The Chatham Islands has some of the most prized black-footed abalone (Haliotis iris) beds in New Zealand. Metabolic disorders are significant in the occurrence and development of malignant tumors. Changes of specific metabolites and metabolic pathways are molecular therapeutic targets. This study aims to determine the metabolic differences between oral squamous cell carcinoma (OSCC) tissues and paired adjacent noncancerous tissues (ANT) through liquid chromatography-mass spectrometry (LC-MS). SPHK1 is a key enzyme in sphingolipid metabolism. This study also investigates the potential role of SPHK1 in OSCC.

This study used LC-MS to analyze metabolic differences between OSCC tissues and paired ANT. Principal component analysis (PCA) and partial least-squares discriminant analysis (PLS-DA) were applied to explain the significance of phospholipid metabolism pathways in the occurrence and development of OSCC. Patients developing metastatic gastrointestinal stromal tumors (mGIST) have heterogenous disease biology and oncologic outcomes; prognostic factors are incompletely characterized. We sought to evaluate predictors of 10-year metastatic survivorship in the era of tyrosine kinase inhibitor (TKI) therapy.

We reviewed patients with mGIST treated at our Comprehensive Cancer Center from 2003 to 2019, including only patients with either mortality or 10 years of follow-up. Ten-year survivorship was evaluated with logistic regression.

We identified 109 patients with a median age of 57 years at mGIST diagnosis. Synchronous disease was present in 57% (n = 62) of patients; liver (n = 48, 44%), peritoneum (n = 40, 37%), and liver + peritoneum (n = 18, 17%) were the most common sites. Forty-six (42%) patients were 10-year mGIST survivors. Following mGIST diagnosis, radiographic progression occurred within 2 years in 53% (n = 58) of patients, 2-5 years in 16% (n = 17), and 5-10 years in 16% (n = 17), with median survivagement of patients with disease progression receiving TKI therapy merits further study.

It is known that weather conditions prior to and after the onset of reproduction can affect the breeding success of birds. However, little is known about how seasonal weather variability can affect birds' breeding performance, particularly for species with a slow pace of life. Long-term studies are key to understanding how weather variability can affect a population's dynamics, especially when extreme weather events are expected to increase with climate change. Using a 32-year population study of the Blue-footed booby (Sula nebouxii) in Mexico, we show that seasonal variation in weather conditions, predominantly during the incubation stage, affects offspring survival and body condition at independence. During most of the incubation period, warm sea surface temperatures were correlated with low hatching success, while rainfall in the middle of the incubation stage was correlated with high fledging success. In addition, chicks from nests that experienced warm sea surface temperatures from the pre-laying stage to near-fledging had lower body condition at 70 days of age. Finally, we show that variable annual SST conditions before and during the incubation stage can impair breeding performance. Assessment of various technologies led to selection of a heterologous vaccine using chimpanzee adenovirus (AdC68) for priming followed by boosts with electroporation of DNA plasmid to deliver T cell antigens to the immune system. We found that priming with AdC68 rapidly activates and expands antigen-specific T cells and does not encounter pre-existing immunity as occurs with the use of a human adenovirus vaccine. The AdC68 vector does, however, induce new anti-virus immune responses, limiting its use for boosting. To circumvent this, boosting with DNA encoding the same antigens can be done repetitively to augment and maintain vaccine responses. Using mouse and monkey models, we found that the activation of both CD4 and CD8 T cells was amplified by combination with anti-CTLA-4 and anti-PD-1 antibodies. These antibodies were administered subcutaneously to target their distribution to vaccination sites and to reduce systemic exposure which may improve their safety. VBIR can break tolerance and activate T cells recognizing tumor-associated self-antigens. This activation lasts more than a year after completing treatment in monkeys, and inhibits tumor growth to a greater degree than is observed using the individual components in mouse cancer models. Even though the population decreased by 11%, noticeable agricultural and forest land areas were turned into urban land. In this work, we analyzed those LULC changes by mapping the spatial characteristics of LULC and urban expansion in the years 2000-2019 in Estonia. Moreover, using the revealed spatiotemporal transitions of LULC, we simulated LULC and urban expansion for 2030. Landsat 5 and 8 data were used to estimate 147 spectral-textural indices in the Google Earth Engine cloud computing platform. Between 2019 and 2030, the other significant changes were the decrease of 34.57 km2 of forest lands and the increase of agricultural lands by 14.90 km2 and wetlands by 9.31 km2. These findings can develop a proper course of action for long-term spatial planning in Estonia. Therefore, a key policy priority should be to plan for the stable care of forest lands to maintain biodiversity.
