Basal-like breast cancers (BBC) exhibit subtype-specific phenotypic and transcriptional responses to stroma, but little research has addressed how stromal-epithelial interactions evolve during early BBC carcinogenesis. It is also unclear how common genetic defects, such as p53 mutations, modify these stromal-epithelial interactions. To address these knowledge gaps, we leveraged the MCF10 progression series of breast cell lines (MCF10A, MCF10AT1, and MCF10DCIS) to develop a longitudinal, tissue-contextualized model of p53-deficient, pre-malignant breast. Acinus asphericity, a morphogenetic correlate of cell invasive potential, was quantified with optical coherence tomography imaging, and gene expression microarrays were performed to identify transcriptional changes associated with p53 depletion and stromal context. Co-culture with stromal fibroblasts significantly increased the asphericity of acini derived from all three p53-deficient, but not p53-sufficient, cell lines, and was associated with the upregulation of 38 genes. Necrosulfonamide in vivo When considered as a multigene score, these genes were upregulated in co-culture models of invasive BBC with increasing stromal content, as well as in basal-like relative to luminal breast cancers in two large human datasets. Taken together, stromal-epithelial interactions during early BBC carcinogenesis are dependent upon epithelial p53 status, and may play important roles in the acquisition of an invasive morphologic phenotype.This paper investigates the associations between stress and fatigue and sexual spontaneity among heterosexual married couples in Singapore, a high-income society with high work performance expectations and long working hours. Data on self-reported menstrual dates, sexual activity within the marriage, and stress and fatigue levels were collected from 657 married women aged 25-34 years through biweekly online diaries over 14 weeks. Using a two-way fixed effects logistic model which controlled for time-invariant individual fixed effects and menstrual cycle fixed effects, the paper tests three hypotheses (1) sexual activity is significantly lower on weekdays compared to weekends and public holidays, (2) sexual activity is significantly lower during periods when women reported higher stress and fatigue, and (3) stress and fatigue are more predictive of sexual activity during weekdays than during weekends. Frequencies of marital sexual intercourse during the diary keeping period were 3.69 and 2.55 per 30 days among women aged 25-29 and 30-34 years, respectively, much lower than ideal frequencies of 6.40 and 5.23. Consistent with all three hypotheses, there was a strong positive weekend effect and a weak but positive public holiday effect, and strong negative effects of both stress and fatigue, especially during weekdays. There was no evidence of compensatory sex on weekends to make up for hectic workweeks. Increased support for work-life balance can contribute to more spontaneous marital sex lives and may reduce underachievement of ideals for sexual intimacy and childbearing in Singapore.
Repeat pulmonary metastasectomy (PM) considerably improves the prognosis of patients with pulmonary metastases of osteosarcoma. Reports have demonstrated a significantly improved prognosis in patients who have undergone repeat metastasectomy for osteosarcoma; however, there have been no reports with more than six metastasectomies. Herein, we describe the long-term survival of a patient following resection of multiple tumors and other treatments for metastatic osteosarcoma.

A 28-year-old woman underwent extensive resection and postoperative adjuvant chemotherapy for right tibial sarcoma. Over the years, she developed repeated pulmonary metastases. First, 116 metastases were removed from the bilateral lungs. After that, multiple PMs of approximately 250 tumors and other treatments for deep metastatic lesions were performed. The patient died of the underlying disease 24years after the primary surgery.

However, mercury concentrations vary among waterbodies and fishes due to a suite of environmental conditions and detection of elevated mercury concentrations is imperfect, resulting in inaccurate consumption guidelines. Occupancy models may be a useful approach for addressing these issues but have not been used for these purposes. Our objectives were to use occupancy modeling to (1) estimate number of samples needed to detect mercury levels surpassing >0.30 mg/kg wet weight in fish at a waterbody (2) identify individual fish-level factors associated with detection probability, and (3) identify environmental-level factors linked to elevated mercury levels in fish at a waterbody. Mercury concentrations were estimated from >500 largemouth bass Micropterus salmoides and walleye Sander vitreus from 30 waterbodies throughout Iowa, USA to identify individuals with concentrations > or 0.30 mg/kg varied between species and increased with fish length; consequently, more samples were needed to detect elevated mercury concentrations in small versus large fish. The probability of a waterbody having fish with elevated mercury levels increased with the percent grassland and declined with percent agriculture in the watershed, providing prioritization metrics for mercury surveillance programs. Our results demonstrate that occupancy models can be a valuable tool for mercury surveillance due to their ability to estimate necessary sample sizes and identify fish sizes and waterbodies with elevated mercury concentrations while accounting for imperfect detection probabilities.Multiple rapid swallow (MRS) is a complementary test performed during high-resolution esophageal manometry (HREM). The present study was done to assess the MRS findings in patients with gastroesophageal reflux disease (GERD) who have normal peristalsis and minor motility disorders. This is a retrospective analysis from a prospectively maintained database of patients referred for HREM study to our center. The recorded details included age, sex, symptoms with duration, and upper gastrointestinal endoscopy reports. HREM data included basal inspiratory and expiratory pressures, median integrated relaxation pressure, and peristaltic pattern. Three MRS sequences were recorded per patient. The inhibition and contraction phases were interpreted for each MRS sequence. A total of 68 patients (38 normal motility, 30 minor motility disorders) formed the study cohort. Hiatus hernia was common in GERD patients with minor motility disorders (p 0.02).
