLM) to estimate the spatial interaction of urban expansion and water pollutant emissions through direct and indirect effects. The results show that from 2011 to 2015, water pollutant emissions in the YRD decreased significantly and that the high-emissions pattern changed from a contiguous to a scattered distribution. The emissions of COD and NH3-N in counties at various distances from the Yangtze River and coastline show a logarithmic curve relationship. The association between urban expansion and water pollutant emissions was significant and stable. In 2015, every 1% increase in the scale of urban expansion resulted in 0.299% and 0.340% increases in local COD and NH3-N emissions, respectively, and emissions in the adjacent counties synchronously increased by 0.068% and 0.084%, respectively. The results show that to break the association and spatial interaction between urban expansion and water pollutant emissions and alleviate the environmental stress on the YRD, in addition to delimiting an urban expansion boundary and strictly restraining the scale of expansion, improvement in the regional environmental carrying capacity through urban water pollutant treatment facilities and pipe network construction is urgently needed.Schaaf-Yang syndrome (SYS) is a congenital disorder characterized by developmental delay, autism spectrum disorder and congenital joint contractures. In this study, a nationwide epidemiological questionnaire-based survey of SYS in the Japanese population was conducted to establish patient numbers, clinical features and genetic information. In the primary survey, we investigated the number of SYS patients. In the secondary survey, we obtained and analyzed detailed clinical and genetic information of SYS patients. This survey collected information on 25 genetically-confirmed patients. The major clinical symptoms included neonatal hypotonia (96% of the patients), poor suck in infancy (82%), developmental delay (100%) and joint contractures (83%). Other main symptoms and findings included characteristic facial features (100%), small hands (92%), eye abnormalities (92%) and short stature (79%). Based on the information collected on activities of daily living, 71% of patients were unable to walk, while 67%, 71%, and 81% of patients required full assistance with eating, toileting and bathing, respectively. Regarding inheritability, the genetic analysis of 21 patients revealed that 14 (67%) carried de novo truncating variants in the melanoma antigen L2 (MAGEL2) gene and seven (33%) had inherited truncating variants from their fathers who were carriers. This survey revealed the clinical and genetic features in Japanese SYS patients. The majority of SYS patients required assistance in many aspects of daily living, and there were a certain number of carriers of the imprinting disorder.Nitrate pollution in groundwater, which is an international problem, threatens human health and the environment. It could take decades for nitrate to transport in the groundwater system. When understanding the impacts of this nitrate legacy on water quality, the nitrate transport velocity (vN) in the unsaturated zone (USZ) is of great significance. Although some local USZ vN data measured or simulated are available, there has been no such a dataset at the global scale. Here, we present a Global-scale unsaturated zone Nitrate transport Velocity dataset (GNV) generated from a Nitrate Time Bomb (NTB) model using global permeability and porosity and global average annual groundwater recharge data. To evaluate GNV, a baseline dataset of USZ vN was created using locally measured data and global lithological data. The results show that 94.50% of GNV match the baseline USZ vN dataset. This dataset will largely contribute to research advancement in the nitrate legacy in the groundwater system, provide evidence for managing nitrate water pollution, and promote international and interdisciplinary collaborations.The growing concern about the negative impact of artificial light at night on biodiversity and human health increases the need of defining a general indicator that could be used for characterizing light pollution as well as performing both spatial and temporal comparisons. In this paper, we show that the traditional indicators based on direct numerical measurements of sky brightness suffer from significant limitation due to calibration bias and lack of reproducibility. Furthermore, these measures are most often performed in periods of clear sky. They do not reflect the wide variety of meteorological conditions that can produce highly inhomogeneous levels of light pollution on a given site. To overcome these issues, we propose a statistical indicator called NSB Dispersion Ratio. This indicator is derived from a statistically significant number of individual night sky brightness measurements, under various meteorological conditions. It is independent of any absolute photometer calibration. It only requires on-time precise corrections of the contribution of natural light sources such as the Galactic plane.Discrimination of ovarian tumors is necessary for proper treatment. In this study, we developed a convolutional neural network model with a convolutional autoencoder (CNN-CAE) to classify ovarian tumors. A total of 1613 ultrasound images of ovaries with known pathological diagnoses were pre-processed and augmented for deep learning analysis. We designed a CNN-CAE model that removes the unnecessary information (e.g., calipers and annotations) from ultrasound images and classifies ovaries into five classes. We used fivefold cross-validation to evaluate the performance of the CNN-CAE model in terms of accuracy, sensitivity, specificity, and the area under the receiver operating characteristic curve (AUC). Gradient-weighted class activation mapping (Grad-CAM) was applied to visualize and verify the CNN-CAE model results qualitatively. In classifying normal versus ovarian tumors, the CNN-CAE model showed 97.2% accuracy, 97.2% sensitivity, and 0.9936 AUC with DenseNet121 CNN architecture. In distinguishing malignant ovarian tumors, the CNN-CAE model showed 90.12% accuracy, 86.67% sensitivity, and 0.9406 AUC with DenseNet161 CNN architecture. Grad-CAM showed that the CNN-CAE model recognizes valid texture and morphology features from the ultrasound images and classifies ovarian tumors from these features. CNN-CAE is a feasible diagnostic tool that is capable of robustly classifying ovarian tumors by eliminating marks on ultrasound images. CNN-CAE demonstrates an important application value in clinical conditions.Data on real-world effectiveness of cyclin-dependent kinase 4/6 inhibitor combination therapy versus endocrine therapy alone are limited. The Flatiron Health Analytic Database was used to assess overall survival (OS) in patients with hormone receptor-positive/human epidermal growth factor receptor 2-negative (HR+/HER2-) metastatic breast cancer (MBC) treated with first-line palbociclib plus an aromatase inhibitor (AI) versus an AI alone in routine US clinical practice. In total, 2888 patients initiated treatment during February 3, 2015-March 31, 2020, with a potential ≥6-month follow-up (cutoff date, September 30, 2020). After stabilized inverse probability treatment weighting, median OS (95% CI) is significantly longer among palbociclib versus AI recipients (49.1 [45.2-57.7] versus 43.2 [37.6-48.0] months; hazard ratio, 0.76 [95% CI, 0.65-0.87]; P less then 0.0001). Progression-free survival (95% CI) is 19.3 (17.5-20.7) versus 13.9 (12.5-15.2) months, respectively (hazard ratio, 0.70 [95% CI, 0.62-0.78]; P less then 0.0001). These data support first-line palbociclib plus an AI treatment for HR+/HER2- MBC.(Trial number NCT05361655).Relativistic electron precipitation (REP) from the Earth's radiation belt plays an important role in mesospheric ozone loss as a connection between space weather and the climate system. However, the rapid (tens of minutes) destruction of mesospheric ozone directly caused by REP has remained poorly understood due to the difficulty of recognizing its location and duration. Here we show a compelling rapid correspondence between localized REP and ozone destruction during a specific auroral phenomenon, the called an isolated proton aurora (IPA). The IPA from the Earth's radiation belt becomes an important spatial and temporal proxy of REP, distinct from other auroral phenomena, and allowing visualizing micro-ozone holes. We found ozone destruction of as much as 10-60% within 1.5 h of the initiation of IPA. Electromagnetic ion cyclotron waves in the oxygen ion band observed as the driver of REP likely affect through resonance with mainly ultra-relativistic (> 2 mega-electron-volts) energy electrons. The rapid REP impact demonstrates its crucial role and direct effect on regulating the atmospheric chemical balance.After vaccination with enterovirus 71 (EV-A71), the prevalence of hand-foot-and-mouth disease (HFMD) remained high, and the spatial-temporal distribution of enteroviruses changed. Therefore, it is essential to define the temporal features, spatial distributions, and epidemiological and etiological characteristics of HFMD in Kunming. Between 2017 and 2020, a total of 36,540 children were diagnosed with HFMD in Kunming, including 32,754 children with enterovirus-positive clinical samples. Demographic, geographical, epidemiological and etiological data of the cases were acquired and analyzed. Other enteroviruses replaced EV-A71, and the incidence of EV-A71 decreased dramatically, whereas coxsackievirus A6 (CV-A6) and coxsackievirus A16 (CV-A16) had substantial outbreaks in 2018 and 2019, respectively. The major and minor peaks all extended for 2-4 months compared to before vaccination with the EV-A71 vaccine. From 2019 to 2020, CV-A6, as the predominant serotype, showed only a single peak.
